Sheet-on-sheet fixed target data collection devices for serial crystallography at synchrotron and XFEL sources

Fixed targets (`chips') offer efficient, high-throughput microcrystal delivery for serial crystallography at synchrotrons and X-ray free-electron lasers (XFELs). Within this family, sheet-on-sheet (SOS) chips offer noteworthy advantages in cost, adaptability, universality and ease of crystal loading. We describe our latest generation of SOS devices, which are now in active use at both synchrotrons and XFELs.

target

Sheet-on-sheet fixed target data collection devices for serial crystallography at synchrotron and XFEL sources

Serial crystallography (SX) efficiently distributes over many crystals the radiation dose absorbed during diffraction data acquisition, enabling structure determination of samples at ambient temperature. SX relies on the rapid and reliable replacement of X-ray-exposed crystals with fresh crystals at a rate commensurate with the data acquisition rate. `Solid supports', also known as `fixed targets' or `chips', offer one approach. These are microscopically thin solid panes into or onto which crystals are deposited to be individually interrogated by an X-ray beam. Solid supports are generally patterned using photolithography methods to produce a regular array of features that trap single crystals. A simpler and less expensive alternative is to merely sandwich the microcrystals between two unpatterned X-ray-transparent polymer sheets. Known as sheet-on-sheet (SOS) chips, these offer significantly more versatility. SOS chips place no constraint on the size or size distribution of the microcrystals or their growth conditions. Crystals ranging from true nanocrystals up to microcrystals can be investigated, as can crystals grown in media ranging from low viscosity (aqueous solution) up to high viscosity (such as lipidic cubic phase). Here, we describe our two SOS devices. The first is a compact and lightweight version designed specifically for synchrotron use. It incorporates a standard SPINE-type magnetic base for mounting on a conventional macromolecular crystallography goniometer. The second and larger chip is intended for both X-ray free-electron laser and synchrotron use and is fully compatible with the fast-scanning XY-raster stages developed for data collection with patterned chips.

Fragment-based screening targeting an open form of the SARS-CoV-2 main protease binding pocket

To identify starting points for therapeutics targeting SARS-CoV-2, the Paul Scherrer Institute and Idorsia decided to collaboratively perform an X-ray crystallographic fragment screen against its main protease. Fragment-based screening was carried out using crystals with a pronounced open conformation of the substrate-binding pocket. Of 631 soaked fragments, a total of 29 hits bound either in the active site (24 hits), a remote binding pocket (three hits) or at crystal-packing interfaces (two hits). Notably, two fragments with a pose that was sterically incompatible with a more occluded crystal form were identified. Two isatin-based electrophilic fragments bound covalently to the catalytic cysteine residue. The structures also revealed a surprisingly strong influence of the crystal form on the binding pose of three published fragments used as positive controls, with implications for fragment screening by crystallography.

Fixed-target pump–probe SFX: eliminating the scourge of light contamination

X-ray free-electron laser (XFEL) light sources have enabled the rapid growth of time-resolved structural experiments, which provide crucial information on the function of macromolecules and their mechanisms. Here, the aim was to commission the SwissMX fixed-target sample-delivery system at the SwissFEL Cristallina experimental station using the PSI-developed micro-structured polymer (MISP) chip for pump–probe time-resolved experiments. To characterize the system, crystals of the light-sensitive protein light–oxygen–voltage domain 1 (LOV1) from Chlamydomonas reinhardtii were used. Using different experimental settings, the accidental illumination, referred to as light contamination, of crystals mounted in wells adjacent to those illuminated by the pump laser was examined. It was crucial to control the light scattering from and through the solid supports otherwise significant contamination occurred. However, the results here show that the opaque MISP chips are suitable for defined pump–probe studies of a light-sensitive protein. The experiment also probed the sub-millisecond structural dynamics of LOV1 and indicated that at Δt = 10 µs a covalent thioether bond is established between reactive Cys57 and its flavin mononucleotide cofactor. This experiment validates the crystals to be suitable for in-depth follow-up studies of this still poorly understood signal-transduction mechanism. Importantly, the fixed-target delivery system also permitted a tenfold reduction in protein sample consumption compared with the more common high-viscosity extrusion-based delivery system. This development creates the prospect of an increase in XFEL project throughput for the field.

