US carried out strikes against Iranian-linked targets in Syria

washington — The U.S. military said Monday that it had carried out strikes against nine targets associated with Iranian groups in Syria.  In a statement, the U.S. military said the strikes were made against two locations in Syria and were a response to several attacks on U.S. personnel in Syria in the past 24 hours.  The U.S. has occasionally carried out strikes against targets linked to Iran in both Iraq and Syria. In February, the U.S. launched airstrikes in Iraq and Syria against more than 85 targets linked to Iran's Islamic Revolutionary Guard Corps and militias it backs, in retaliation for a deadly attack on U.S. troops.  "These strikes will degrade the Iranian backed groups' ability to plan and launch future attacks on U.S. and coalition forces," the U.S. military said after the most recent strikes.  The U.S. has 900 troops in Syria and 2,500 in neighboring Iraq, on a mission to advise and assist local forces trying to prevent a resurgence of Islamic State, which in 2014 seized large swaths of both countries but was later defeated.  The U.S. has sent warships and fighter aircraft to the region since the Israel-Hamas conflict erupted on October 7, 2023, to try to deter Iran and Iran-backed groups from getting involved in the conflict.  U.S. forces have also helped shoot down projectiles that Iran launched toward Israel this year.

the hague, netherlands — Dozens of people armed with sticks and firecrackers set a tram on fire in Amsterdam on Monday, police said, while the city is facing tensions following violence last week targeting fans of an Israeli soccer club. Police said the fire was quickly extinguished and riot officers cleared the square. Images online showed people damaging property and setting off firecrackers. Police said it was not clear who started the unrest or whether it was related to what happened last week. But they noted the tense atmosphere since five people were treated in the hospital and dozens detained Thursday following a Maccabi Tel Aviv-Ajax match. Youths on scooters and on foot went in search of Israeli fans, punching and kicking them and then fleeing to evade police, according to Amsterdam’s mayor. Dutch police announced five new arrests Monday in their investigation into that earlier violence. The suspects are men aged 18 to 37 and are from Amsterdam or surrounding cities. Four are still in custody; the fifth has been released but remains a suspect. Earlier, police said that four other men who had been arrested last week would remain in custody while the investigation continued. Two of those are minors, a 16-year-old and a 17-year-old from Amsterdam. The other two men are from Amsterdam and a nearby city. Police said they have identified over 170 witnesses and have taken forensics evidence from dozens. Prime Minister Dick Schoof said they were also examining videos posted to social media. Reports of antisemitic speech, vandalism and violence have been on the rise in Europe since the start of the war in Gaza, and tensions mounted in Amsterdam ahead of Thursday night’s match. Pro-Palestinian demonstrators were banned by local authorities from gathering outside the stadium. Before the match, Maccabi fans also tore a Palestinian flag off a building in Amsterdam and chanted anti-Arab slogans on their way to the stadium. There were also reports of Maccabi fans starting fights. The mayor has banned all demonstrations in the city and declared several parts of Amsterdam risk zones where police can stop and check anyone. Dozens were detained on Sunday for taking part in a pro-Palestinian demonstration in central Amsterdam that had been outlawed. Israeli Foreign Minister Gideon Saar rushed to the Netherlands on Friday and offered Israel’s help in the police investigation. He met on Saturday with the Dutch prime minister and said in a statement that the attacks and demands to show passports “were reminiscent of dark periods in history.”

Chromatin interaction maps identify oncogenic targets of enhancer duplications in cancer [RESEARCH]

As a major type of structural variants, tandem duplication plays a critical role in tumorigenesis by increasing oncogene dosage. Recent work has revealed that noncoding enhancers are also affected by duplications leading to the activation of oncogenes that are inside or outside of the duplicated regions. However, the prevalence of enhancer duplication and the identity of their target genes remains largely unknown in the cancer genome. Here, by analyzing whole-genome sequencing data in a non-gene-centric manner, we identify 881 duplication hotspots in 13 major cancer types, most of which do not contain protein-coding genes. We show that the hotspots are enriched with distal enhancer elements and are highly lineage-specific. We develop a HiChIP-based methodology that navigates enhancer–promoter contact maps to prioritize the target genes for the duplication hotspots harboring enhancer elements. The methodology identifies many novel enhancer duplication events activating oncogenes such as ESR1, FOXA1, GATA3, GATA6, TP63, and VEGFA, as well as potentially novel oncogenes such as GRHL2, IRF2BP2, and CREB3L1. In particular, we identify a duplication hotspot on Chromosome 10p15 harboring a cluster of enhancers, which skips over two genes, through a long-range chromatin interaction, to activate an oncogenic isoform of the NET1 gene to promote migration of gastric cancer cells. Focusing on tandem duplications, our study substantially extends the catalog of noncoding driver alterations in multiple cancer types, revealing attractive targets for functional characterization and therapeutic intervention.

