A molybdenum tris­(di­thiol­ene) complex coordinates to three bound cobalt centers in three different ways

The synthesis and structural characterization of the mol­ecular compound (μ3-benzene-1,2-di­thiol­ato)hexa­carbonyl­bis­(μ3-1,1,1,4,4,4-hexafluorobut-2-ene-2,3-dithiolato)tricobaltmolybdenum, [Co3Mo(C4F6S2)2(C6H4S2)(CO)6] or Mo(tfd)2(bdt)(Co(CO)2)3 (tfd is 1,1,1,4,4,4-hexafluorobut-2-ene-2,3-dithiolate and bdt is benzene-1,2-di­thiol­ate), are reported. The structure of the mol­ecule contains the molybdenum tris­(di­thiol­ene) complex Mo(tfd)2(bdt) coordinated as a multidentate ligand to three cobalt dicarbonyl units. Each of the three cobalt centers is relatively close to molybdenum, with Co⋯Mo distances of 2.7224 (7), 2.8058 (7), and 2.6320 (6) Å. Additionally, each of the cobalt centers is bound via main-group donor atoms, but each one in a different way: the first cobalt atom is coordinated by two sulfur atoms from different di­thiol­enes (bdt and tfd). The second cobalt atom is coordinated by one sulfur from one tfd and two olefinic carbons from another tfd. The third cobalt is coordinated by one sulfur from bdt and two sulfurs from tfd. This is, to the best of our knowledge, the first structurally characterized example of a molybdenum (tris­)di­thiol­ene complex that coordinates to cobalt. The F atoms of two of the –CF3 groups were refined as disordered over two sets of sites with ratios of refined occupancies of 0.703 (7):0.297 (7) and 0.72 (2):0.28 (2).

Structural insights into conformational switching in latency-associated peptide between transforming growth factor β-1 bound and unbound states

Transforming growth factor β-1 (TGFβ-1) is a secreted signalling protein that directs many cellular processes and is an attractive target for the treatment of several diseases. The primary endogenous activity regulatory mechanism for TGFβ-1 is sequestration by its pro-peptide, latency-associated peptide (LAP), which sterically prohibits receptor binding by caging TGFβ-1. As such, recombinant LAP is promising as a protein-based therapeutic for modulating TGFβ-1 activity; however, the mechanism of binding is incompletely understood. Comparison of the crystal structure of unbound LAP (solved here to 3.5 Å resolution) with that of the bound complex shows that LAP is in a more open and extended conformation when unbound to TGFβ-1. Analysis suggests a mechanism of binding TGFβ-1 through a large-scale conformational change that includes contraction of the inter-monomer interface and caging by the `straight-jacket' domain that may occur in partnership through a loop-to-helix transition in the core jelly-roll fold. This conformational change does not appear to include a repositioning of the integrin-binding motif as previously proposed. X-ray scattering-based modelling supports this mechanism and reveals possible orientations and ensembles in solution. Although native LAP is heavily glycosylated, solution scattering experiments show that the overall folding and flexibility of unbound LAP are not influenced by glycan modification. The combination of crystallography, solution scattering and biochemical experiments reported here provide insight into the mechanism of LAP sequestration of TGFβ-1 that is of fundamental importance for therapeutic development.

Structures of substrate- and product-bound forms of a multi-domain copper nitrite reductase shed light on the role of domain tethering in protein complexes

Copper-containing nitrite reductases (CuNiRs) are found in all three kingdoms of life and play a major role in the denitrification branch of the global nitro­gen cycle where nitrate is used in place of di­oxy­gen as an electron acceptor in respiratory energy metabolism. Several C- and N-terminal redox domain tethered CuNiRs have been identified and structurally characterized during the last decade. Our understanding of the role of tethered domains in these new classes of three-domain CuNiRs, where an extra cytochrome or cupredoxin domain is tethered to the catalytic two-domain CuNiRs, has remained limited. This is further compounded by a complete lack of substrate-bound structures for these tethered CuNiRs. There is still no substrate-bound structure for any of the as-isolated wild-type tethered enzymes. Here, structures of nitrite and product-bound states from a nitrite-soaked crystal of the N-terminal cupredoxin-tethered enzyme from the Hyphomicrobium denitrificans strain 1NES1 (Hd1NES1NiR) are provided. These, together with the as-isolated structure of the same species, provide clear evidence for the role of the N-terminal peptide bearing the conserved His27 in water-mediated anchoring of the substrate at the catalytic T2Cu site. Our data indicate a more complex role of tethering than the intuitive advantage for a partner-protein electron-transfer complex by narrowing the conformational search in such a combined system.

High-resolution crystal structures of Escherichia coli FtsZ bound to GDP and GTP

Bacterial cytokinesis is mediated by the Z-ring, which is formed by the prokaryotic tubulin homolog FtsZ. Recent data indicate that the Z-ring is composed of small patches of FtsZ protofilaments that travel around the bacterial cell by treadmilling. Treadmilling involves a switch from a relaxed (R) state, favored for monomers, to a tense (T) conformation, which is favored upon association into filaments. The R conformation has been observed in numerous monomeric FtsZ crystal structures and the T conformation in Staphylococcus aureus FtsZ crystallized as assembled filaments. However, while Escherichia coli has served as a main model system for the study of the Z-ring and the associated divisome, a structure has not yet been reported for E. coli FtsZ. To address this gap, structures were determined of the E. coli FtsZ mutant FtsZ(L178E) with GDP and GTP bound to 1.35 and 1.40 Å resolution, respectively. The E. coli FtsZ(L178E) structures both crystallized as straight filaments with subunits in the R conformation. These high-resolution structures can be employed to facilitate experimental cell-division studies and their interpretation in E. coli.

SALEM – The Virginia Department of Transportation encourages race fans driving to and from Martinsville Speedway for the race on Sunday, October 28, to follow recommended traffic patterns as posted on signs and message boards. Drivers travelling from Roanoke to the Martinsville Speedway for the race events this weekend should plan for possible delays due to a left lane closure on Route 220 southbound approaching the Boones Mill area. VDOT will place many signs and changeable message boards throughout the region to assist travelers. Vehicular and pedestrian traffic will be heavy. Traffic is being routed to the speedway in certain patterns to minimize overall congestion as much as possible. Watch for the speed limit to reduce gradually from 65 mph to 35 mph on the Route 220/Route 58 Bypass as you approach the speedway. From Roanoke and areas north of Martinsville: Race fans traveling south on Route 220 are encouraged to allow for extra travel time. A left lane closure is in place for a pipe failure in the median about eight miles south of Roanoke and 1.7 miles north of Boones Mill. Drivers can take the exit onto Route 58 to Martinsville to reach the track or they can continue on the Route 220/Route 58 Bypass past the speedway interchange and turn right onto the access road that leads into the speedway. The entrance is located one mile east of the Route 220 interchange and may provide easier access. Signs are in place designating the entrance to the speedway.  From Danville and areas east of Martinsville: Motorists on the Route 58 Bypass should follow signs directing them to use the track’s back gate. This entrance is located off the bypass, east of the track. From Greensboro and areas south of Martinsville: Route 220 northbound traffic will split into three lanes shortly before reaching the speedway.  Motorists should watch for electronic message boards and obey police directing traffic. Through traffic should use the left lane. From Stuart and areas west of Martinsville: Motorists should stay on Route 58 east and follow signs and message boards to the track.  ADA Parking: Race fans needing ADA Parking are encouraged to enter the Speedway via Old Sand Roa Those attending the race should closely follow directional information and plan to arrive early and expect heavy traffic and delays.