In situ counter-diffusion crystallization and long-term crystal preservation in microfluidic fixed targets for serial crystallography

Compared with batch and vapor diffusion methods, counter diffusion can generate larger and higher-quality protein crystals yielding improved diffraction data and higher-resolution structures. Typically, counter-diffusion experiments are conducted in elongated chambers, such as glass capillaries, and the crystals are either directly measured in the capillary or extracted and mounted at the X-ray beamline. Despite the advantages of counter-diffusion protein crystallization, there are few fixed-target devices that utilize counter diffusion for crystallization. In this article, different designs of user-friendly counter-diffusion chambers are presented which can be used to grow large protein crystals in a 2D polymer microfluidic fixed-target chip. Methods for rapid chip fabrication using commercially available thin-film materials such as Mylar, propylene and Kapton are also detailed. Rules of thumb are provided to tune the nucleation and crystal growth to meet users' needs while minimizing sample consumption. These designs provide a reliable approach to forming large crystals and maintaining their hydration for weeks and even months. This allows ample time to grow, select and preserve the best crystal batches before X-ray beam time. Importantly, the fixed-target microfluidic chip has a low background scatter and can be directly used at beamlines without any crystal handling, enabling crystal quality to be preserved. The approach is demonstrated with serial diffraction of photoactive yellow protein, yielding 1.32 Å resolution at room temperature. Fabrication of this standard microfluidic chip with commercially available thin films greatly simplifies fabrication and provides enhanced stability under vacuum. These advances will further broaden microfluidic fixed-target utilization by crystallographers.

Drill Program Targets High-Grade Gold Veins in British Columbia

Source: Streetwise Reports 11/06/2024

Independence Group NL (IGO:ASX) has begun a comprehensive diamond drill program at its fully-owned 3Ts Project, located in British Columbia.Read more about the 25 planned drill holes aimed at unlocking high-grade intercepts and the promising exploration targets at the 3Ts Project.

Independence Gold Corp. (IGO:TSX.V; IEGCF:OTCMKTS) has begun a comprehensive diamond drill program at its fully-owned 3Ts Project, located in British Columbia. Positioned 16 km from Artemis Gold Inc.'s Blackwater Project, the 3Ts Project covers 8,840 hectares within a prolific epithermal quartz-carbonate vein district on the Nechako Plateau. The program will consist of approximately 25 drill holes, totaling a minimum of 7,500 meters. The targets are the Ted-Mint and Tommy Vein Systems, with a primary emphasis on unexplored depth zones to identify high-grade intercepts for mineral resource expansion.

The 3Ts Project encompasses multiple identified veins, with strike lengths from 50 to over 1,100 meters and true widths of up to 25 meters. Additional exploration will be directed at the Ian, Johnny, and Larry Veins, focusing on mineralization both along strike and at depth. The Ootsa and Balrog targets, identified through geophysical and geological data collected during the summer 2024 exploration program, are also set to undergo further investigation.

President and CEO Randy Turner stated in the press release, "We look forward to building on the success of recent drill programs at 3Ts. With a larger and more extensive drill program planned, including deeper holes to test the major vein systems below the microdiorite sill and further testing of the newly discovered Ootsa and Balrog targets, we anticipate a very busy and exciting year ahead."

Upon hearing this news, Jeff Clark of The Gold Advisor wrote, "And they're off! This is the THIRD drill program this year at 3Ts, an aggressive schedule that, as investors, we're very happy to see."

He noted that these results will help expand the current resource. He continued, "Remember, management just raised a whopping US$6.65 million, more than double the initial goal, due to strong investor interest. They thus have the financial firepower to conduct all this drilling before winter sets in. The stock isn't reacting to the news, but this isn't something that would normally have a big impact on it. It's cooled from its recent spike so offers a very attractive entry point if you don't have the shares you want. This is an overweight position for me, and it's my belief we'll see more spikes just like the one we witnessed. More news and potential catalysts ahead. This is definitely one to own for the gold bull market."

Looking Into Gold

On October 29, Kitco reported that gold prices approached US$2,800. This reflects a substantial 35% increase for the year. According to the report, this growth resulted from multiple factors, including geopolitical conflicts, Federal Reserve interest rate normalization, strong central bank demand, and political uncertainties surrounding the upcoming presidential election. Analysts described these elements as a "perfect storm," which significantly bolstered investor sentiment and reinforced gold's appeal as a hedge against economic instability.

"This is definitely one to own for the gold bull market," Jeff Clark of The Gold Advisor Wrote.

LiveMint, on October 30, noted the strong performance of precious metals, emphasizing that silver had outpaced gold over the past year. Ankit Gohel from LiveMint mentioned, "Gold has delivered a substantial return of over 33.5% since Dhanteras last year," but highlighted that silver had achieved an even more impressive rally of over 40.5%. Despite this, gold continued to attract attention, with Chintan Mehta, CEO of Abans Holdings, emphasizing gold's role as a safe haven during times of uncertainty. He said, "Gold stands out in times of uncertainty . . . It's a complete safe-haven unlike silver, which always has that industrial component attached to it, adding an extra layer of risk."

In a November 4 article, Egon von Greyerz of Matterhorn Asset Management provided a historical perspective on gold's consistent role in preserving wealth. Von Greyerz discussed how gold had risen 78 times since 1971, when the dollar lost its gold backing, emphasizing that "gold held in the investor's name in safe vaults and jurisdictions outside the financial system is the ultimate form of wealth preservation." He argued that gold's ascent had only just begun, driven by the devaluation of fiat currencies and ongoing global debt expansion.