Targeted and complete genomic sequencing of the major histocompatibility complex in haplotypic form of individual heterozygous samples [RESEARCH]

The human major histocompatibility complex (MHC) is a ~4 Mb genomic segment on Chromosome 6 that plays a pivotal role in the immune response. Despite its importance in various traits and diseases, its complex nature makes it challenging to accurately characterize on a routine basis. We present a novel approach allowing targeted sequencing and de novo haplotypic assembly of the MHC region in heterozygous samples, using long-read sequencing technologies. Our approach is validated using two reference samples, two family trios, and an African-American sample. We achieved excellent coverage (96.6%–99.9% with at least 30x depth) and high accuracy (99.89%–99.99%) for the different haplotypes. This methodology offers a reliable and cost-effective method for sequencing and fully characterizing the MHC without the need for whole-genome sequencing, facilitating broader studies on this important genomic segment and having significant implications in immunology, genetics, and medicine.

Best Patient Care Practices for Administering PSMA-Targeted Radiopharmaceutical Therapy

Optimal patient management protocols for metastatic castration-resistant prostate cancer (mCRPC) are poorly defined and even further complexified with new therapy approvals, such as radiopharmaceuticals. The prostate-specific membrane antigen (PSMA)–targeted agent 177Lu vipivotide tetraxetan ([177Lu]Lu-PSMA-617), approved after the phase III VISION study, presents physicians with additional aspects of patient management, including specific adverse event (AE) monitoring and management, as well as radiation safety. Drawing on our experience as VISION study investigators, here we provide guidance on best practices for delivering PSMA-targeted radiopharmaceutical therapy (RPT) to patients with mCRPC. After a comprehensive review of published evidence and guidelines on RPT management in prostate cancer, we identified educational gaps in managing the radiation safety and AEs associated with [177Lu]Lu-PSMA-617. Our results showed that providing sufficient education on AEs (e.g., fatigue and dry mouth) and radiation safety principles is key to effective delivery and management of patient expectations. Patient counseling by health care professionals, across disciplines, is a cornerstone of optimal patient management during PSMA-targeted RPT. Multidisciplinary collaboration is crucial, and physicians must adhere to radiation safety protocols and counsel patients on radiation safety considerations. Treatment with [177Lu]Lu-PSMA-617 is generally well tolerated; however, additional interventions may be required, such as dosing modification, medications, or transfusions. Urinary incontinence can be challenging in the context of radiation safety. Multidisciplinary collaboration between medical oncologists and nuclear medicine teams ensures that patients are monitored and managed safely and efficiently. In clinical practice, the benefit-to-risk ratio should always be evaluated on a case-by-case basis.

Somatostatin: Linking Cognition and Alzheimer Disease to Therapeutic Targeting [Review Article]

Over 4 decades of research support the link between Alzheimer disease (AD) and somatostatin [somatotropin-releasing inhibitory factor (SRIF)]. SRIF and SRIF-expressing neurons play an essential role in brain function, modulating hippocampal activity and memory formation. Loss of SRIF and SRIF-expressing neurons in the brain rests at the center of a series of interdependent pathological events driven by amyloid-β peptide (Aβ), culminating in cognitive decline and dementia. The connection between the SRIF and AD further extends to the neuropsychiatric symptoms, seizure activity, and inflammation, whereas preclinical AD investigations show SRIF or SRIF receptor agonist administration capable of enhancing cognition. SRIF receptor subtype-4 activation in particular presents unique attributes, with the potential to mitigate learning and memory decline, reduce comorbid symptoms, and enhance enzymatic degradation of Aβ in the brain. Here, we review the links between SRIF and AD along with the therapeutic implications.

Significance Statement

Somatostatin and somatostatin-expressing neurons in the brain are extensively involved in cognition. Loss of somatostatin and somatostatin-expressing neurons in Alzheimer disease rests at the center of a series of interdependent pathological events contributing to cognitive decline and dementia. Targeting somatostatin-mediated processes has significant therapeutic potential for the treatment of Alzheimer disease.

Neuroactive Kynurenines as Pharmacological Targets: New Experimental Tools and Exciting Therapeutic Opportunities [75th Anniversary Celebration Collection Special Section]

Both preclinical and clinical studies implicate functional impairments of several neuroactive metabolites of the kynurenine pathway (KP), the major degradative cascade of the essential amino acid tryptophan in mammals, in the pathophysiology of neurologic and psychiatric diseases. A number of KP enzymes, such as tryptophan 2,3-dioxygenase (TDO2), indoleamine 2,3-dioxygenases (IDO1 and IDO2), kynurenine aminotransferases (KATs), kynurenine 3-monooxygenase (KMO), 3-hydroxyanthranilic acid oxygenase (3-HAO), and quinolinic acid phosphoribosyltransferase (QPRT), control brain KP metabolism in health and disease and are therefore increasingly considered to be promising targets for the treatment of disorders of the nervous system. Understanding the distribution, cellular expression, and regulation of KP enzymes and KP metabolites in the brain is therefore critical for the conceptualization and implementation of successful therapeutic strategies.

Significance Statement

Studies have implicated the kynurenine pathway of tryptophan in the pathophysiology of neurologic and psychiatric diseases. Key enzymes of the kynurenine pathway regulate brain metabolism in both health and disease, making them promising targets for treating these disorders. Therefore, understanding the distribution, cellular expression, and regulation of these enzymes and metabolites in the brain is critical for developing effective therapeutic strategies. This review endeavors to describe these processes in detail.:
