My UVM Adapter looks like: Issue: When I use basic reg.write, my write access are working well, as that is managed by the driver i.e. once adapter gives the packet to the driver, the driver supplies the address and the control signals to the DUT on the first clock cycle and then the write data on the next clock cycle. But when I am performing the read operation, somehow the UVM adapter is reading the data at the same clock cycle where read address + Controls are supplied and this is triggering read failure messages from the `uvm_reg_hw_reset_seq` sequence. What should I modify in the driver/sequencer/adapter so that the UVM adapter can read the data on the next cycle instead of the same clock cycle. Just FYI: The waveforms of the read operation are correct, it is just the Adapter and the `uvm_reg_hw_reset_seq`. The AHB Driver + AHB Monitor is fully proven and verified to be working correctly. axi Factors Influencing the Central Nervous System (CNS) Distribution of the Ataxia Telangiectasia Mutated and Rad3-Related Inhibitor Elimusertib (BAY1895344): Implications for the Treatment of CNS Tumors [Metabolism, Transport, and Pharmacogenetics] By jpet.aspetjournals.org Published On :: 2024-10-18T07:04:15-07:00 Glioblastoma (GBM) is a disease of the whole brain, with infiltrative tumor cells protected by an intact blood-brain barrier (BBB). GBM has a poor prognosis despite aggressive treatment, in part due to the lack of adequate drug permeability at the BBB. Standard of care GBM therapies include radiation and cytotoxic chemotherapy that lead to DNA damage. Subsequent activation of DNA damage response (DDR) pathways can induce resistance. Various DDR inhibitors, targeting the key regulators of these pathways such as ataxia telangiectasia mutated and Rad3-related (ATR), are being explored as radio- and chemosensitizers. Elimusertib, a novel ATR kinase inhibitor, can prevent repair of damaged DNA, increasing efficacy of DNA-damaging cytotoxic therapies. Robust synergy was observed in vitro when elimusertib was combined with the DNA-damaging agent temozolomide; however, we did not observe improvement with this combination in in vivo efficacy studies in GBM orthotopic tumor-bearing mice. This in vitro–in vivo disconnect was explored to understand factors influencing central nervous system (CNS) distribution of elimusertib and reasons for lack of efficacy. We observed that elimusertib is rapidly cleared from systemic circulation in mice and would not maintain adequate exposure in the CNS for efficacious combination therapy with temozolomide. CNS distribution of elimusertib is partially limited by P-glycoprotein efflux at the BBB, and high binding to CNS tissues leads to low levels of pharmacologically active (unbound) drug in the brain. Acknowledging the potential for interspecies differences in pharmacokinetics, these data suggest that clinical translation of elimusertib in combination with temozolomide for treatment of GBM may be limited. SIGNIFICANCE STATEMENT This study examined the disconnect between the in vitro synergy and in vivo efficacy of elimusertib/temozolomide combination therapy by exploring systemic and central nervous system (CNS) distributional pharmacokinetics. Results indicate that the lack of improvement in in vivo efficacy in glioblastoma (GBM) patient-derived xenograft (PDX) models could be attributed to inadequate exposure of pharmacologically active drug concentrations in the CNS. These observations can guide further exploration of elimusertib for the treatment of GBM or other CNS tumors. Full Article

In this report, we present a family with three affected individuals found to have a heterozygous missense c.800C > T (predicted p.Thr267Met) who present clinically with a SCA29-like syndrome. All three individuals presented with varying degrees of ataxia, developmental delay, and apparent intellectual disability, as well as craniofacial involvement—an uncommon finding in patients with SCA29. The variant was identified using clinical exome sequencing and validated with Sanger sequencing. It is presumed to be inherited via parental germline mosaicism. We present our findings to provide additional evidence for germline mosaic inheritance of SCA29, as well as to expand the clinical phenotype of the syndrome. Full Article

Yesterday I spent a few hours tweaking various text attributes like kerning and quote height, and finished up the two-column version of the poster. It’ll be a 16″x20″ thing, and will look something like this… If you want to get your hands on one of these posters, perhaps for the wall of your office, or maybe the local kindergarten, jump in on the Backerkit project today. We’ll be printing extras, of course, but backing the project is the only way to ensure that we set one aside for you. And speaking of Backerkit… this project is an experiment, a stress-test of a new soup-to-nuts crowdfunding service, an alternative to Kickstarter. For several projects we’ve used Backerkit in conjunction with Kickstarter, because Backerkit makes fulfilment easier for complex projects. They’ve been around for a while, and we love working with them. We still like working with Kickstarter, but it’s good to have an alternative—especially since Kickstarter briefly flirted with adding NFTs to their blockchain infrastructure, sending much of their community scrambling for other options. They’ve backed away from that ledge, at least for now, which makes us happy. Also, we are happy to be trying out a different service. We like having options. Unsurprisingly, there are a couple of maxims that may apply here: 50: If it only works in exactly the way the manufacturer intended, it is defective. 30: A little trust goes a long way. Here, then is a nice origin story for you, the origin of Maxim 32. I was signing and sketching at GenCon Indy a decade or so ago when some young men approached the table. They were what a friend of mine likes to call “baby sailors”: relatively new members of the US Navy. One of them said “we have a suggestion for a maxim.” I smiled. “Let’s hear it.” “Anything is amphibious if you can fit it into an AAV.” I chuckled. “That’s pretty good, but the term ‘AAV’ is too specific for Schlock Mercenary use.” Then I went silent, stared off into the distance, and I guess this made everyone uncomfortable because our Booth Captain, Darren, spoke next. “Shhh… don’t interrupt him. The magic is happening.” It’s true, I’d been wondering how this US Navy aphorism could be repurposed, but I had expected to be able to mull it over all day. Now, however, Darren had turned it into the promise of performance art. Did I curse silently? Maybe. I don’t remember, because I was panicking. Still staring into space, trying not to show fear, I dove into the “formulae” for the maxims. I knew that many of the maxims were subversions of existing aphorisms. Several of them formed thematic couplets, like Maxims 2 and 3 (“a sergeant in motion” and “an ordnance tech in motion”) are a great example of this. And Maxim 23, “Anything is air-droppable at least once,” seemed like a good candidate for pairing with what the Navy boys had suggested, especially since “Anything is air-droppable” and “Anything is amphibious” were already pretty close. All I needed to do was break the amphibious-ness in the same way I’d broken the air-droppability… and I think it was that moment, when I contemplated “breaking” amphibious-ness, when the final text arrived in my head. “Anything is amphibious if you can get it back out of the water.” A quick note. The United States Navy exists to keep things DRY. Everything except the hulls, really. The very idea of dropping something into the water that is not already a boat, runs counter to Navy thinking. So it’s no surprise that those Navy boys were visibly horrified by my subversion of their aphorism. “That’s terrible” one of them said. And then they started to laugh. And then I wrote Maxim 32 in my notebook, because obviously it was perfect. Full Article
