Further evaluation of the shape of atomic Hirshfeld surfaces: M⋯H contacts and homoatomic bonds

It is well known that Hirshfeld surfaces provide an easy and straightforward way of analysing inter­molecular inter­actions in the crystal environment. The use of atomic Hirshfeld surfaces has also demonstrated that such surfaces carry information related to chemical bonds which allow a deeper evaluation of the structures. Here we briefly summarize the approach of atomic Hirshfeld surfaces while further evaluating the kind of information that can be retrieved from them. We show that the analysis of the metal-centre Hirshfeld surfaces from structures refined via Hirshfeld Atom Refinement (HAR) allow accurate evaluation of contacts of type M⋯H, and that such contacts can be related to the overall shape of the surfaces. The com­pounds analysed were tetra­aqua­bis­(3-carb­oxy­propionato)metal(II), [M(C4H3O4)2(H2O)4], for metal(II)/M = manganese/Mn, cobalt/Co, nickel/Ni and zinc/Zn. We also evaluate the sensitivity of the surfaces by an investigation of seemingly flat surfaces through analysis of the curvature functions in the direction of C—C bonds. The obtained values not only demonstrate variations in curvature but also show a correlation with the hybridization of the C atoms involved in the bond.

Site-specific contacts enable distinct modes of TRPV1 regulation by the potassium channel Kv{beta}1 subunit [Molecular Biophysics]

Transient receptor potential vanilloid 1 (TRPV1) channel is a multimodal receptor that is responsible for nociceptive, thermal, and mechanical sensations. However, which biomolecular partners specifically interact with TRPV1 remains to be elucidated. Here, we used cDNA library screening of genes from mouse dorsal root ganglia combined with patch-clamp electrophysiology to identify the voltage-gated potassium channel auxiliary subunit Kvβ1 physically interacting with TRPV1 channel and regulating its function. The interaction was validated in situ using endogenous dorsal root ganglia neurons, as well as a recombinant expression model in HEK 293T cells. The presence of Kvβ1 enhanced the expression stability of TRPV1 channels on the plasma membrane and the nociceptive current density. Surprisingly, Kvβ1 interaction also shifted the temperature threshold for TRPV1 thermal activation. Using site-specific mapping, we further revealed that Kvβ1 interacted with the membrane-distal domain and membrane-proximal domain of TRPV1 to regulate its membrane expression and temperature-activation threshold, respectively. Our data therefore suggest that Kvβ1 is a key element in the TRPV1 signaling complex and exerts dual regulatory effects in a site-specific manner.

HARI SREENIVASAN: In the day’s other news: Attorney General Jeff Sessions insisted he never lied to the Senate Judiciary Committee about his conversations with the Russian ambassador during the presidential campaign. At a hearing today, he bridled at Democratic Senator Al Franken’s accusation that he’d — quote — “moved the goalposts” on the nature of his discussions.

SEN. AL FRANKEN, D-Minn.: First it was, I didn’t have communications with Russians, which wasn’t true. Then it was, I never met with any Russians to discuss any political campaign, which may or may not be true.

Now it’s, I didn’t discuss interference in the campaign.

JEFF SESSIONS, Attorney General: Well, let me just say without hesitation, that I conducted no improper discussions with the Russians at any time regarding the campaign or any other item facing this country.

HARI SREENIVASAN: Sessions has recused himself from the Justice Department’s investigation into Russia’s election meddling.

President Trump had new criticism today for former FBI Director James Comey over the Hillary Clinton e-mail probe. He complained again that Comey decided to clear Clinton before she was even interviewed. That’s based on newly released draft statements by Comey from May of 2016. FBI officials say it was already clear that no charges were warranted.

On another issue, the president faced fallout over the death of Army Sergeant La David Johnson in Niger this month. Congresswoman Frederica Wilson says she was with Mrs. Johnson when the president called. The Florida Democrat told The Washington Post that Mr. Trump said — quote — “He knew what he was signing up for, but I guess it hurts anyway.”

The sergeant’s mother confirmed it, but the president denied it, and White House Press Secretary Sarah Huckabee Sanders went after Wilson.

