The Minor Phytocannabinoid Delta-8-Tetrahydrocannabinol Attenuates Collagen-Induced Arthritic Inflammation and Pain-Depressed Behaviors [Special Section: Cannabinoid Signaling in Human Health and Disease]

Patients with arthritis report using cannabis for pain management, and the major cannabinoid delta-9-tetrahydrocannabinol (9-THC) has anti-inflammatory properties, yet the effects of minor cannabinoids on arthritis are largely unknown. The goal of the present study was to determine the antiarthritic potential of the minor cannabinoid delta-8-tetrahydrocannabinol (8-THC) using the collagen-induced arthritis (CIA) mouse model. Adult male DBA/1J mice were immunized and boosted 21 days later with an emulsion of collagen and complete Freund’s adjuvant. Beginning on the day of the booster, mice were administered twice-daily injections of 8-THC (3 or 30 mg/kg), the steroid dexamethasone (2 mg/kg), or vehicle for two weeks. Dorsal-ventral paw thickness and qualitative measures of arthritis were recorded daily, and latency to fall from an inverted grid was measured on alternating days, to determine arthritis severity and functional impairment. On the final day of testing, spontaneous wire-climbing behavior and temperature preference in a thermal gradient ring were measured to assess CIA-depressed behavior. The 8-THC treatment (30 mg/kg) reduced paw swelling and qualitative signs of arthritis. 8-THC also blocked CIA-depressed climbing and CIA-induced preference for a heated floor without producing locomotor effects but did not affect latency to fall from a wire grid. In alignment with the morphologic and behavioral assessments in vivo, histology revealed that 8-THC reduced synovial inflammation, proteoglycan loss and cartilage and bone erosion in the foot joints in a dose-dependent manner. Together, these findings suggest that 8-THC not only blocked morphologic changes but also prevented functional loss caused by collagen-induced arthritis.

SIGNIFICANCE STATEMENT

Despite increasing use of cannabis products, the potential effects of minor cannabinoids are largely unknown. Here, the minor cannabinoid delta-8-tetrahydrocannabinol blocked the development of experimentally induced arthritis by preventing both pathophysiological as well as functional effects of the disease model. These data support the development of novel cannabinoid treatments for inflammatory arthritis.

In accordance with one or more embodiments of the present disclosure, methods and apparatus are provided for flexible and user-friendly position editing of loaded media in a multi-media presentation. In one embodiment, a method for editing the position of loaded media comprises loading a page of a collage document to a client device, the page having a plurality of layers with each layer being associated with a media object, and creating a list of layers of the loaded page with each layer indexed by at least a position in the collage document. The method further includes selecting a first media object, selecting a position editing tool to group the first media object and at least one other media object adjacent to the first media object; and moving the grouped first media object and the at least one other media object to a different position in the collage document. A client device for position editing loaded media is also disclosed.

Preservation of the biological activity of undenatured type II collagen

The instant invention is the storage of Type II collagen containing tissue in carbonated water. Such Type II collagen is useful for alleviating the symptoms of arthritis in mammals as well as the treatment of arthritis in mammals. The instant invention is also a method for the preparation of a nutritional supplement that includes the steps of: (a) separating water-insoluble undenatured Type II collagen containing animal tissue from animal tissue not containing Type II collagen; (b) subdividing and sterilizing said tissue under conditions which do not change the original structure of the Type II collagen to produce a subdivided and sterilized product; (c) packaging the subdivided and sterilized product in carbonated water.

Collagen fiber reconstituted rawhide and process for making

This invention describes a collagen fiber reconstituted rawhide useful in the production of pet chews, and a method for making the same. Beginning materials include animal skins with hair, depilated limed splits and leftovers from the manufacture of rawhide pet chews. The materials are pretreated, ground, subjected to acid treatment to swell and separate the collagen fibers that comprise the animal hide, further ground into pulp, vacuum filtered, mixed with a dehydration agent that chemically shrinks the expanded fibers, drained, formed and dried into sheet and finally exposed to a cross-linking agent to increase the adhesive strength of the end product. Edible materials, including meats, may be added to the solution prior to draining, to enhance the nutritional value and palatability of the final product, and the reconstituted rawhide sheets may be bleached for appearance.

Washer Collage

Hi! I've been stuck inside for weeks, probably just like you. I go out running every day, dodging people, but otherwise it's lockdown-mode. Our washing machine promptly broke, so I had to replace that thing. It became a project, because (aside from the difficult but mostly uninteresting process of getting it onto the second floor) one of the things that contributed to the last one's failure was its not-very-stable footing, and I wanted to do this one well. The thing resides inside a nice (but probably unnecessary) tile basin, which poses a few problems: It would make it impossible to get to the bottom doors on the machine, and it makes it impossible to adjust the feet in situ for leveling purposes, and the basin is not at all flat. The weirdly-shaped surface meant that my CAD prospectus was not very useful, which is annoying because I like to measure like 200 times in CAD and then cut once.

