noc A thermal deformation optimization method for cryogenically cooled silicon crystal monochromators under high heat load By journals.iucr.org Published On :: 2024-01-22 A method to optimize the thermal deformation of an indirectly cryo-cooled silicon crystal monochromator exposed to intense X-rays at a low-emittance diffraction-limited synchrotron radiation source is presented. The thermal-induced slope error of the monochromator crystal has been studied as a function of heat transfer efficiency, crystal temperature distribution and beam footprint size. A partial cooling method is proposed, which flattens the crystal surface profile within the beam footprint by modifying the cooling contact area to optimize the crystal peak temperature. The optimal temperature varies with different photon energies, which is investigated, and a proper cooling strategy is obtained to fulfil the thermal distortion requirements over the entire photon energy range. At an absorbed power up to 300 W with a maximum power density of 44.8 W mm−2 normal incidence beam from an in-vacuum undulator, the crystal thermal distortion does not exceed 0.3 µrad at 8.33 keV. This method will provide references for the monochromator design on diffraction-limited synchrotron radiation or free-electron laser light sources. Full Article text
noc Modelling the power threshold and optimum thermal deformation of indirectly liquid-nitrogen cryo-cooled Si monochromators By journals.iucr.org Published On :: 2024-04-09 Maximizing the performance of crystal monochromators is a key aspect in the design of beamline optics for diffraction-limited synchrotron sources. Temperature and deformation of cryo-cooled crystals, illuminated by high-power beams of X-rays, can be estimated with a purely analytical model. The analysis is based on the thermal properties of cryo-cooled silicon crystals and the cooling geometry. Deformation amplitudes can be obtained, quickly and reliably. In this article the concept of threshold power conditions is introduced and defined analytically. The contribution of parameters such as liquid-nitrogen cooling efficiency, thermal contact conductance and interface contact area of the crystal with the cooling base is evaluated. The optimal crystal illumination and the base temperature are inferred, which help minimize the optics deformation. The model has been examined using finite-element analysis studies performed for several beamlines of the Diamond-II upgrade. Full Article text

noc Characterization of novel mevalonate kinases from the tardigrade Ramazzottius varieornatus and the psychrophilic archaeon Methanococcoides burtonii By journals.iucr.org Published On :: 2024-02-27 Mevalonate kinase is central to the isoprenoid biosynthesis pathway. Here, high-resolution X-ray crystal structures of two mevalonate kinases are presented: a eukaryotic protein from Ramazzottius varieornatus and an archaeal protein from Methanococcoides burtonii. Both enzymes possess the highly conserved motifs of the GHMP enzyme superfamily, with notable differences between the two enzymes in the N-terminal part of the structures. Biochemical characterization of the two enzymes revealed major differences in their sensitivity to geranyl pyrophosphate and farnesyl pyrophosphate, and in their thermal stabilities. This work adds to the understanding of the structural basis of enzyme inhibition and thermostability in mevalonate kinases. Full Article text
noc STEM SerialED: achieving high-resolution data for ab initio structure determination of beam-sensitive nanocrystalline materials By journals.iucr.org Published On :: 2024-01-01 Serial electron diffraction (SerialED), which applies a snapshot data acquisition strategy for each crystal, was introduced to tackle the problem of radiation damage in the structure determination of beam-sensitive materials by three-dimensional electron diffraction (3DED). The snapshot data acquisition in SerialED can be realized using both transmission and scanning transmission electron microscopes (TEM/STEM). However, the current SerialED workflow based on STEM setups requires special external devices and software, which limits broader adoption. Here, we present a simplified experimental implementation of STEM-based SerialED on Thermo Fisher Scientific STEMs using common proprietary software interfaced through Python scripts to automate data collection. Specifically, we utilize TEM Imaging and Analysis (TIA) scripting and TEM scripting to access the STEM functionalities of the microscope, and DigitalMicrograph scripting to control the camera for snapshot data acquisition. Data analysis adapts the existing workflow using the software CrystFEL, which was developed for serial X-ray crystallography. Our workflow for STEM SerialED can be used on any Gatan or Thermo Fisher Scientific camera. We apply this workflow to collect high-resolution STEM SerialED data from two aluminosilicate zeolites, zeolite Y and ZSM-25. We demonstrate, for the first time, ab initio structure determination through direct methods using STEM SerialED data. Zeolite Y is relatively stable under the electron beam, and STEM SerialED data extend to 0.60 Å. We show that the structural model obtained using STEM SerialED data merged from 358 crystals is nearly identical to that using continuous rotation electron diffraction data from one crystal. This demonstrates that accurate structures can be obtained from STEM SerialED. Zeolite ZSM-25 is very beam-sensitive and has a complex structure. We show that STEM SerialED greatly improves the data resolution of ZSM-25, compared with serial rotation electron diffraction (SerialRED), from 1.50 to 0.90 Å. This allows, for the first time, the use of standard phasing methods, such as direct methods, for the ab initio structure determination of ZSM-25. Full Article text
