The first was the abduction and murder of three Israeli boys by a Hamas-linked group. Then there was an act of reprisal—the torture, burning, and murder of a Palestinian teen-ager named Mohammed Abu Khdeir—by Israeli right-wing extremists. Even by the standards of this conflict, the killings were shocking. “Our Boys,” a co-production of HBO and the Israeli Keshet Studios, examines the forces that led to Abu Khdeir’s killing. It is not for the faint of heart, David Remnick says, but the series is as complex and deep a portrayal of the conflict as he has ever seen. Remnick spoke with two of the creators: Hagai Levi, an Israeli Jew, and Tawfiq Abu Wael, a Palestinian living in Israel. The FBI's annual tally counted 7,120 hate crimes reported last year, 55 fewer than the year before. The main concern for extremism trackers, however, is the rising level of violence — the report showed an increase in the number of "crimes against persons," such as intimidation, assault and homicide. "We're seeing a leaner and meaner type of hate crime going on," said Brian Levin, director of the Center for the Study of Hate and Extremism at California State University San Bernardino. "Homicides were up and crimes against persons were up and that's an important thing to look at." Hate crimes targeting people accounted for 61% of all hate crimes in 2018, according to Levin, who is co-author of a report released Tuesday that analyzes law enforcement data. We are living in Vancouver right now, and we love a lot about it, but we could buy a place in the suburbs right now (which might not be true six months or a year from now). We are really torn, and I want some perspective on what moving to the suburbs is really like, and if owning is that much better than renting.We've been renting a flat in East Vancouver for a year and love a lot of things about it. The proximity to work downtown, the neighbourhood feel, proximity to beaches and attractions, the kids' school (both elementary-aged), cherry blossoms, shopping, all the things people love about Vancouver. We haven't been saving any money though, because our rent is outrageously high. We can buy a 2000 sq ft condo in Surrey for less than the rent of 1000 sq ft in East Van. We have a small down-payment saved up, but we're not adding to it anymore, so if we are going to buy now is the time. There are some very motivated sellers at the moment and prices have come down, which they NEVER do in the area. But we are torn. Suburbs mean longer commute (and paying for transit instead of biking to work), longer travel time to all the fun things we love, changing the kids' school, further to the airport/ferry, the awfulness of moving, etc. We would gain some space, some privacy, some autonomy (paint walls! get a hamster!) and some equity. Have you moved to the suburbs with kids? Was it worth it? Additional details: I'm a stay-at-home-mom and my wife works right downtown in Vancouver. Both of our kids have ADHD and are ROWDY. Moving to another (cheaper) rental is out-of-the-question. Even though our current place isn't perfect, its good enough that if we continue to rent we just wanna stay here. If we bought, it would be into a strata, with all that entails. We have owned a house before but not in this province. Refrigerated storage of RBCs induces oxidative stress. We hypothesized that G6PD-deficient donor RBCs would have inferior storage quality for transfusion as compared with G6PD-normal RBCs.METHODS Male volunteers were screened for G6PD deficiency; 27 control and 10 G6PD-deficient volunteers each donated 1 RBC unit. After 42 days of refrigerated storage, autologous 51-chromium 24-hour posttransfusion RBC recovery (PTR) studies were performed. Metabolomics analyses of these RBC units were also performed.RESULTS The mean 24-hour PTR for G6PD-deficient subjects was 78.5% ± 8.4% (mean ± SD), which was significantly lower than that for G6PD-normal RBCs (85.3% ± 3.2%; P = 0.0009). None of the G6PD-normal volunteers (0/27) and 3 G6PD-deficient volunteers (3/10) had PTR results below 75%, a key FDA acceptability criterion for stored donor RBCs. As expected, fresh G6PD-deficient RBCs demonstrated defects in the oxidative phase of the pentose phosphate pathway. During refrigerated storage, G6PD-deficient RBCs demonstrated increased glycolysis, impaired glutathione homeostasis, and increased purine oxidation, as compared with G6PD-normal RBCs. In addition, there were significant correlations between PTR and specific metabolites in these pathways.CONCLUSION Based on current FDA criteria, RBCs from G6PD-deficient donors would not meet the requirements for storage quality. Importantly, we found that the eukaryote-specific host signaling molecule inositol hexakisphosphate (IP6) is required for Lpg2603 kinase activity. Crystal structures of Lpg2603 in the apo-form and when bound to IP6 revealed an active-site rearrangement that allows for ATP binding and catalysis. hate Metabolic regulation of the lysosomal cofactor bis(monoacylglycero)phosphate in mice [Research Articles] By feedproxy.google.com Published On :: 2020-04-29T07:33:43-07:00 Bis(monoacylglycero)phosphate (BMP), also known as lysobisphosphatidic acid (LBPA), is a phospholipid that promotes lipid sorting in late endosomes/lysosomes by activating lipid hydrolases and lipid transfer proteins. Changes in the cellular BMP content therefore reflect an altered metabolic activity of the endo-lysosomal system. Surprisingly, little is known about the physiological regulation of BMP. In this study, we investigated the effects of nutritional and metabolic factors on BMP profiles of whole tissues and parenchymal and non-parenchymal cells. Tissue samples were obtained from fed, fasted, two-hours refed, and insulin-treated mice, as well as from mice housed at 5°C, 22°C, or 30°C. These tissues exhibited distinct BMP profiles, which were regulated by the nutritional state in a tissue-specific manner. Insulin treatment was not sufficient to mimic refeeding-induced changes in tissue BMP levels indicating that BMP metabolism is regulated by other hormonal or nutritional factors. Tissue fractionation experiments revealed that fasting drastically elevates BMP levels in hepatocytes and pancreatic cells. Furthermore, we observed that the BMP content in brown adipose tissue strongly depends on housing temperatures. In conclusion, our observations suggest that BMP concentrations adapt to the metabolic state in a tissue-and cell type-specific manner in mice. Drastic changes observed in hepatocytes, pancreatic cells, and brown adipocytes suggest that BMP possesses a role in the functional adaption to nutrient starvation and ambient temperature.
