BRCA1 Promotes Repair of DNA Damage in Cochlear Hair Cells and Prevents Hearing Loss

Cochlear hair cells (HCs) sense sound waves and allow us to hear. Loss of HCs will cause irreversible sensorineural hearing loss. It is well known that DNA damage repair plays a critical role in protecting cells in many organs. However, how HCs respond to DNA damage and how defective DNA damage repair contributes to hearing loss remain elusive. In this study, we showed that cisplatin induced DNA damage in outer hair cells (OHCs) and promoted OHC loss, leading to hearing loss in mice of either sex. Cisplatin induced the expression of Brca1, a DNA damage repair factor, in OHCs. Deficiency of Brca1 induced OHC and hearing loss, and further promoted cisplatin-induced DNA damage in OHCs, accelerating OHC loss. This study provides the first in vivo evidence demonstrating that cisplatin mainly induces DNA damage in OHCs and that BRCA1 promotes repair of DNA damage in OHCs and prevents hearing loss. Our findings not only demonstrate that DNA damage–inducing agent generates DNA damage in postmitotic HCs but also suggest that DNA repair factors, like BRCA1, protect postmitotic HCs from DNA damage–induced cell death and hearing loss.

Last summer, news reports of orcas deliberately tearing the propellers off of yachts in the Strait of Gibraltar thrilled observers who were eager to cast these intelligent and social pack hunters as class warriors striking a blow for the “common mammals” against the one percent. That turned out to be wishful thinking, according to guest Lori Marino, a biopsychologist who studies whale and dolphin intelligence. She told us that these six-ton whales were just having fun—if they wanted to harm the occupants of those boats, we’d know it.  Even so, these encounters are becoming a predictable seasonal occurrence between the months of May and August: A 50-foot charter vessel sank after its hull and rudder were damaged in an orca encounter near the Strait of Gibraltar on May 12. How to identify old Orcad Schematic entry version


Good morning,
I dug up an old project from 2005 and I should open the schematic to check some things.
This is the schematic of a XILINX XC95108-pq160 CPLD which the XILINX ISE 6.1 software then translated and compiled, to generate a JEDEC file to burn CPLD.

My problem is that I can't open schematics with the versions of Orcad Schematic Entry that I have.
Can anyone help me understand which version of Orcad Schematic Entry I need to install to see these files?

How to transfer custom title block from Orcad Capture to PCB Editor

Hi,

So I was trying to update the title block of a schematic that I have. The title block that was on there was out of date . I clicked on place --> title block and was able to find the title block that I need. I also have a .OLB file that contains that title block. Then I created a Netlist with the old BRD file as the input file (To keep it as is but modify changes) but when I do that I still do not see / cannot place the title block that I need. Under Place --> format symbols in Allegro , I do see a title block that is coming from the database (But it's the old one). I don't know what to do at this point and would appreciate any tips. I did make sure that the path to where the library is , is defined in the user preferences. 
I also tried copying the title block under the library folder in capture before creating my Netlist and that did not work either.

Orcad PCB (allegro) not using GPU over USB

Hi,

I have a monitor plugged to my laptop using a HDMI to USB adapter. When using this adapter, Allegro runs very slowly. It seems that it is not using my video card.

Is this a known issue with a workaround I can try?

Migrating from files Orcad Layout 16.2

I have managed to convert our old schematic and PCD file to from Layout 16.2 to 17.4

I have exported the footprints and moved them to the correct lib directory. 

I get no DRC errors and I can build a new netlist file. The problem is I can't get the PCB editor to update using the new netlist and get the following error:

No windows cascading in OrCAD Capture 17.4

Hello All,

I'm a novice to this forum and probably this subject has been already discussed here.

My company has purchased OrCAD Capture 17.4 tools that have a new GUI if compared to my earlier used OrCAD Capture 9.2. I have been using Capture 9.2 for ~18 years and its GUI is really convenient. The GUI of 17.4 looks to be a modern one with new icons and really has improved features and new capabilities.

However, my main complain about GUI 17.4 is that the schematic windows cannot be cascaded. Although they can be set floating, this is even more annoying because all toolbars remain in the Capture window and when you select a tool, the Capture window pops over already open schematic window and you need a lot of useless extra clicks to return back to the currently edited schematic page. I always used cascading of schematic windows before because my complex designs includes many pages, not speaking about the library windows that are typically open simulatneously. My view is that the lack of CASCADING in Capture GUI 17.4 is critical and unacceptable for complex projects, and I would very highly appreciate if the Cadence guys will return back the CASCADING capability for schematic pages. In case this will be done, this will make the GUI really great and comfortable to use.

Does anybody have opinion on this issue?

If so, should the floating license server be upgraded as well?

Update Package_Height_Max from Orcad Capture

I am using OrCAD PCB Designer Standard version 17.4-2019. I want to force update the Package_Height_Max property on the place bound top shape. The footprint library that we've created has that property set in the dra file, but I'd like to override that from capture so I can be certain that the height is correct.

This is coming from a place where we have created a very large footprint library over that past ++ years. Everyone who creates a new footprint is supposed to make sure that we add Package_Height_Max to the footprint, but of course footprints get reused for various parts, not all of which will have the same package height. What I want to do is export a list of package heights from our part database and then import the package heights into Capture and override the package height in the footprint.

I have found a post here  Using Height Property from Orcad Capture which says its not possible, but it also says its from 15 years ago, so maybe things have changed?

