However, the mechanism by which these PFFs induce toxicity is unknown. Iron is implicated in PD pathophysiology, so we investigated whether α-synuclein PFFs induce ferroptosis, an iron-dependent cell death pathway. A range of ferroptosis inhibitors were added to a striatal neuron-derived cell line (STHdhQ7/7 cells), a dopaminergic neuron–derived cell line (SN4741 cells), and WT primary cortical neurons, all of which had been intoxicated with α-synuclein PFFs. Viability was not recovered by these inhibitors except for liproxstatin-1, a best-in-class ferroptosis inhibitor, when used at high doses. High-dose liproxstatin-1 visibly enlarged the area of a cell that contained acidic vesicles and elevated the expression of several proteins associated with the autophagy-lysosomal pathway similarly to the known lysosomal inhibitors, chloroquine and bafilomycin A1. Consistent with high-dose liproxstatin-1 protecting via a lysosomal mechanism, we further de-monstrated that loss of viability induced by α-synuclein PFFs was attenuated by chloroquine and bafilomycin A1 as well as the lysosomal cysteine protease inhibitors, leupeptin, E-64D, and Ca-074-Me, but not other autophagy or lysosomal enzyme inhibitors. We confirmed using immunofluorescence microscopy that heparin prevented uptake of α-synuclein PFFs into cells but that chloroquine did not stop α-synuclein uptake into lysosomes despite impairing lysosomal function and inhibiting α-synuclein toxicity. Together, these data suggested that α-synuclein PFFs are toxic in functional lysosomes in vitro. Therapeutic strategies that prevent α-synuclein fibril uptake into lysosomes may be of benefit in PD. Here, we analyzed the effects of a congenital stationary night blindness type 2 (CSNB2)-causing mutation, I745T (IT), in Cav1.4 channels typical of those in human retina: Cav1.4 splice variants with or without exon 47 (Cav1.4+ex47 and Cav1.4Δex47, respectively), and the auxiliary subunits, β2X13 and α2δ-4. We find that IT caused both Cav1.4 splice variants to activate at significantly more negative voltages and with slower deactivation kinetics than the corresponding WT channels. These effects of the IT mutation, along with unexpected alterations in ion selectivity, were generally larger in channels lacking exon 47. The weaker ion selectivity caused by IT led to hyperpolarizing shifts in the reversal potential and large outward currents that were evident in channels containing the auxiliary subunits β2X13 and α2δ-4 but not in those with β2A and α2δ-1. We conclude that the IT mutation stabilizes channel opening and alters ion selectivity of Cav1.4 in a manner that is strengthened by exclusion of exon 47 and inclusion of β2X13 and α2δ-4. Our results reveal complex actions of IT in modifying the properties of Cav1.4 channels, which may influence the pathological consequences of this mutation in retinal photoreceptors. Full Article

This approach, which is similar to that used in the rotation method of crystallographic data collection at synchrotrons, propagates error estimates from photon-counting statistics to the merged data. Here, it is demonstrated that the application of this approach to SX data provides better SAD phasing ability, enabling the autobuilding of a protein structure that had previously failed to be built. Estimating the error in the merged reflection intensities requires the understanding and propagation of all of the sources of error arising from the measurements. One type of error, which is well understood, is the counting error introduced when the detector counts X-ray photons. Thus, if other types of random errors (such as readout noise) as well as uncertainties in systematic corrections (such as from X-ray attenuation) are completely understood, they can be propagated along with the counting error, as appropriate. In practice, most software packages propagate as much error as they know how to model and then include error-adjustment terms that scale the error estimates until they explain the variance among the measurements. If this is performed carefully, then during SAD phasing likelihood-based approaches can make optimal use of these error estimates, increasing the chance of a successful structure solution. In serial crystallography, SAD phasing has remained challenging, with the few examples of de novo protein structure solution each requiring many thousands of diffraction patterns. Here, the effects of different methods of treating the error estimates are estimated and it is shown that using a parametric approach that includes terms proportional to the known experimental uncertainty, the reflection intensity and the squared reflection intensity to improve the error estimates can allow SAD phasing even from weak zinc anomalous signal. Internet-enabled smartphones and tablets provide access to information and a channel of communication for users. Building wireless networks to support mobile devices requires large capital investments from wireless carriers who must purchase wireless spectrum and infrastructure. To ensure that mobile services are reliable and affordable, national governments must allocate enough wireless spectrum to commercial carriers to satisfy demand. This is the subject of a new paper from Shamika Ravi and Darrell M. West titled “Spectrum Policy in India." A scarce resource Mobile devices typically operate on frequencies from 30 kHz to 300 GHz on the radio spectrum. Unless spectrum is allocated efficiently, the scarcity of available frequencies leads to poor quality and high costs for mobile broadband. The growing demand for mobile service in India currently exceeds the amount of spectrum available to wireless carriers. The scarcity of wireless spectrum limits reliable Internet access for mobile subscribers who have no alternative point of access. According to the Cellular Operators Association of India, nearly 60 percent of Internet users only have access through their mobile phones. Mobile service in India is relatively expensive for many consumers because the Indian military reserves so much spectrum for their own use. Much of this spectrum goes underutilized, even as commercial carriers plead for more spectrum to be released. When the Indian government does release spectrum, it is typically through auctions with high starting bids. Setting high starting bids for blocks of spectrum can lead to high selling prices that force wireless carriers to take out large loans. Higher prices for spectrum raise costs for consumers and reduce private sector investment in wireless infrastructure. Rather than make spectrum artificially scarce, the Indian government should work with wireless carriers to lower the prices for consumers. Investing in India’s future Reliable mobile service has the potential to greatly enhance economic growth in India. Analysis from the Boston Consulting Group found that the India’s mobile sector grew at 12.4 percent annually from 2009-2014; it now accounts for 2.2 percent of India’s gross domestic product. Potential growth comes from filling gaps in educational and health care spending in rural communities. Innovative mobile applications provide a low cost method of sending education and health care resources to underserved rural communities that lack physical infrastructure. In India’s rapidly growing cities, mobile services are seen as a way to improve the quality of government services and promote entrepreneurship. Prime Minister Narendra Modi recently designated 100 “smart cities” that would use technology to overcome the challenges of India’s rapid urbanization. India could free up spectrum by adopting the “NATO Band” of spectrum for military uses and auctioning off the remaining spectrum. The NATO band is used by the militaries of NATO member countries and several of their allies, and it already overlaps with much of the Indian military’s spectrum. Furthermore, the Indian government must lower the minimum bids at spectrum auctions and lower taxes so that wireless carriers have enough profits to build their networks. Mobile technologies are rapidly evolving, and each new generation has greater demands for spectrum. Regulators in India will not only have to maintain affordable prices for the current generation of mobile technology, but also anticipate upgrades that will deliver more data at faster speeds.