The other problem is that I didn't have the right wood for this, and although Home Depot claimed to be able to do a same-day shipment, they gave me the runaround for over a week (I still don't have it). It's understandable, but our piles of laundry were getting a bit dire, so I just had to make do with what I had. In figure 1(a) I sawed through these 6x6 timbers with a 3.5" saw, which took like an hour. Then I used the also-too-small table saw to mill that into the smaller size I actually wanted (figure 1(b)). Then, I painstakingly test fit the logs in the basin, and sawed/planed/chiseled/sanded them until they were sitting stably on that curved surface without wobble. This was a real pain. The best advice I have for doing this was to get the tile sopping wet, then place the wood there for a moment, and then see where the high spots are based on where the wood is wet. (It would work better with some dye or something, but I didn't want to ruin the tile, ugh.) At that point we have some logs that were nice and sturdy, but not necessarily level gravity-wise. My solution here was router-out cups for each of the washer's feet, which I could set the depth of so that the washer would be level without any adjustment. (This also has the nice advantage that the washer can't jump around more than a few millimeters!) This was accomplished by using a laser level for an accurate level, and then putting some objects of fixed height (here the feet from the old washer, which will be disassembled for its more exciting pieces) into each cup, and iteratively routing the depth until they all touch the laser line exactly (Figure 1(c)). All that work paid off, though, because when we finally dropped the washer into place, it was as level as a spirit level can possibly indicate (Figure 1(d)). No pictures of the install here because this is like in my bathroom and that seems weirdly intimate to put on the internet for some reason.

SIGBOVIK is tomorrow, but this year there is no in-person event due to the shelter-in-place order! The proceedings is shaping up nicely though, and there is some "podcast" expected. I have a few silly papers in there, but I'll save those for tomorrow. No talks from me this year; the whole situation in the world has been sort of draining my creative energy, but hopefully I will start feeling good again soon.

Cinnamaldehyde Inhibits Inflammation of Human Synoviocyte Cells Through Regulation of Jak/Stat Pathway and Ameliorates Collagen-Induced Arthritis in Rats [Inflammation, Immunopharmacology, and Asthma]

Cinnamaldehyde (Cin), a bioactive cinnamon essential oil from traditional Chinese medicine herb Cinnamomum cassia, has been reported to have multipharmacological activities including anti-inflammation. However, its role and molecular mechanism of anti-inflammatory activity in musculoskeletal tissues remains unclear. Here, we first investigated the effects and molecular mechanisms of Cin in human synoviocyte cells. Then in vivo therapeutic effect of Cin on collagen-induced arthritis (CIA) also studied. Cell Counting Kit ‎CCK-8 assay was performed to evaluate the cell cytotoxicity. Proinflammatory cytokine expression was evaluated using quantitative polymerase chain reaction and ELISA. Protein expression was measured by western blotting. The in vivo effect of Cin (75 mg/kg per day) was evaluated in rats with CIA by gavage administration. Disease progression was assessed by clinical scoring, radiographic, and histologic examinations. Cin significantly inhibited interleukin (IL)-1β–induced IL-6, IL-8, and tumor necrosis factor-α release from human synoviocyte cells. The molecular analysis revealed that Cin impaired IL-6–induced activation of Janus kinase 2 (JAK2), signal transducer and activator of transcription 1 (STAT1), and STAT3 signaling pathway by inhibiting the phosphorylation of JAK2, STAT1, and STAT3, without affecting NF-B pathway. Cin reduced collagen-induced swollen paw volume of arthritic rats. The anti-inflammation effects of Cin were associated with decreased severity of arthritis, joint swelling, and reduced bone erosion and destruction. Furthermore, serum IL-6 level was decreased when Cin administered therapeutically to CIA rats. Cin suppresses IL-1β–induced inflammation in synoviocytes through the JAK/STAT pathway and alleviated collagen-induced arthritis in rats. These data indicated that Cin might be a potential traditional Chinese medicine–derived, disease-modifying, antirheumatic herbal drug.

SIGNIFICANCE STATEMENT

In this study, we found that cinnamaldehyde (Cin) suppressed proinflammatory cytokines secretion in rheumatology arthritis synoviocyte cells by Janus kinase/signal transducer and activator of transcription pathway. The in vivo results showed that Cin ameliorated collagen-induced arthritis in rats. These findings indicate that Cin is a potential traditional Chinese medicine–derived, disease-modifying, antirheumatic herbal drug.