SARAH SANDERS, White House Press Secretary: This is a president who loves our country very much, who has the greatest level of respect for men and women in uniform and wanted to call and offer condolences to the family, and I think to try to create something from that, that the congresswoman is doing, is, frankly, appalling and disgusting.

HARI SREENIVASAN: The Post also reported on another incident today. It quoted the father of a soldier killed in Afghanistan as saying the president offered $25,000 from his personal account, but never followed through.

The death toll in Northern California’s wildfires rose to 42 today. Officials in Sonoma County found the remains of the latest victim, as they searched hundreds of burned homes. Meanwhile, fire crews made new gains overnight with the help of cooler weather and low winds.

A two-time Olympic medalist says the former team doctor for U.S. women’s gymnastics sexually abused her for years. McKayla Maroney is the highest profile athlete to come forward in the scandal. In a statement today, she said Dr. Larry Nassar began molesting her when she was just 13. He’s awaiting sentencing on a child pornography charge, but has denied any sexual abuse.

More questions tonight about drug pricing. A new study finds the costs of injectable cancer drugs, approved since 1996, rose an average of 25 percent over eight years. That’s far higher than the rate of inflation. The study was based at Emory University and published in “The Journal of Clinical Oncology.”

And on Wall Street, health insurers and IBM fueled a surge in stocks today. The Dow Jones industrial average gained 160 points, more than half-a-percent, to close above 23000 for the first time. The Nasdaq rose just a fraction, and the S&P 500 was up two points.

The title tetra­nuclear copper complex, [Cu4Cl6O(C6H9N3O3)4] or [Cu4Cl6O­(MET)4] [MET is 1-(2-hy­droxy­eth­yl)-2-methyl-5-nitro-1H-imidazole or metronidazole], contains a tetra­hedral arrangement of copper(II) ions. Each copper atom is also linked to the other three copper atoms in the tetra­hedron via bridging chloride ions. A fifth coordination position on each metal atom is occupied by a nitro­gen atom of the monodentate MET ligand. The result is a distorted CuCl3NO trigonal–bipyramidal coordination polyhedron with the axial positions occupied by oxygen and nitro­gen atoms. The extended structure displays O—H⋯O hydrogen bonding, as well as unusual short O⋯N inter­actions [2.775 (4) Å] between the nitro groups of adjacent clusters that are oriented perpendicular to each other. The scattering contribution of disordered water and methanol solvent mol­ecules was removed using the SQUEEZE procedure [Spek (2015). Acta Cryst. C71, 9–16] in PLATON [Spek (2009). Acta Cryst. Osh6 and Osh7 are lipid transfer proteins (LTPs) that move phosphatidylserine (PS) from the endoplasmic reticulum (ER) to the plasma membrane (PM). High PS level at the PM is key for many cellular functions. Intriguingly, Osh6/7 localize to ER-PM contact sites, although they lack membrane-targeting motifs, in contrast to multidomain LTPs that both bridge membranes and convey lipids. We show that Osh6 localization to contact sites depends on its interaction with the cytosolic tail of the ER-PM tether Ist2, a homologue of TMEM16 proteins. We identify a motif in the Ist2 tail, conserved in yeasts, as the Osh6-binding region, and we map an Ist2-binding surface on Osh6. Mutations in the Ist2 tail phenocopy osh6 osh7 deletion: they decrease cellular PS levels, and block PS transport to the PM. Our study unveils an unexpected partnership between a TMEM16-like protein and a soluble LTP, which together mediate lipid transport at contact sites.

Seizing the Opportunity to Expand People to People Contacts in Cuba

INTRODUCTION 

Last year, President Obama delivered the first step in his promise to reach out to the Cuban people and support their desire for freedom and self-determination. Premised on the belief that Cuban Americans are our best ambassadors for freedom in Cuba, the Obama administration lifted restrictions on travel and remittances by Cuban Americans. The pent-up demand for Cuban American contact with the island revealed itself: within three months of the new policy, 300,000 Cuban Americans traveled to Havana -- 50,000 more than for all of the previous year.  Experts estimate that over $600 million in annual remittances has flowed from the United States to Cuba in 2008 and 2009 and informal flows of consumer goods is expanding rapidly.