Independence Catalysts

According to the company's September 2024 investor presentation, the 3Ts Project remains a high-priority asset with substantial growth potential. The updated NI 43-101 compliant resource estimate for the Tommy, Ted, and Mint veins, totaling 522,330 ounces of gold and 13.83 million ounces of silver, is expected to expand with new discoveries and continued drilling. Recent metallurgical testing has returned gold recoveries of up to 97.9%, and the strategic location near Artemis Gold's Blackwater Mine adds further credibility to the project's prospects. [OWNERSHIP_CHART-7643]

The fall 2024 drill program, with a budget of CA$4.5 million, will test high-grade zones and underexplored targets, building on over 63,000 meters of historical drilling. Additionally, new targets such as the Balrog and Ootsa anomalies present significant exploration upside, underscoring the project's potential for resource expansion and discovery.

Ownership and Share Structure

According to Refinitiv, about 4.38% of the company is held by insiders and management.

7.97% is with strategic investor Newmont Corp.

The rest is retail.

Its market cap is CA$29.28 million with 167.8 million shares outstanding. It trades in a 52-week range of CA$0.34 and CA$0.12.

In Suing Georgia, Justice Department Says State's New Voting Law Targets Black Voters

Updated June 25, 2021 at 12:54 PM ET

Attorney General Merrick Garland announced Friday that the U.S. Justice Department is suing the state of Georgia over its new voting law, saying that the controversial measure is intended to restrict ballot access to Black voters.

"Our complaint alleges that recent changes to Georgia's election laws were enacted with the purpose of denying or abridging the right of Black Georgians to vote on account of their race or color, in violation of Section 2 of the Voting Rights Act," Garland said at a news conference.

The lawsuit marks the first major action from the Biden administration to combat a series of new restrictive voting measures passed by Republican-led state legislatures. And it came on the eighth anniversary of the U.S. Supreme Court decision to gut another key provision of the landmark Voting Rights Act, Section 5.

Garland noted that Georgia experienced record voter turnout and participation in the 2020 election cycle.

In March, Georgia Gov. Brian Kemp, a Republican, signed Senate Bill 202, a 98-page omnibus measure that makes sweeping changes to the state's absentee voting rules, adds new voter identification mandates and nearly cuts in half the amount of time for voters to request a mail-in ballot. It also expands early voting access for most counties and formally codifies Sunday voting hours as optional.

The legislation outlaws passing out food or drinks to voters within 150 feet of a polling place or too close to voters waiting in line, a provision that Assistant Attorney General Kristen Clarke, who heads the department's Civil Rights Division, highlighted at the press conference.

"Historically, minority voters in Georgia have been disproportionately more likely to wait in long lines to vote in person on Election Day," she said. "Given those long and protracted wait times, civic groups, including churches, have at times provided food and water to voters in line to make their wait more comfortable. As we allege in our complaint, this needless ban was passed with unlawful discriminatory intent."

Clarke also said the Georgia Legislature passed the bill through "a rushed process that departed from normal practice and procedure."

"The version of the bill that passed the state Senate ... was three pages long. Days later, the bill ballooned into over 90 pages in the House. The House held less than two hours of floor debate on the newly inflated SB 202 before Gov. Kemp signed it into law the same day," she said. "These legislative actions occurred at a time when the Black population in Georgia continues to steadily increase, and after a historic election that saw record voter turnout across the state, particularly for absentee voting, which Black voters are now more likely to use than white voters."

Garland said the lawsuit is the first of "many steps" the department is taking to protect the right to vote for all eligible voters. He said the Civil Rights Division will continue to examine voting laws that other states have passed.

"We will not hesitate to act," Garland said.

The Justice Department announced this month it would vigorously defend voting rights. Garland said that the department will double the number of voter enfranchisement lawyers and focus attention on litigation related to voting rights.

In response to the filing, Kemp said the lawsuit is "born out of the lies and misinformation the Biden administration has pushed against Georgia's Election Integrity Act from the start."

"[Biden and his allies] are weaponizing the U.S. Department of Justice to carry out their far-left agenda that undermines election integrity and empowers federal government overreach in our democracy," he said in a statement.

Georgia Secretary of State Brad Raffensperger, another Republican who notably defended the state's administration of the 2020 election, said in a statement he "looks forward to ... beating [the administration] in court."

Garland's announcement comes just days after Senate Republicans united to block Democrats' attempts to pass sweeping voting rights legislation.

Senate Judiciary Chairman Dick Durbin, D-Ill., tweeted his approval of the lawsuit shortly after the announcement Friday.

"If Republicans think the fight for voting rights ended with their filibuster of the For the People Act, they are sorely mistaken," he wrote. "Glad to see the Biden Administration is joining this effort. We must protect our democracy."

The Republican National Committee also linked the failed Senate vote to the Department of Justice's lawsuit.

"After failing to sell the partisan federal election takeover known as H.R. 1 to the American people, Joe Biden is now weaponizing the Justice Department to attack election integrity," RNC Chair Ronna McDaniel said in a statement.
