Reply to Sysel et al.: Comment on the importance of using nitric oxide gas in the synthesis of nitrosylcobalamin and ICH-validated methods to assess purity and stability [Letters to the Editor]

In their comment (1) on our publication (2), the authors make two points: (i) they raise concerns about the possible effect of residual NONOate in our study, and (ii) they promote nitrosylcobalamin (NOCbl) supplied by their own company. Both points lack merit for the following reasons. The authors make the astonishing claim that the spectra of nitric oxide (NO•) and cobalamins overlap. Unlike NO•, cobalamin absorbs in the visible region, permitting unequivocal spectral assignment of NOCbl as reported (3). We demonstrated that whereas NOCbl is highly unstable in solution, it is stabilized by the B12 trafficking protein CblC. So even if present, residual NONOate (which is unstable at neutral pH and is removed during the work-up (3)) could not account for the observed difference.The authors then misrepresent our synthetic method, claiming that anaerobic conditions were used to generate nitrocobalamin (NO2Cbl), which results in the transient formation of NOCbl. We synthesized NO2Cbl aerobically using nitrite as described (4); NOCbl is not an intermediate in this ligand exchange reaction. The aerobic instability of NOCbl has been rigorously described by inorganic chemists (3, 5) and raises obvious questions about its purported biological effects as exemplified by the authors' own 2003 JBC publication, which was later withdrawn.As to promoting NOCbl from their company, the authors refer to a synthetic route from a mixture of NO• gas and aquocobalamin. Comment on the importance of using nitric oxide gas in the synthesis of nitrosylcobalamin and ICH-validated methods to assess purity and stability [Letters to the Editor]

After a thorough read of this paper (1), we wish to clarify that the authors' anaerobic method of synthesis for the production of nitrocobalamin results in the transient formation of nitrosylcobalamin, an unstable intermediate upon exposure to air. We concur that the authors' method results in the production of nitrocobalamin based on the UV-visible data as shown. The authors' adapted anaerobic method consists of mixing hydroxocobalamin hydrochloride with diethylamine NONOate diethylammonium salt in aqueous solution. Of concern, the UV spectrum of nitric oxide overlaps that of all cobalamin species under anaerobic conditions, making any assignments of the binding of nitric oxide to hydroxocobalamin suspect (2). Additionally, the use of acetone to precipitate the authors' product causes precipitation of diethylamine NONOate, resulting in an impure product. As a result, its utility for drawing experimental conclusions is faulty.The product from the authors' anaerobic synthetic method has not been assessed for purity, and the synthetic method itself has not been validated using a stability-indicating method as required by the International Conference on Harmonization (ICH) (ICH Q2B, Validation of Analytical Procedures) methodology, which is a hallmark for analytical characterization. Our nitrosylcobalamin synthesis involves reacting nitric oxide gas with hydroxocobalamin acetate as a heterogeneous mixture in a non-electron-donating solvent followed by rotary evaporation. Our nitrosylcobalamin product is stable in air, releases nitric oxide gas in situ (3), and meets ICH stability guidelines (4). Additionally, our nitrosylcobalamin product demonstrates biological activity, which has not been observed for nitrocobalamin (3, 5).

Opening ASBMB publications freely to all [Editorial]

We are extremely excited to announce on behalf of the American Society for Biochemistry and Molecular Biology (ASBMB) that the Journal of Biological Chemistry (JBC), Molecular & Cellular Proteomics (MCP), and the Journal of Lipid Research (JLR) will be published as fully open-access journals beginning in January 2021. This is a landmark decision that will have huge impact for readers and authors. As many of you know, many researchers have called for journals to become open access to facilitate scientific progress, and many funding agencies across the globe are either already requiring or considering a requirement that all scientific publications based on research they support be published in open-access journals. The ASBMB journals have long supported open access, making the accepted author versions of manuscripts immediately and permanently available, allowing authors to opt in to the immediate open publication of the final version of their paper, and endorsing the goals of the larger open-access movement (1). However, we are no longer satisfied with these measures. To live up to our goals as a scientific society, we want to freely distribute the scientific advances published in JBC, MCP, and JLR as widely and quickly as possible to support the scientific community. How better can we facilitate the dissemination of new information than to make our scientific content freely open to all?For ASBMB journals and others who have contemplated or made the transition to publishing all content open access, achieving this milestone generally requires new financial mechanisms. How to transfer etch/conductor delays from Allegro Package Designer (APD) to pin delays in Allegro PCB Editor

