Calcium-ligand variants of the myocilin olfactomedin propeller selected from invertebrate phyla reveal cross-talk with N-terminal blade and surface helices

Olfactomedins are a family of modular proteins found in multicellular organisms that all contain five-bladed β-propeller olfactomedin (OLF) domains. In support of differential functions for the OLF propeller, the available crystal structures reveal that only some OLF domains harbor an internal calcium-binding site with ligands derived from a triad of residues. For the myocilin OLF domain (myoc-OLF), ablation of the ion-binding site (triad Asp, Asn, Asp) by altering the coordinating residues affects the stability and overall structure, in one case leading to misfolding and glaucoma. Bioinformatics analysis reveals a variety of triads with possible ion-binding characteristics lurking in OLF domains in invertebrate chordates such as Arthropoda (Asp–Glu–Ser), Nematoda (Asp–Asp–His) and Echinodermata (Asp–Glu–Lys). To test ion binding and to extend the observed connection between ion binding and distal structural rearrangements, consensus triads from these phyla were installed in the myoc-OLF. All three protein variants exhibit wild-type-like or better stability, but their calcium-binding properties differ, concomitant with new structural deviations from wild-type myoc-OLF. Taken together, the results indicate that calcium binding is not intrinsically destabilizing to myoc-OLF or required to observe a well ordered side helix, and that ion binding is a differential feature that may underlie the largely elusive biological function of OLF propellers.

Vertebrate population losses and declines: Earth’s ongoing mass extinction may be more severe than previously estimated

A new study suggests that Earth’s ongoing mass extinction episode is more severe than generally perceived. Rather than focusing on the complete extinction of entire species, researchers analysed the losses and declines of populations in a sample of 27 600 vertebrate species. Population declines and losses are often a prelude to species extinctions. Researchers also conducted a more in-depth analysis of population losses between 1900–2015 in 177 mammal species. The results reveal that rates of population loss and decline in vertebrates are extremely high, even in common “species of low concern”. The data indicates that, in addition to significant species extinction rates, the Earth is experiencing a huge episode of population decline and loss, which will have a significant effect on ecosystem functioning and services. The researchers warn that the window for effective action is closing rapidly and emphasise the need for an urgent response.

Effects of extreme weather, climate and pesticides on farmland invertebrates

Cereal fields provide a staple food, but are also home to a wide array of invertebrates. This study analysed over 40 years of data to investigate the effects of extreme weather, climate and pesticide use on invertebrates in cereal fields in southern England. As pesticide use had a greater effect on abundance than temperature or rainfall, the authors also recommend reducing pesticide use.

Dietary exposure to neonicotinoid-contaminated plant material poses risk to leaf-shredding invertebrates

Neonicotinoids are pesticides applied to plants to protect them from insects. The use of neonicotinoids may lead to contamination of aquatic environments through, among other routes, the input of contaminated plant material into waterways. While it is well established that direct exposure to contaminated water endangers aquatic invertebrates, scientists have now published findings indicating that dietary exposure through the consumption of contaminated plant material puts leaf-shredding species at increased risk. The researchers recommend that policymakers registering systemic insecticides (those whose active ingredients are transported throughout the plant tissues) consider dietary exposure, and its potential implications for ecosystem integrity, in addition to other exposure pathways.

estaba localizado cerca de la latitud 30.3 norte, longitud 75.1 oeste.
Humberto se mueve hacia el este-noreste a cerca de 8 mph (13 km/h).

DISCUSION Y PRONOSTICO
-----------------------
A las 1000 PM CDT (0300 UTC), el centro de Pos-Tropical Ciclon Olga
estaba localizada en la latitud 27.8 norte, longitud 92.2 oeste. La
pos-tropical ciclon esta moviendose hacia el noreste a cerca de 17
mph (28 km/h) Se pronostica que Olga se mueva rapidamente hacia el
norte a norte-noreste el sabado y despues girarse hacia el noreste
tarde el sabado o doming. En la trayectoria pronosticada, el centro
del ciclon pos-tropical debe moverse a traves del Mississippi Valley
manana y hacis los Great Lakes mas tarde el fin de semana.

Los vientos maximos sostenidos estan cerca de 50 mph (85 km/h) con
rafagas mas fuertes. Se espera que Olga se debilite despues que el
sistema se mueva sobre tierra el sabado por la manana.

Vientos con fuerza de tormenta tropical se extienden hasta 140
millas (220 km) del centro.

La presion minima central estimada basado sobre los datos anteriores
reportado por el Avion Cazahuracan y observaciones de la superficie
sobre el norte del Golfo de Mexico es de 999 mb (29.50 pulgadas).


PELIGROS AFECTANDO TIERRA

VIENTO: Vientos con fuerza de galerna asociado con Olga y sus remenantes
deben extenderse sobre sectores de la costa norte del Golfo esta noche y
sabado.

LLUVIA: El ciclon pos-tropical, y lluvia frente al sistema a lo
largo y al norte de la zona frontal a traves de la costa del Golfo
Central deben producir acumulaciones totales de lluvia de 3 a 6
pulgadas con totales maximos de 8 pulgadas a traves de la costa del
Golfo Central hasta sectores del Lower Mississippi Valley y el oeste
del Tennessee Valley hasta el sabado en la manana. Estas lluvias
pudieran producir inundaciones repentinas a traves de la costa del
Golfo Central hacia Lower Mississippi Valley y y el oeste del
Tennessee Valley.

INUNDACIONES COSTERAS: Mareas sobre lo normal e inundaciones costeras
asociadas son posibles a traves de sectores de la costa norte del
