It’s a good principle that, except in exceptional circumstances, architecture critics see the buildings that they review. You wouldn’t want a food critic to recommend a restaurant based on photographs of loaded plates, nor a theatre critic to base their judgments on films of performances. You’re meant to sniff, feel and sense the things you describe, to experience them in the round.

There’s a downside to this good principle, which is that buildings don’t come to you, and that many fine works are done all over the world which it would be absurdly expensive and eco-reckless to visit. Circumstances are, what’s more, what can only be described as exceptional. Given that almost everywhere is now almost equally inaccessible, whether another county or another continent, I’ve decided to make an opportunity out of a problem, and to offer a world tour of the very best new architecture, as seen through the portal of a laptop.

Good design in any discipline usually carries a structure of order and harmony. Since the Renaissance, artists and architects have used a strong understanding of proportions to create aesthetically pleasing architecture. Many of these classical design principles have followed us into modern times and can be found today in effective web design.

Take an A4 piece of paper for example. If you take it and halve it, the resulting size is A5 with the same exact proportions. No other proportion has the same properties. 16th century architect, Andrea Palladio knew this well. It is believed that because, fundamentally, most architects—like Palladio—use a similar system of proportions to plan and design spaces, buildings can look very different while remaining similar at their cores.

Plasmodium vivax and human hexokinases share similar active sites but display distinct quaternary architectures

Malaria is a devastating disease caused by a protozoan parasite. It affects over 300 million individuals and results in over 400 000 deaths annually, most of whom are young children under the age of five. Hexokinase, the first enzyme in glucose metabolism, plays an important role in the infection process and represents a promising target for therapeutic intervention. Here, cryo-EM structures of two conformational states of Plasmodium vivax hexokinase (PvHK) are reported at resolutions of ∼3 Å. It is shown that unlike other known hexokinase structures, PvHK displays a unique tetrameric organization (∼220 kDa) that can exist in either open or closed quaternary conformational states. Despite the resemblance of the active site of PvHK to its mammalian counterparts, this tetrameric organization is distinct from that of human hexokinases, providing a foundation for the structure-guided design of parasite-selective antimalarial drugs.