noc Orientational analysis of atomic pair correlations in nanocrystalline indium oxide thin films By journals.iucr.org Published On :: 2024-01-01 The application of grazing-incidence total X-ray scattering (GITXS) for pair distribution function (PDF) analysis using >50 keV X-rays from synchrotron light sources has created new opportunities for structural characterization of supported thin films with high resolution. Compared with grazing-incidence wide-angle X-ray scattering, which is only useful for highly ordered materials, GITXS/PDFs expand such analysis to largely disordered or nanostructured materials by examining the atomic pair correlations dependent on the direction relative to the surface of the supporting substrate. A characterization of nanocrystalline In2O3-derived thin films is presented here with in-plane-isotropic and out-of-plane-anisotropic orientational ordering of the atomic structure, each synthesized using different techniques. The atomic orientations of such films are known to vary based on the synthetic conditions. Here, an azimuthal orientational analysis of these films using GITXS with a single incident angle is shown to resolve the markedly different orientations of the atomic structures with respect to the planar support and the different degrees of long-range order, and hence, the terminal surface chemistries. It is anticipated that orientational analysis of GITXS/PDF data will offer opportunities to extend structural analyses of thin films by providing a means to qualitatively determine the major atomic orientation within nanocrystalline and, eventually, non-crystalline films. noc Structural analysis of nanocrystals by pair distribution function combining electron diffraction with crystal tilting By journals.iucr.org Published On :: 2024-02-16 As an important characterization method, pair distribution function (PDF) has been extensively used in structural analysis of nanomaterials, providing key insights into the degree of crystallinity, atomic structure, local disorder etc. The collection of scattering signals with good statistics is necessary for a reliable structural analysis. However, current conventional electron diffraction experiments using PDF (ePDF) are limited in their ability to acquire continuous diffraction rings for large nanoparticles. Herein, a new method – tilt-ePDF – is proposed to improve the data quality and compatibility of ePDF by a combination of electron diffraction and specimen tilting. In the present work, a tilt-series of electron diffraction patterns was collected from gold nanoparticles with three different sizes and a standard sample polycrystalline aluminium film for ePDF analysis. The results show that tilt-ePDF can not only enhance the continuity of diffraction rings, but can also improve the signal-to-noise ratio in the high scattering angle range. As a result, compared with conventional ePDF data, tilt-ePDF data provide structure parameters with a better accuracy and lower residual factors in the refinement against the crystal structure. This method provides a new way of utilizing ePDF to obtain accurate local structure information from nanoparticles. Full Article text

noc ODIHR Director Link and IHRA Chair Constantinescu, on day to commemorate genocide against Roma and Sinti, say greater efforts needed to protect endangered memorial sites and ensure dignity of victims By feeds.osce.org Published On :: Tue, 02 Aug 2016 08:28:11 +0000 WARSAW / BUCHAREST, 2 August 2016 – Michael Georg Link, Director of the OSCE Office for Democratic Institutions and Human Rights (ODIHR) and Ambassador Mihnea Constantinescu, Chair of the International Holocaust Remembrance Alliance (IHRA), called today for greater efforts to protect endangered memorial sites related to the Roma and Sinti genocide during World War II. Speaking on the occasion of the commemoration of the liquidation of the “Gypsy family camp” at Auschwitz-Birkenau in 1944, when the close to 3,000 remaining Roma and Sinti in the camp were murdered, they stressed that states have to do more to demonstrate their sincere and strong commitment to education about and remembrance of the genocide. “Positively, we have seen increasing attention in recent years on the part of OSCE participating States to commemorate the Roma and Sinti genocide, and to educate people about this horrible event. This practice and these experiences should be widely shared and replicated,” Director Link said. “Promoting understanding of the Holocaust and its effect on different communities can help to create empathy and promote equality and non-discrimination for all.” “Accurate and ethical education about the Holocaust includes the respectful and dignified preservation of memorial sites,” said Ambassador Constantinescu. “States have to take resolute action to protect endangered memorial sites and continue to do more to commemorate the Roma and Sinti victims. The history of these endangered sites should be included as part of broader efforts to educate about the consequences of indifference to racism.” They called on governments to ensure that endangered memorial sites for Roma and Sinti victims are preserved and protected, to include this history as an integral part of civic and human rights education in their countries. They stressed that current developments, including a disturbing rise in xenophobic public rhetoric and racism, mean it is even more essential to build strong alliances among different communities. In 2003, with the Action Plan on Improving the Situation of Roma and Sinti within the OSCE Area, the OSCE participating States committed themselves to strengthen education about the Roma and Sinti genocide. Through its Contact Point for Roma and Sinti Issues, ODIHR promotes knowledge about and recognition of the plight of Roma and Sinti during the Holocaust to counter present-day discrimination and racism, and to promote tolerance. According to ODIHR’s recent publication Teaching about and Commemorating the Roma and Sinti Genocide: Practices within the OSCE Area, seven OSCE participating States officially commemorate the Roma and Sinti genocide on 2 August, while a larger number of states commemorate the Roma and Sinti victims on the International Holocaust Memorial Day, 27 January.