The packaging group has finished their design in Allegro Package Designer (APD) and I want to use the etch/conductor delay information from the mcm file in the board design in Allegro PCB Designer. Is there a method to do this?

This can be done by exporting the etch/conductor data from APD and importing it as PIN_DELAY information into Allegro PCB Editor.

If you are generating a length report for use in Allegro Pin Delay, you should consider changing the APD units to Mils and uncheck the Time Delay Report.

In Allegro Package Designer:

  1. Select File > Export > Board Level Component.
  2. Select HDL for the Output format and select OK.

       3. Choose a padstack for use when generating the component and select OK.

This will create a file, package_pin_delay.rpt, in the component subdirectory of the current working directory. This file will contain the etch/conductor delay information that can be imported into Allegro.

In Allegro PCB Editor:

  1. Make sure that the device you want to import delays to is placed in your board design and is visible.
  2. Select File > Import > Pin delay.
  3. Browse to the component directory and select package_pin_delay.rpt. The browser defaults to look for *.csv files so you will need to change the Files of type to *.* to select the file.
  4. You may be prompted with an error message stating that the component cannot be found and you should select one. If so, select the appropriate component.
  5. Select Import.
  6. Once the import is completed, select Close.

Note: It is important that all non-trace shapes have a VOLTAGE property so they will not be processed by the the 2D field solver. You should run Reports > Net Delay Report in APD prior to generating the board-level component. This will display the net name of each net as it is processed. If you miss a VOLTAGE property on a net, the net name will show in the report processing window, and you will know which net needs the property.

How to transfer custom title block from Orcad Capture to PCB Editor

Hi,

So I was trying to update the title block of a schematic that I have. The title block that was on there was out of date . I clicked on place --> title block and was able to find the title block that I need. I also have a .OLB file that contains that title block. Then I created a Netlist with the old BRD file as the input file (To keep it as is but modify changes) but when I do that I still do not see / cannot place the title block that I need. Under Place --> format symbols in Allegro , I do see a title block that is coming from the database (But it's the old one). I don't know what to do at this point and would appreciate any tips. I did make sure that the path to where the library is , is defined in the user preferences. 
I also tried copying the title block under the library folder in capture before creating my Netlist and that did not work either.

Purging duplicate vias in pcb editor

How do we purge/remove the duplicated vias in the same location of the PCB editor? These vias are not the one stacked and they are just blind vias running in internal layers 12-14. I find there is an additional copy of the blind via at the same location. Not sure what caused this issue.

Launch footprint editor from Capture or PCB Editor?

I'd like to be able to edit a footprint for a part in my design without needing to find the footprint filepath and directly open that file in PCB Editor. I see that I can view footprints from Capture, and that doing so shows me the footprint path, but I can't find any way to launch the editor. Is there any way to go directly from a part in a Capture schematic or a placed part in a PCB Editor board design to editing that part's footprint?

Display Resource Editor: Different Colors for Schematic and Layout Axis

Hi

In the environment I'm currently working, axes are shown for schematic, symbol, and layout views.For schematics and symbols, I'd prefer a dim gray, such that the axes are just visible but not dominant. For the layout, I'd prefer a brighter color. Is there a way to realize this? So far when I change the color of the 'axis' layer in the display resource editor, the axes in all three views get changed together:
