Now, 10 years on, they are revisiting the support available for family carers to see whether recent policy initiatives and investment have delivered the much-needed change. A total of 264 family carers responded to their survey on short breaks provision and experiences of caring. They also sent Freedom of Information requests to all 152 local authorities in England that provide social care services. This report looks at short breaks provision in a climate of cuts to central and local government budgets. It examines the extent to which these cuts have impacted on the lives of people with a learning disability and their family carers. It also looks at the state of affairs for family carers of children and young people across the full spectrum of learning disability; from people with mild and moderate learning disabilities, to people with severe and profound disabilities. repo Hexie 2 Report - 733 Hexie Flowers Finished! By beadlust.blogspot.com Published On :: Sat, 14 May 2016 23:04:00 +0000 I started cutting scraps of fabric to make hexie flowers on September 1, 2015. After 9 months of labor, I have just finished stitching the 733rd flower! I'll do the math for you... That's 5,131 hexies. They are small ones, 3/4 inch per side. The finished flowers measure just under 4" in diameter. To give you an idea, this is what 44 hexie flowers look like. In case you don't know, for each hexie, the fabric is cut, and then basted over a paper, hexagon-shaped form. The hexies are then hand-stitched together to make the flowers. It takes about 1 hour to make one hexie flower, start-to-finish; thus 44 hours to make the batch above. Click the photo to see better detail of this process, which is called English paper piecing. And here are all 733 hexie flowers! Each flower is a different fabric. Although a few of the flower petal fabrics were also used for flower centers, I'm certain that there are over 1,000 different fabrics used in these flowers. What's next? Well, I'm going to build a design wall using sound-proofing foam-board covered with a king-sized flannel sheet. I haven't quite figured out how to make it yet. When the design wall is ready, I'll get out my hexie flowers and start to "paint" with them. Who knows what will emerge? Not me. I only have a vague idea that I might want to try "painting" an abstract view of our island shoreline. When the "painting" process is finished, I'll stitch the flowers together in small groups, and then stitch the groups together. The result, hopefully in my lifetime, will be a queen-sized, non-traditional style, hexie quilt! TNF is synthesized as a transmembrane protein, which is further matured via proteolytic cleavage by metalloproteases such as ADAM17, a process known as shedding. At present, TNF is mainly detected by measuring the precursor or the mature cytokine of bulk cell populations by techniques such as ELISA or immunoblotting. However, these methods do not provide information on the exact timing and extent of TNF cleavage at single-cell resolution and they do not allow the live visualization of shedding events. Here, we generated C-tag TNF as a genetically encoded reporter to study TNF shedding at the single-cell level. The functionality of the C-tag TNF reporter is based on the exposure of a cryptic epitope on the C terminus of the transmembrane portion of pro-TNF on cleavage. In both denatured and nondenatured samples, this epitope can be detected by a nanobody in a highly sensitive and specific manner only upon TNF shedding. As such, C-tag TNF can successfully be used for the detection of TNF cleavage in flow cytometry and live-cell imaging applications. We furthermore demonstrate its applicability in a forward genetic screen geared toward the identification of genetic regulators of TNF maturation. In summary, the C-tag TNF reporter can be employed to gain novel insights into the complex regulation of ADAM-dependent TNF shedding. Full Article

The elevation of cytokines in blood is crucial to induce cytokine storm and immunosuppression in the transition of severity in COVID-19 patients. However, the comprehensive changes of serum proteins in COVID-19 patients throughout the SARS-CoV-2 infection is unknown. In this work, we developed a high-density antibody microarray and performed an in-depth proteomics analysis of serum samples collected from early COVID-19 (n = 15) and influenza (n = 13) patients. We identified a large set of differentially expressed proteins (n = 132) that participate in a landscape of inflammation and immune signaling related to the SARS-CoV-2 infection. Furthermore, the significant correlations of neutrophil and lymphocyte with the CCL2 and CXCL10 mediated cytokine signaling pathways was identified. These information are valuable for the understanding of COVID-19 pathogenesis, identification of biomarkers and development of the optimal anti-inflammation therapy. Full Article

This means Chatham House is needed more than ever. Our world-leading convening and cutting-edge research on the major challenges facing the world, from building more sustainable economic growth and tackling climate change to easing geopolitical tensions, has continued despite the challenges of lockdown. For that I pay tribute to the resilience and ingenuity of the staff who have found innovative solutions to the obstacles presented by the pandemic, all while working largely from home. This year I am also delighted to note the exceptional gift of £10 million from the MAVA Foundation to enable us to launch the Sustainability Accelerator. This initiative builds on the Hoffmann Centre’s last five years of innovative convening and activity. And it puts sustainability at the core of the institute’s work in this critical year for addressing climate change and biodiversity. So, as I prepare to step down from my three years as chair of the institute, I would like to thank my fellow Council members and Robin Niblett and his team for their hard work and ambition to deliver on Chatham House’s mission. I have also been especially pleased to see how we are engaging younger, more diverse audiences through the next generation initiatives, including our Panel of Young Advisers, the Common Futures Conversations project, the QEII Academy Ambassadors, our Internship Programme and the Chatham House-SNF CoLab. I am particularly pleased about the Chatham House Summer School, where 16–18-year-olds can now engage with experts on international affairs and get an insight into careers within the charity and not-for-profit sector.
