The study ‘The State of the Cryosphere 2024’, released on Tuesday on the sidelines of COP29 in Baku, urged urgent action to control emissions to save glaciers, which are melting at a rapid pace due to global warming. “Under a high emissions scenario…Hindu Kush Himalaya (HKH), may experience up to 80% of ice loss. With very low emissions however, up to 40% of glacier ice in the HKH region could be preserved,” it said, adding that projections in a few glacier regions even show slow re-growth beginning between 2100 and 2300, but only with very low emissions and essentially carbon neutrality by 2050. Against this backdrop, the environment ministers from the HKH met on Tuesday at the Baku Olympics Stadium to come together to save the “third pole” and to keep global temperatures below 1.5 Celsius. This gathering aimed to discuss the rapidly increasing climate risks and vulnerabilities in the region and beyond, while identifying areas for urgent collective actions, inevitable to addressing the pressing challenges and fulfilling the hopes of the quarter of humanity impacted by these changes, said a statement. It stated that over the past decade, the rate of glacier melting in the HKH has accelerated by 65 per cent compared to the previous decade (2000-2010) and the trend is projected to continue. “Over the last decade, the rate of glacier melting in the HKH has accelerated by 65% compared to the previous decade (2000- 2010), and the trend is projected to continue.” Speaking at the event, Bhutan Prime Minister Tshering Tobgay said this was an opportune time for the region to unite to push for a new collective quantified role that would directly address the need of the countries which were most vulnerable to climate change. Pakistan Prime Minister Adviser on Climate Change Romina Khurshid Alam said no country across the HKH region could tackle the climate crisis in isolation and besides regional unity, international response was essential. She said Pakistan stood for regional partnership aiming to save the ecosystem and species, and build climate resilience. She argued for easy access to climate finance to ensure these countries could erect safeguards to protect themselves from climate change. She said Pakistan was experiencing first-hand the impacts of climate change, increasing the risk of natural disasters in the form of GLOFs and threatening water security and agriculture as well as biodiversity. Other speakers included delegates from China, Bangladesh, India, and Nepal. The event was organised by the Kingdom of Bhutan and the International Centre for Integrated Mountain Development. Bleak state of Cryosphere According to the State of Cryosphere 2024 report, if the current Nationally Determined Contributions (NDCs) are met, global temperatures will likely reach 2.3°C by 2100, leading to irreversible ice loss, significant sea-level rise, and severe impacts on coastal regions, mountain communities, and polar ecosystems. In case of a high emissions scenario, the temperature may rise to 3-3.5°C, which will cause extreme damage, including rapid ice sheet loss, the disappearance of glaciers, and widespread permafrost thaw. However, the 1.5°C temperature in line with the Paris Agreement can help stabilise the cryosphere and preserve part of glaciers but that cannot happen unless there is a drastic cut in emissions. “This requires urgent action, however, with emergency-scale tightening of mitigation commitments and fossil fuel emissions declining 40% by 2030,” the report added. In many species, including humans, the poly(A) signal is an AAUAAA hexamer, but we recently found that the deeply branching eukaryote Giardia lamblia uses a distinct hexamer (AGURAA) and lacks any known auxiliary elements. Our discovery prompted us to explore the evolutionary dynamics of poly(A) signals and auxiliary elements in the eukaryotic kingdom. We use direct RNA sequencing to determine poly(A) signals for four protists within the Metamonada clade (which also contains G. lamblia) and two outgroup protists. These experiments reveal that the AAUAAA hexamer serves as the poly(A) signal in at least four different eukaryotic clades, indicating that it is likely the ancestral signal, whereas the unusual Giardia version is derived. We find that the use and relative strengths of auxiliary elements are also plastic; in fact, within Metamonada, species like G. lamblia make use of a previously unrecognized auxiliary element where nucleotides flanking the poly(A) signal itself specify genuine cleavage sites. Thus, despite the fundamental nature of pre-mRNA cleavage for the expression of all protein-coding genes, the motifs controlling this process are dynamic on evolutionary timescales, providing motivation for future biochemical and structural studies as well as new therapeutic angles to target eukaryotic pathogens. It was therefore proposed as a potential drug target for cancer therapy. In our previous study, it was found that low-dose X-ray and carbon ion irradiation both upregulated the expression of OATP family member OATP1A2 and in turn, led to a more dramatic killing effect when cancer cells were cotreated with antitumor drugs such as methotrexate. In the present study, the underlying mechanism of the phenomenon was explored in breast cancer cell line MCF-7. It was found that the nonreceptor tyrosine kinase v-YES-1 Yamaguchi sarcoma viral oncogene homolog 1 (YES-1) was temporally coordinated with the change of OATP1A2 after irradiation. The overexpression of YES-1 significantly increased OATP1A2 both at the mRNA and protein level. The signal transducer and activator of transcription 3 (STAT3) pathway is likely the downstream target of YES-1 because phosphorylation and nuclear accumulation of STAT3 were both enhanced after overexpressing YES-1 in MCF-7 cells. Further investigation revealed that there are two possible binding sites of STAT3 localized at the upstream sequence of SLCO1A2, the encoding gene of OATP1A2. Electrophoretic mobility shift assay and chromatin immunoprecipitation analysis suggested that these two sites bound to STAT3 specifically and the overexpression of YES-1 significantly increased the association of the transcription factor with the putative binding sites. Finally, inhibition or knockdown of YES-1 attenuated the induction effect of radiation on the expression of OATP1A2. SIGNIFICANCE STATEMENT The present study found that the effect of X-rays on v-YES-1 Yamaguchi sarcoma viral oncogene homolog 1 (YES-1) and organic anion transporting polypeptides (OATP)1A2 was temporally coordinated. YES-1 phosphorylates and increases the nuclear accumulation of signal transducer and activator of transcription 3, which in turn binds to the upstream regulatory sequences of SLCO1A2, the coding gene for OATP1A2. Amnesty International has accused Myanmar’s security forces of killing hundreds of men, women and children during a systematic campaign to expel the Rohingya. More than 580,000 refugees have fled to neighboring Bangladesh since late August. “We really hold the military leadership accountable for what’s happening,” Tillerson said at the Center for Strategic and International Studies, a Washington think tank. “What’s most important to us is that the world can’t just stand idly by and be witness to the atrocities that are being reported in that area.” He also called Wednesday for the U.S. and India to expand strategic ties. He pointedly criticized China, which he accused of challenging international norms needed for global stability. He said the world needed the U.S. and India to have a strong partnership. The U.N. has called the actions taken by Myanmar forces against the group “a textbook example of ethnic cleansing.” A report released by Amnesty International on Wednesday documents widespread rape, killings and burnings of Rohingya across the Rakhine State in Myanmar. The report includes extensive interviews of Rohingya refugees who tell stories of live burnings, sexual violence and mass shootings at the hands of soldiers. To escape persecution, Rohingya refugees are fleeing in droves to neighboring Bangladesh, a country described by some as a reluctant host for the thousands of refugees behind its borders. Conditions within Bangladesh show refugee camps beyond capacity, as organizations struggle to keep up with humanitarian aid. They are not considered one of the country’s official ethnic groups. As such, their lack of official identity bars them from government services and travel.