The administration’s new policy has the potential to create new conditions for change in Cuba. However, if U.S. policy is to be truly forward looking it must further expand its focus from the Castro government to the well-being of the Cuban people. Recent developments on the island, including the ongoing release of dozens of political prisoners, have helped create the right political moment to take action.  The administration should institute a cultural diplomacy strategy that authorizes a broad cross-section of American private citizens and civil society to travel to the island to engage Cuban society and share their experiences as citizens of a democratic country.  Reducing restrictions on people-to-people contact is not a “concession,” but a strategic tool to advance U.S. policy objectives to support the emergence of a Cuban nation in which the Cuban people determine their political and economic future.

The President has the authority to reinstate a wide range of “purposeful,” non-touristic travel to Cuba in order to implement a cultural diplomacy strategy. Under President Clinton, the Baltimore Orioles played baseball in Havana and in return the Cuban national team was invited to Baltimore. U.S. students studied abroad in Cuba and engaged in lively discussions with their fellow students and host families. U.S. religious groups provided food and medicines to community organizations, helping them assist their membership.  However, in 2004, such travel was curtailed, severely limiting U.S. insights about the needs, interests and organizational capacities of community groups and grassroots organizations. Today, visitors traveling under an educational license, for example, number a meager 2,000 annually.

COVID-19: BMC slow to test high-risk contacts in Govandi

It took the BMC a full week to quarantine over two dozen high-risk patients, including the family of a Govandi resident who was posthumously found to have COVID-19. Right after the woman's death on Sunday, six neighbours got themselves tested at a fever camp and four returned positive over the next few days.

The woman, 55, was admitted at Rajawadi Hospital on April 24 and died on Sunday. A local social group identified 21 people as her high-risk contacts with symptoms.

While protocol states that family members and high-risk contacts should be tested and quarantined immediately, around 25 people were isolated at Vishnu Nagar in Mahul village only on Friday.

Of the six that visited the Mankhurd fever camp on Monday, one tested positive on Wednesday and was quarantined, two on Thursday. One positive diabetic patient is still at home as they require heightened care at a hospital.

Shankar Patil, president of the Govandi-based social group Rahul Seva Mandal, who highlighted the case in an email to CM Uddhav Thackeray on April 29, said, "The woman already had hypertension and diabetes. Later, her relatives, some family members and neighbours started showing symptoms. All live in Lumbini Baug in Panchsheel chawl so it is impossible to practise social distancing. They also use common toilets."


A neighbour whose husband has tested positive

The mandal had conducted its survey in the chawl on Saturday. Shankar said, "Had they not decided to get themselves tested, it would have been impossible to contain the spread."

'BMC's efforts taking time'

A neighbour — whose husband is one of the positive patients — went to the fever camp on Friday. She has a fever of almost 103°C. "We did not know that the woman had COVID-19. My husband, cousins and other family members took care of her as she already had diabetes," she said.

"After her death, BMC visited the chawl to take the names of high-risk contacts and said they will visit again the next day but no one came. All 21 people should have been quarantined immediately."

"We usually go out to buy vegetables and if we are positive, we don't want to spread the disease. Most of the people who tested positive were showing mild symptoms. They don't have too high a fever or difficulty breathing," she said. "I understand if the government doesn't test or quarantine neighbours, but at least test those who live in the same house. BMC is doing its work — but it is slow. We cannot directly blame them."

'Testing symptomatic cases'

M East ward Assistant Commissioner Sudhanshu Dwivedi said efforts was doubled at the fever camp in KC International School on Friday. When asked about the lack of testing of high-risk contacts, he said, "The ICMR guidelines say only symptomatic cases need to be tested. Earlier, we had set up a fever camp in a one-km radius of the chawl. It was in Govandi but in surrounding areas such as Deonar, Lotus Colony, etc. We also barricaded the chawl on Saturday, a day after the deceased woman was taken to Rajawadi."

"We also sent out Community Health Volunteers to check on residents' health. They told residents that if anyone is feeling symptomatic, they should visit the nearest fever camp and contact them.
