testing Species Distribution and Comparison between EUCAST and Gradient Concentration Strips Methods for Antifungal Susceptibility Testing of 112 Aspergillus Section Nigri Isolates [Susceptibility] By aac.asm.org Published On :: 2020-04-20T08:36:46-07:00 Aspergillus niger, the third species responsible for invasive aspergillosis has been considered as a homogeneous species until DNA-based identification uncovered many cryptic species. These species have been recently reclassified into the Aspergillus section Nigri. However little is yet known among the section Nigri about the species distribution and the antifungal susceptibility pattern of each cryptic species. A total of 112 clinical isolates collected from 5 teaching hospitals in France and phenotypically identified as A. niger were analyzed. Identification to the species level was carried out by nucleotide sequence analysis. The Minimum Inhibitory Concentrations (MICs) of itraconazole, voriconazole, posaconazole, isavuconazole and amphotericin B were determined by both the EUCAST and gradient concentration strips methods. Aspergillus tubingensis (n=51, 45.5%) and A. welwitschiae (n=50, 44.6%) were the most common species while A. niger accounted for only 6.3% (n=7). The MICs of azoles drugs were higher for A. tubingensis than for A. welwitschiae. The MIC of amphotericin B was 2 mg/L or less for all isolates. Importantly, MICs determined by EUCAST showed no correlation with those determined by gradient concentration strips methods, these latter being lower than the former (Spearman's rank correlation tests ranging - depending on the antifungal agent - from 0.01 to 0.25; p>0.4). In conclusion, A. niger should be considered as a minority species in the section Nigri. The differences in MICs between species for different azoles underline the importance of accurate identification. Significant divergences in the determination of MIC between EUCAST and gradient concentration strips methods require further investigation. Full Article

Guidelines recommend HIV testing with incident STIs. Prevalence and factors associated with HIV testing in acute STIs are unknown in adolescents. Our objective was to determine the prevalence of completed HIV testing among adolescents with incident STIs and identify patient and health care factors associated with HIV testing. METHODS: Retrospective study of STI episodes (gonorrhea, Chlamydia, trichomoniasis, or syphilis) of adolescents between 13 and 24 years old from July 2014 to December 2017 in 2 urban primary care clinics. We performed mixed effects logistic regression modeling to identify patient and health care factors associated with HIV testing within 90 days of STI diagnosis. RESULTS: The 1313 participants contributed 1816 acute STI episodes. Mean age at STI diagnosis was 17.2 years (SD = 1.7), 75% of episodes occurred in females, and 97% occurred in African Americans. Only half (55%) of acute STI episodes had a completed HIV test. In the adjusted model, female sex, previous STIs, uninsured status, and confidential sexual health encounters were associated with decreased odds of HIV testing. Patients enrolled in primary care at the clinics, compared with those receiving sexual health care alone, and those with multipathogen STI diagnoses were more likely to have HIV testing. CONCLUSIONS: HIV testing rates among adolescents with acute STIs are suboptimal. Patient and health care factors were found to be associated with receipt of testing and should be considered in clinical practice. Full Article

Every month, this trio reviews a newly-published paper in academia that pertains to verification and discusses its implications. The purpose of metamorphic testing is to define a verification approach where is there is no “golden reference.” This situation comes up a lot now as designs grow in complexity, and it begs the question: “how does one know the design is verified if there is no standard to compare to?”. Metamorphic testing addresses the problem of not having a “gold standard” to compare to by comparing the results of related tests instead. The paper reviewed by this team used metamorphic testing to study methods of managing JavaScript tags. Paul saw this as a valuable new class of coverage. Metamorphic testing represents a way to create better distribution analyses through understanding the relationships among tests. This can reveal critical-but-complex issues that traditional verification methods may overlook. He saw this as an emerging class of coverage that new verification tools could be built around. Paul asserted that a future metamorphic-testing-based tool’s main contribution to the field of verification would be to better analyze noisy performance results where the noise is multi-modal. It could be useful in detecting race conditions and similar hard-to-debug anomalies. Paul also sees metamorphic testing as ripe for ML techniques. Overall—Paul sees a bright future for metamorphic testing in verification. Jim is reminded of Solido and Spice—these metamorphic testing capabilities are “more than just a feature”—they might be a product. Maybe even a whole new class of verification tools, as Paul said.
