The biggest problem with this is that when she doesn't eat, she gets sick. Out both ends. And there's sometimes blood. This is unpleasant enough for Dan and I, but has to be even worse for her. The problem could usually be solved just by making absolutely sure that she ate her meals, but I recently took her to the vet to see if there was another problem.Come to find out that GSDs often have digestive issues and now we are in the middle of trying to diagnose hers. It could be as simple as a parasite (though none shows up in the tests) or as complicated as IBD. For now, she is taking some antibiotics and just finished her first course of dewormer, so we'll see where things go from there. I'm happy to report that she's been eating well, at least, and hasn't had any issues.Does anyone have any experience with this? I'm told it's very very common in GSDs, especially! That's what some people chanted when the news of Osama Bin Laden's killing broke. But will it have any impact on the President Barack Obama's politics and popularity? Mr Obama has gone out of his way to stress that "get Bin Laden" was his direct instruction and that the arch villain's death is, in part, his victory. White House officials are doing all they can to capitalise on what looks like a mood of nationwide elation. Any president who "got" Bin Laden would benefit. Former President Bill Clinton's efforts were mocked by George W Bush. Then he failed too, losing Bin Laden in the caves along the border land, as US soldiers stood by. But perhaps Mr Obama will benefit more than most. His style of decision making is to take time, to deliberate, to chew over every option. His critics call it dithering. There are now some excellent "tick tocks" as they are called here - blow by blow accounts of the decision making process. But you always have to remember all sources are in the circle, and liable to portray the president positively. It sounds as if Mr Obama gave this decision as much time and thought as all the others but away from the public gaze. Not only did Mr Obama's security advisor John Brennan praise him, but Republicans have even called his decision "gutsy". He did not simply go for bombs or drones but rather a helicopter raid. One insider is quoted as saying that Black Hawk Down was mentioned a few times in the discussions. When that helicopter did go down, Mr Obama surely thought of Jimmy Carter and Iran. So he's a risk taker, too. It also makes him look focused on what is truly in the US's national interest. You can argue Iraq wasn't, Libya wasn't, even Afghanistan no longer is. But getting the head of al-Qaeda clearly was a number one priority in the minds of many Americans, and Mr Obama decided it was his as well. At a reception for Republicans and Democrats last night, he got a standing ovation. So the wind is behind him. Whence will he sail? At a White House dinner for members of Congress, he used Bin Laden's killing as a call for unity. He said: "We were reminded again that there is a pride in what this nation stands for, and what we can achieve, that runs far deeper than party, far deeper than politics." From Bin Laden, he moved effortlessly to domestic public enemy number one, the deficit. "It is my fervent hope that we can harness some of that unity and some of that pride to confront the many challenges that we still face," Mr Obama said. On Thursday, Mr Obama will travel to New York City to remember those who died in Bin Laden's assault on America. I expect more talk of unity but perhaps some big foreign policy themes as well. There are those who think the halo of success makes it easier for the president to confront a military that wants July's Afghanistan wind-down to be small and fairly insignificant. Others, however, think the momentum runs the other way, and that it gives all the more reason to stay and finish the job. So the killing sends waves that will wash against these shores and those of a wider world. Some are saying this moment assures Mr Obama's re-election. It assures no such thing. Apart from the obvious point that there can be many other unexpected events that will have an impact, positive or negative, It just doesn't work like that. However huge this event snow seems, wait a couple of months. In the relentless frenzy of the 24-hour media cycle, it will probably be half forgotten by the the time of the election. This far out, only events that mean change to people's lives on a day-to-day basis have that sort of game changing impact. But image is important. The president has burnished his in the eyes of many Americans and looks like a resolute commander-in-chief. He knows it, and intends to milk the moment for all it is worth. Full Article

The journal Science published a survey of the field and the Cambridge, Massachusetts start-up Holobiome that hopes to use insight into this "psychobiome" to develop treatments for depression, insomnia, and other conditions with a neurological side to them. From Science: For example, many people with irritable bowel syndrome are also depressed, people on the autism spectrum tend to have digestive problems, and people with Parkinson’s are prone to constipation. Researchers have also noticed an increase in depression in people taking antibiotics—but not antiviral or antifungal medications that leave gut bacteria unharmed. Last year, Jeroen Raes, a microbiologist at the Catholic University of Leuven, and colleagues analyzed the health records of two groups—one Belgian, one Dutch—of more then 1000 people participating in surveys of their types of gut bacteria. People with depression had deficits of the same two bacterial species, the authors reported in April 2019 in Nature Microbiology. Researchers see ways in which gut microbes could influence the brain. Some may secrete messenger molecules that travel though the blood to the brain. Other bacteria may stimulate the vagus nerve, which runs from the base of the brain to the organs in the abdomen. Bacterial molecules might relay signals to the vagus through recently discovered “neuropod” cells that sit in the lining of the gut, sensing its biochemical milieu, including microbial compounds. Here, we quantified liver function, bile acid pools, and gut microbial