Home Respiratory Strategies in Patients With COPD With Chronic Hypercapnic Respiratory Failure

BACKGROUND:Home noninvasive ventilation (NIV) may improve chronic hypercarbia in COPD and patient-important outcomes. The efficacy of home high-flow nasal cannula (HFNC) as an alternative is unclear.METHODS:We searched MEDLINE, Embase, Cochrane CENTRAL, Scopus, and ClinicalTrials.gov for randomized trials of subjects from inception to March 31, 2023, and updated the search on July 14, 2023. We performed a frequentist network meta-analysis and assessed the certainty of the evidence using the Grading of Recommendations Assessment, Development, and Evaluation approach. We analyzed randomized controlled trials (RCTs) comparing NIV, HFNC, or standard care in adult subjects with COPD with chronic hypercapnic respiratory failure. Outcomes included mortality, COPD exacerbations, hospitalizations, and quality of life (St George Respiratory Questionnaire [SGRQ]).RESULTS:We analyzed 24 RCTs (1,850 subjects). We found that NIV may reduce the risk of death compared to standard care (relative risk 0.82 [95% CI 0.66–1.00]) and probably reduces exacerbations (relative risk 0.71 [95% CI 0.58–0.87]). HFNC probably reduces exacerbations compared to standard care (relative risk 0.77 [0.68–0.88]), but its effect on mortality is uncertain (relative risk 1.20 [95% CI 0.63–2.28]). HFNC probably improves SGRQ scores (mean difference −7.01 [95% CI −12.27 to −1.77]) and may reduce hospitalizations (relative risk 0.87 [0.69–1.09]) compared to standard care. No significant difference was observed between HFNC and NIV in reducing exacerbations.CONCLUSIONS:Both NIV and HFNC reduce exacerbation risks in subjects with COPD compared to standard care. HFNC may offer advantages in improving quality of life.

Synchronous T-lymphoblastic lymphoma and neuroblastoma in a 3-yr-old with novel germline SMARCA4 and EZH2 variants [RAPID CANCER COMMUNICATION]

T-lymphoblastic lymphoma (T-LLy) is the most common lymphoblastic lymphoma in children and often presents with a mediastinal mass. Lymphomatous suprarenal masses are possible but rare. Here, we discuss the case of a previously healthy 3-yr-old male who presented with mediastinal T-LLy with bilateral suprarenal masses. Following initial treatment, surgical biopsy of persisting adrenal masses revealed bilateral neuroblastoma (NBL). A clinical genetics panel for germline cancer predisposition did not identify any pathogenic variants. Combination large panel (864 genes) profiling analysis in the context of a precision oncology study revealed two novel likely pathogenic heterozygous variants: SMARCA4 c.1420-1G > T p.? and EZH2 c.1943G > C p.(Ile631Phefs*44). Somatic analysis revealed potential second hits/somatic variants in EZH2 (in the T-LLy) and a segmental loss in Chromosome 19p encompassing SMARCA4 (in the NBL). Synchronous cancers, especially at a young age, warrant genetic evaluation for cancer predisposition; enrollment in a precision oncology program assessing germline and tumor DNA can fulfill that purpose, particularly when standard first-line genetic testing is negative and in the setting of tumors that are not classic for common cancer predisposition syndromes.

Analysis of Sleep and Circadian Rhythms from Drosophila Activity-Monitoring Data Using SCAMP

Sleep is a fundamental feature of life for virtually all multicellular animals, but many questions remain about how sleep is regulated and what biological functions it plays. Substantial headway has been made in the study of both circadian rhythms and sleep in the fruit fly Drosophila melanogaster, much of it through studies of individual fly activity using beam break counts from Drosophila activity monitors (DAMs). The number of laboratories worldwide studying sleep in Drosophila has grown from only a few 20 years ago to hundreds today. The utility of these studies is limited by the quality of the metrics that can be extracted from the data. Many software options exist to help analyze DAM data; however, these are often expensive or have significant limitations. Therefore, we describe here a method for analyzing DAM-based data using the sleep and circadian analysis MATLAB program (SCAMP). This user-friendly software has an advantage of combining several analyses of both sleep and circadian rhythms in one package and produces graphical outputs as well as spreadsheets of the outputs for further statistical analysis. The version of SCAMP described here is also the first published software package that can analyze data from multibeam DAM5Ms, enabling determination of positional preference over time.

Neural Stimulation during Drosophila Activity Monitor (DAM)-Based Studies of Sleep and Circadian Rhythms in Drosophila melanogaster

Sleep is a fundamental feature of life for virtually all multicellular animals, but many questions remain about how sleep is regulated by circadian rhythms, homeostatic sleep drive that builds up with wakefulness, and modifying factors such as hunger or social interactions, as well as about the biological functions of sleep. Substantial headway has been made in the study of both circadian rhythms and sleep in the fruit fly Drosophila melanogaster, much of it through studies of individual fly activity using Drosophila activity monitors (DAMs). Here, we describe approaches for the activation of specific neurons of interest using optogenetics (involving genetic modifications that allow for light-based neuronal activation) and thermogenetics (involving genetic modifications that allow for temperature-based neuronal activation) so that researchers can evaluate the roles of those neurons in controlling rest and activity behavior. In this protocol, we describe how to set up a rig for simultaneous optogenetic or thermogenetic stimulation and activity monitoring for analysis of sleep and circadian rhythms in Drosophila, how to raise appropriate flies, and how to perform the experiment. This protocol will allow researchers to assess the causative role in the regulation of sleep and activity rhythms of any genetically tractable subset of cells.

Management of central sleep apnoea: a review of non-hypercapnic causes

Central sleep apnoea (CSA) is characterised by recurrent episodes of airway cessation or reduction in the absence of respiratory effort. Although CSA is less common than obstructive sleep apnoea, it shares similar symptoms. CSA can be secondary to various medical conditions, high altitude and medication exposure. CSA can also emerge during obstructive sleep apnoea therapy. There are a range of treatment options and selecting the right therapy requires an understanding of the pathophysiology of CSA.
