YY1 knockout in pro-B cells impairs lineage commitment, enabling unusual hematopoietic lineage plasticity [Research Papers]

During B-cell development, cells progress through multiple developmental stages, with the pro-B-cell stage defining commitment to the B-cell lineage. YY1 is a ubiquitous transcription factor that is capable of both activation and repression functions. We found here that knockout of YY1 at the pro-B-cell stage eliminates B lineage commitment. YY1 knockout pro-B cells can generate T lineage cells in vitro using the OP9-DL4 feeder system and in vivo after injection into sublethally irradiated Rag1–/– mice. These T lineage-like cells lose their B lineage transcript profile and gain a T-cell lineage profile. Single-cell RNA-seq experiments showed that as YY1 knockout pro-B cells transition into T lineage cells in vitro, various cell clusters adopt transcript profiles representing a multiplicity of hematopoietic lineages, indicating unusual lineage plasticity. In addition, YY1 KO pro-B cells in vivo can give rise to other hematopoietic lineages in vivo. Evaluation of RNA-seq, scRNA-seq, ChIP-seq, and scATAC-seq data indicates that YY1 controls numerous chromatin-modifying proteins leading to increased accessibility of alternative lineage genes in YY1 knockout pro-B cells. Given the ubiquitous nature of YY1 and its dual activation and repression functions, YY1 may regulate commitment in multiple cell lineages.

Chrome for iOS now lets you add text to Google Lens visual searches

If you use Chrome on your iPhone, you’re about to see some features sliding over from the Android version. Google Lens will let you add text to your image searches, and you can save files and pictures directly to Google Drive and Photos. You can get “Shopping Insights” for products you’re browsing.

Chrome for iOS now lets you add words to your Google Lens visual searches, allowing you to add nuance to your query or “perform more complex and specific searches,” as Chrome Product Manager Katia Muradyan wrote in a blog post. After activating Lens by tapping the camera icon in the Chrome search bar, you can ask questions about the object you’re snapping a pic of, and it will produce corresponding results. Google says AI Overviews will also appear for some of these search results.

The feature shares some common ground with an Apple Intelligence feature for iPhone 16 owners in iOS 18.2, which is currently in beta. Visual Intelligence lets you point your camera at something and get info about it, including asking ChatGPT questions about it or searching for it on Google.

Chrome for iPhone now has a feature that lets you save a file directly to Google Drive or Google Photos, sparing you from using your phone’s internal storage. When saving files from Chrome, you’ll see a new option to save the file to Drive. Similarly, when browsing a photo you want to save, long-press on it, and you’ll see a new “Save in Google Photos” option in the context menu. Of course, the feature requires you to be signed into a Google account.

Chrome for iOS also adds a feature that pops up a mini-map when you click on an address. Look for an underlined link to specific addresses; clicking on it will take you to the mini-map without leaving the browser.

Finally, Google is adding Shopping Insights for US users. The company frames it as a way to help you find great prices on items you’re shopping for, but it’s hard to imagine this feature exists strictly from the kindness of Google's heart. Regardless, you’ll soon see a “Good Deal Now” alert in Chrome’s address bar when browsing for products for which it’s available. You’ll see details like price history / tracking and buying options if you tap it.

IMF holds unusual talks with Pakistan over $9.4 billion bailout

ISLAMABAD — The International Monetary Fund's Pakistan mission chief Nathan Porter on Tuesday (Nov 12) opened unusual talks with Pakistan over a US$7 billion (S$9.4 billion) bailout approved by its board in September, the finance ministry and sources said. The unscheduled visit of the IMF mission and talks beginning with meeting the country's finance team are too early for first review of the IMF's Extended Fund Facility (EFF), which is due in the first quarter of 2025. The chiefs of Pakistan's central bank and federal board of revenue also attended the meeting besides other officials from both the sides, the statement said. The ministry and the IMF have not officially released details of the visit. Sources in the finance ministry said the Nov 11-15 visit will discuss recent developments and programme performance to date, adding the mission was not part of the first review. The sources declined to be identified as they were not authorised to speak with the media. Pakistan has been struggling with boom-and-bust economic cycles for decades, leading to 23 IMF bailouts since 1958.

A Visual Approach to Help Pages

As the functionality and scope of Web sites and applications has grown over the years, so has the prevalence of Help pages. Nearly every feature has an explanatory article outlining how to use it and why. But most Help pages are walls of text making them hard to act on. So a few years ago, we tried something different.

First let's look at the status quo. This Help page from Amazon is both pretty typical and by those standards, pretty good. It's specific to one topic, brief, outlines steps clearly, and includes links to help people accomplish their intended task. Companies iterated to these kinds of Help pages because they mostly work and because they're less work.

Keeping Help text up to date and accurate is less labor-intensive than updating images or videos with the same information. But as the old saying goes, a picture is worth a lot of words and there's a reason many people turn to video tutorials to learn how to do things instead of reading about how to do them.

When building Polar several years ago, we wanted a more approachable and fun way of helping people learn how to use our product. And while you might say "the best Help pages are no Help pages -just make your app easy to use" not all Help pages are smearing over usability issues. Some introduce higher level concepts, others outline capabilities, and some serve as marketing for specific features.

So with those goals in mind, we iterated to a simple formula. Each concept or feature gets a Help page that has a title alongside 1-2 sentences and as many sections consisting of a title, 1-2 sentences, plus a graphic as needed.

This approach meant people primarily relied on images (or their alt tags if visually impaired) to figure out how to get things done. So we iterated a fair amount on the images to find the right balance of detail and abstraction. Make the UI too realistic and it becomes hard to focus on the relevant elements. Realistic UI images also need updating anytime the actual product UI changes. Conversely, make the image too simplistic and it doesn't provide enough detail for people to actually learn how to do things.

Of course, not all Help topics are well suited to an image but the process of trying to create one often triggers ideas on how to simplify the actual UI or concepts within a product. So it's worth the iteration.

But is a visual approach to Help pages able to scale? Assuming it works, can companies invest the time and effort needed to generate all these images and keep them up to date? Perhaps in a time of image generation AI models, it's increasingly possible through automated or supervised pipelines. Time will tell!