and metabolite profiles in premature, parenterally fed pigs given a soy-oil lipid emulsion, Intralipid (IL); a multi component lipid emulsion, SMOFlipid (SMOF); a novel emulsion with a modified fatty-acid composition (EXP); or a control enteral diet (ENT) for 22 days. We assayed serum cholestasis markers; measured total bile acid levels in plasma, liver, and gut contents; and analyzed colonic bacterial 16S rRNA gene sequences and metabolomic profiles. Serum cholestasis markers (i.e. bilirubin, bile acids, and g-glutamyl transferase) were highest in IL-fed pigs and normalized in those given SMOF, EXP, or ENT. Gut bile acid pools were lowest in the IL treatment and were increased in the SMOF and EXP treatments and comparable to ENT. Multiple bile acids, especially their conjugated forms, were higher in the colon contents of SMOF and EXP than in IL pigs. Colonic microbial communities of SMOF and EXP pigs had lower relative abundance of several Gram-positive anaerobes, including Clostridrium XIVa, and higher abundance of Enterobacteriaceae than those of IL and ENT pigs. Differences in lipid and microbial-derived compounds were also observed in colon metabolite profiles. These results indicate that multi-component lipid emulsions prevent cholestasis and restore enterohepatic bile flow in association with gut microbial and metabolomic changes. We conclude that sustained bile flow induced by multi-component lipid emulsions likely exerts a dominant effect in reducing bile acid–sensitive, Gram-positive bacteria. Full Article

Synthesized by both host and microbial pathways, BAs are increasingly understood as participating in the regulation of numerous pathways relevant to metabolic diseases, including lipid and glucose metabolism, energy expenditure, and inflammation. Quantitative analyses of BAs in biological matrices can be problematic due to their unusual and diverse physicochemical properties, making optimization of a method that shows good accuracy, precision, efficiency of extraction, and minimized matrix effects across structurally distinct human and murine BAs challenging. Herein we develop and clinically validate a stable-isotope-dilution LC/MS/MS method for the quantitative analysis of numerous primary and secondary BAs in both human and mouse biological matrices. We also utilize this tool to investigate gut microbiota participation in the generation of structurally specific BAs in both humans and mice. We examine circulating levels of specific BAs and in a clinical case-control study of age- and gender-matched type 2 diabetes mellitus (T2DM) versus nondiabetics. BAs whose circulating levels are associated with T2DM include numerous 12α-hydroxyl BAs (taurocholic acid, taurodeoxycholic acid, glycodeoxycholic acid, deoxycholic acid, and 3-ketodeoxycholic acid), while taurohyodeoxycholic acid was negatively associated with diabetes. The LC/MS/MS-based platform described should serve as a robust, high-throughput investigative tool for studying the potential involvement of structurally specific BAs and the gut microbiome on both physiological and disease processes. However, the effects of GPR120 activation on dietary fat intake or obesity are not clearly understood. We investigated to determine whether the binding of TUG891, a novel GPR120 agonist, to lingual GPR120 modulates fat preference in mice. We explored the effects of TUG891 on obesity-related hormones and conducted behavioral choice tests on mice to better understand the physiologic relevance of the action of TUG891. In cultured mouse and human taste bud cells (TBCs), TUG891 induced a rapid increase in Ca2+ by acting on GPR120. A long-chain dietary fatty acid, linoleic acid (LA), also recruited Ca2+ via GPR120 in human and mouse TBCs. Both TUG891 and LA induced ERK1/2 phosphorylation and enhanced in vitro release of glucagon-like peptide-1 from cultured human and mouse TBCs. In situ application of TUG891 onto the tongue of anesthetized mice triggered the secretion of pancreatobiliary juice, probably via the tongue-brain-gut axis. Furthermore, lingual application of TUG891 altered circulating concentrations of cholecystokinin and adipokines, associated with decreased circulating LDL, in conscious mice. In behavioral tests, mice exhibited a spontaneous preference for solutions containing either TUG891 or LA instead of a control. However, addition of TUG891 to a solution containing LA significantly curtailed fatty acid preference. Our study demonstrates that TUG891 binds to lingual GPR120 receptors, activates the tongue-brain-gut axis, and modulates fat preference. These findings may support the development of new fat taste analogs that can change the approach to obesity prevention and treatment. Full Article
gut Longitudinal Analysis of Serum Cytokine Levels and Gut Microbial Abundance Links IL-17/IL-22 with Clostridia and Insulin Sensitivity in Humans By diabetes.diabetesjournals.org Published On :: 2020-05-04T10:07:04-07:00 Recent studies using mouse models suggest that interaction between the gut microbiome and IL-17/IL-22 producing cells plays a role in the development of metabolic diseases. We investigated this relationship in humans using data from the prediabetes study of the Integrated Human Microbiome Project (iHMP). Specifically, we addressed the hypothesis that early in the onset of metabolic diseases there is a decline in serum levels of IL-17/IL-22, with concomitant changes in the gut microbiome. Clustering iHMP study participants on the basis of longitudinal IL-17/IL-22 profiles identified discrete groups. Individuals distinguished by low levels of IL-17/IL-22 were linked to established markers of metabolic disease, including insulin sensitivity. These individuals also displayed gut microbiome dysbiosis, characterized by decreased diversity, and IL-17/IL-22-related declines in the phylum Firmicutes, class Clostridia, and order Clostridiales. This ancillary analysis of the iHMP data therefore supports a link between the gut microbiome, IL-17/IL-22 and the onset of metabolic diseases. This raises the possibility for novel, microbiome-related therapeutic targets that may effectively alleviate metabolic diseases in humans as they do in animal models. Full Article
