oda Ex-PM Abbasi to brief sugar probe body today By feedproxy.google.com Published On :: Sat, 09 May 2020 04:38:30 +0500 ISLAMABAD: Former prime minister and senior leader of the Pakistan Muslim League-Nawaz (PML-N) Shahid Khaqan Abbasi will appear on Saturday (today) before the commission of inquiry investigating the recent shortage and price hike of sugar in the country. Mr Abbasi told Dawn on Friday he would inform the commission how sugar scandals develop, in the light of his experience as a former chief executive of the country. He said he had written a letter to the commission, offering his services to it by sharing his experiences in probing scams. “I wrote the letter to the commission, as under the Commission of Inquiry Act any citizen can assist such commissions in investigating such scams,” he said. Upon receiving the letter, Mr Abbasi said, the commission asked him to provide a written statement, or any other document, that would help in investigating the scandal. “I told the commission that I will not give any written statement or document, but will assist it on the basis of my experience, I being a former member of the Economic Coordination Committee (of the cabinet) and former head of the federal cabinet,” he said. Mr Abbasi, who served as the prime minister in the last PML-N government, said he would explain to the commission how the sugar crisis occurred. When asked to share some of the reasons behind the crisis, the former premier said allowing export of the commodity was one of the main reasons. “The commission has to ascertain who gave the approval for export of sugar when its local demand had not been taken care of.” He said he would request the commission to keep his meeting with its members open to all. Another PML-N leader and former defence minister Khurram Dastagir Khan would be accompanying Mr Abbasi to the meeting. The commission is investigating the recent shortage of sugar and wheat in the country, in which some key members and allies of the ruling Pakistan Tehreek-i-Insaf were allegedly involved. Here, we identified a novel cis-acting sequence (5'-GUA AAG CUA CGG GAC GGU-3'), intronic splicing enhancer 3 (ISE3), which lies 72 nucleotides upstream of the second splice acceptor (A2-2) site of the second intron that defines the exon of the mRNA encoding the 11-kDa viral nonstructural protein. RNA binding motif protein 45 (RBM45) specifically binds to ISE3 with high affinity (equilibrium dissociation constant [KD] = 33 nM) mediated by its RNA recognition domain and 2-homo-oligomer assembly domain (RRM2-HOA). Knockdown of RBM45 expression or ectopic overexpression of RRM2-HOA in human erythroid progenitor cells (EPCs) expanded ex vivo significantly decreased the level of viral mRNA spliced at the A2-2 acceptor but not that of the mRNA spliced at A2-1 that encodes VP2. Moreover, silent mutations of ISE3 in an infectious DNA of B19V significantly reduced 11-kDa expression. Notably, RBM45 also specifically interacts in vitro with ISE2, which shares the octanucleotide (GGGACGGU) with ISE3. Taken together, our results suggest that RBM45, through binding to both ISE2 and ISE3, is an essential host factor for maturation of 11-kDa-encoding mRNA. IMPORTANCE Human parvovirus B19 (B19V) is a human pathogen that causes severe hematological disorders in immunocompromised individuals. B19V infection has a remarkable tropism with respect to human erythroid progenitor cells (EPCs) in human bone marrow and fetal liver. During B19V infection, only one viral precursor mRNA (pre-mRNA) is transcribed by a single promoter of the viral genome and is alternatively spliced and alternatively polyadenylated, a process which plays a key role in expression of viral proteins. Our studies revealed that a cellular RNA binding protein, RBM45, binds to two intron splicing enhancers and is essential for the maturation of the small nonstructural protein 11-kDa-encoding mRNA. The 11-kDa protein plays an important role not only in B19V infection-induced apoptosis but also in viral DNA replication. Thus, the identification of the RBM45 protein and its cognate binding site in B19V pre-mRNA provides a novel target for antiviral development to combat B19V infection-caused severe hematological disorders. Signal complexity could lead to higher cognitive load, impairing performance, or might benefit pollinators by facilitating learning, memory and decision-making. Here, we evaluate learning and memory in foragers of the bumble bee Bombus impatiens trained to simple (unimodal) vs. complex signals (bimodal) under restrained conditions. Use of a proboscis extension response protocol enabled us to control the timing and duration of stimuli presented during absolute and differential learning tasks. Overall, we observed broad variation in the performance under the two conditions, with bees trained to compound bimodal signals learning and remembering as well as, better, or more poorly than bees trained to unimodal signals. Interestingly, the outcome of training was affected by the specific colour-odour combination. Among unimodal stimuli, the performance with odour stimuli was higher than with colour stimuli, suggesting that olfactory signals played a more significant role in the compound bimodal condition. This was supported by the fact that after 24 h, most bimodal-treatment bees responded to odour but not visual stimuli. We did not observe differences in latency of response, suggesting that signal composition affected decision accuracy, not speed. We conclude that restrained bumble bee workers exhibit broad variation of responses to bimodal stimuli and that components of the bimodal signal may not be used equivalently. The analysis of bee performance under restrained conditions enables accurately control the multimodal stimuli provided to individuals and to study the interaction of individual components within a compound.
