High-throughput sequencing (HTS) technologies have been instrumental in investigating biological questions at the bulk and single-cell levels. Comparative analysis of two HTS data sets often relies on testing the statistical significance for the difference of two negative binomial distributions (DOTNB). Although negative binomial distributions are well studied, the theoretical results for DOTNB remain largely unexplored. Here, we derive basic analytical results for DOTNB and examine its asymptotic properties. As a state-of-the-art application of DOTNB, we introduce DEGage, a computational method for detecting differentially expressed genes (DEGs) in scRNA-seq data. DEGage calculates the mean of the sample-wise differences of gene expression levels as the test statistic and determines significant differential expression by computing the P-value with DOTNB. Extensive validation using simulated and real scRNA-seq data sets demonstrates that DEGage outperforms five popular DEG analysis tools: DEGseq2, DEsingle, edgeR, Monocle3, and scDD. DEGage is robust against high dropout levels and exhibits superior sensitivity when applied to balanced and imbalanced data sets, even with small sample sizes. We utilize DEGage to analyze prostate cancer scRNA-seq data sets and identify marker genes for 17 cell types. Furthermore, we apply DEGage to scRNA-seq data sets of mouse neurons with and without fear memory and reveal eight potential memory-related genes overlooked in previous analyses. The theoretical results and supporting software for DOTNB can be widely applied to comparative analyses of dispersed count data in HTS and broad research questions.

This review explores the concept of synergy in pharmacology, emphasizing its importance in optimizing treatment outcomes through the combination of drugs with different mechanisms of action. Synergy, defined as an effect greater than the expected additive effect elicited by individual agents according to specific predictive models, offers a promising approach to enhance therapeutic efficacy while minimizing adverse events. The historical evolution of synergy research, from ancient civilizations to modern pharmacology, highlights the ongoing quest to understand and harness synergistic interactions. Key concepts, such as concentration-response curves, additive effects, and predictive models, are discussed in detail, emphasizing the need for accurate assessment methods throughout translational drug development. Although various mathematical models exist for synergy analysis, selecting the appropriate model and software tools remains a challenge, necessitating careful consideration of experimental design and data interpretation. Furthermore, this review addresses practical considerations in synergy assessment, including preclinical and clinical approaches, mechanism of action, and statistical analysis. Optimizing synergy requires attention to concentration/dose ratios, target site localization, and timing of drug administration, ensuring that the benefits of combination therapy detected bench-side are translatable into clinical practice. Overall, the review advocates for a systematic approach to synergy assessment, incorporating robust statistical analysis, effective and simplified predictive models, and collaborative efforts across pivotal sectors, such as academic institutions, pharmaceutical companies, and regulatory agencies. By overcoming critical challenges and maximizing therapeutic potential, effective synergy assessment in drug development holds promise for advancing patient care.

It will likely release in 2026 and feature Apple Intelligence, according to a reliable analyst.

New iPhone sales will rest entirely in the next quarter this time.

The grant program provides hardware startups with reimbursement for working with local manufacturers.

It's no secret that most technology nowadays isn't exactly built to last forever, with new product models regularly on sale on replace malfunctioning items. However, Apple, which famously announces new products each September, is taking new steps to let people repair their own devices. Apple has launched parts and tools for the iPhone 16, including the Plus, Pro and Pro Max. 

Now, if you have an iPhone 16 model that's acting up, you can go to Apple's self service repair store and — possibly — get what you need to fix it. Choose the product type and model then pick from the iPhone 16's choices of back glass, battery, camera, top speaker, TrueDepth camera, SIM tray and more. Apple's iPhone 16 should be easier to fix than its predecessor, receiving a 7/10 on the iFixit repairability scale, compared to a 4/10 for the iPhone 15. 

You can either get kits or just the necessary part with prices varying based on your phone model. For example, a battery and screw kit for the iPhone 16 Pro Max is $119, but its $99 for the regular iPhone 16. You can get quite a lot of money back on the kit prices if you return the replaced piece afterward. The iPhone 16 Pro Max, for instance, goes down to $61.88 after you send back the old battery. 

Apple created its self-service program in 2022, giving its customers access to actual Apple parts when something in their device failed. However, Apple states that the service "is intended for individuals with the knowledge and experience to repair electronic devices. If you are experienced with the complexities of repairing electronic devices." So, repairs might not be as accessible if you or someone you know isn't savvy in making the swaps. 

Black Friday is right around the corner and the deals are already coming in hot. You can pick up a four-pack of Apple AirTags for just $70, as an early Black Friday promotion. This is a discount of 30 percent and represents a record low price.

Apple AirTags easily made our list of the best Bluetooth trackers, particularly for regular iPhone users. We love the vast finding network, which really helps when you misplace a tag. Just think of all of those AirTags, iPhones and other Apple devices out there helping to create this network.

Apple AirTags also offer the ability to tap into the ultra-wideband (UWB) wireless protocol. This creates a sort of game out of finding lost items, as long as the object is within 25 feet of the phone. The screen will display directional arrows and a distance meter so you can zero in on the lost item without having to ring the AirTag.

There are some caveats. These trackers only really work with iPhones, so Android users should buy something else. Also, the ringer only goes for seven seconds at a time, which isn’t always enough time to track something down. Finally, there’s no attachment point for connecting to a keychain or a related item. However, there are plenty of good AirTag accessories to solve that problem.

If you have an iPhone that can run iOS 18, you can now download and install the public beta of iOS 18.2. One of the most useful new features is the capability to share the location of a lost item connected to Apple's Find My network with a third party. You can already share an item's location with people in your contacts list, but Apple says this could help in instances wherein you have to rely on the help of a stranger.

When you choose "Share Item Location" in the Find My app on iPhone, iPad or Mac, you'll get the option to share a link that shows the location of your missing item. The link's recipient will be able to open it on any device, and they'll also be able to see your Apple account email and/or phone number so that they can get in touch with you. You'll be able to see how many people visited the link, which expires after you're reunited with your lost belonging or after a week if you haven't found it by then.

Along with benevolent strangers, you'll be able to share the locations of misplaced AirTags and Find My accessories with select airlines. Apple has worked with several of them to integrate this feature into their systems. Only a small number of airline staff can access each Share Item Location link, and Apple says they'll first need to authenticate themselves with an Apple Account or partner email address.

More than 15 airlines will start supporting this feature in the coming months, including Aer Lingus, Air Canada, Air New Zealand, Austrian Airlines, British Airways, Brussels Airlines, Delta Air Lines, Eurowings, Iberia, KLM Royal Dutch Airlines, Lufthansa, Qantas, Singapore Airlines, Swiss International Air Lines, Turkish Airlines, United, Virgin Atlantic and Vueling. Apple says that more airlines will get involved over time to help track down mishandled or delayed bags.

Air transport tech company SITA is also incorporating Share Item Location into WorldTracer, its baggage-tracing system. According to Apple, this is used by more than 500 airlines and ground handlers at more than 2,800 airports.

Of course, installing beta software does have its downsides. There's a higher chance of running into bugs, and some apps or features might still not work as Apple had intended. If you don't mind and want to test iOS 18.2, you'll have to enable Beta Updates under Software Updates in your device's Settings menu. Apple is expected to roll out iOS 18.2 proper in December.

Meanwhile, as luck would have it, there's an early Black Friday deal on AirTags at the time of writing. You can scoop up a four-pack for a record low of $70.
