In May, the company also made VMware Workstation Pro and Fusion Pro free for personal use, allowing students and home users to set up virtualized test labs and experiment with other OSs by running virtual machines and Kubernetes clusters on Windows, Linux, and macOS devices. Starting this week, the Pro versions and the two products will no longer be available under a paid subscription model. "Effective immediately, both VMware Fusion and VMware Workstation will transition away from the paid subscription model, meaning you can now utilize these tools without any cost. The paid versions of these offerings -- Workstation Pro and Fusion Pro -- are no longer available for purchase," said Broadcom product marketing director Himanshu Singh. "If you're currently under a commercial contract, you can rest easy knowing that your agreement will remain in effect until the end of your term. You will continue to receive the full level of service and enterprise-grade support as per your contract." While the free versions will include all the features available in the paid products, Broadcom will no longer provide users with support ticketing for troubleshooting. Broadcom plans to continue developing new features and improvements and ensure that updates are rolled out promptly. "We're actively investing in new features, usability improvements, and other valuable enhancements," Singh added. "Our engineering teams are committed to maintaining our high standards for stability, with timely updates and reliable performance." We tested how the mutations causing recessive congenital stationary night blindness (CSNB), recessive Leber's congenital amaurosis (LCA1), and dominant cone–rod dystrophy-6 (CORD6) affected RetGC1 activity and regulation by RetGC-activating proteins (GCAPs) and retinal degeneration-3 protein (RD3). CSNB mutations R666W, R761W, and L911F, as well as LCA1 mutations R768W and G982VfsX39, disabled RetGC1 activation by human GCAP1, -2, and -3. The R666W and R761W substitutions compromised binding of GCAP1 with RetGC1 in HEK293 cells. In contrast, G982VfsX39 and L911F RetGC1 retained the ability to bind GCAP1 in cyto but failed to effectively bind RD3. R768W RetGC1 did not bind either GCAP1 or RD3. The co-expression of GUCY2D allelic combinations linked to CSNB did not restore RetGC1 activity in vitro. The CORD6 mutation R838S in the RetGC1 dimerization domain strongly dominated the Ca2+ sensitivity of cyclase regulation by GCAP1 in RetGC1 heterodimer produced by co-expression of WT and the R838S subunits. It required higher Ca2+ concentrations to decelerate GCAP-activated RetGC1 heterodimer—6-fold higher than WT and 2-fold higher than the Ser838-harboring homodimer. The heterodimer was also more resistant than homodimers to inhibition by RD3. The observed biochemical changes can explain the dominant CORD6 blindness and recessive LCA1 blindness, both of which affect rods and cones, but they cannot explain the selective loss of rod function in recessive CSNB. The properties of these channels vary with distinct combinations of Cav subunits and alternative splicing of the encoding transcripts. Therefore, the impact of disease-causing mutations affecting these channels may depend on the identities of Cav subunits and splice variants. Here, we analyzed the effects of a congenital stationary night blindness type 2 (CSNB2)-causing mutation, I745T (IT), in Cav1.4 channels typical of those in human retina: Cav1.4 splice variants with or without exon 47 (Cav1.4+ex47 and Cav1.4Δex47, respectively), and the auxiliary subunits, β2X13 and α2δ-4. We find that IT caused both Cav1.4 splice variants to activate at significantly more negative voltages and with slower deactivation kinetics than the corresponding WT channels. These effects of the IT mutation, along with unexpected alterations in ion selectivity, were generally larger in channels lacking exon 47. The weaker ion selectivity caused by IT led to hyperpolarizing shifts in the reversal potential and large outward currents that were evident in channels containing the auxiliary subunits β2X13 and α2δ-4 but not in those with β2A and α2δ-1. We conclude that the IT mutation stabilizes channel opening and alters ion selectivity of Cav1.4 in a manner that is strengthened by exclusion of exon 47 and inclusion of β2X13 and α2δ-4. Our results reveal complex actions of IT in modifying the properties of Cav1.4 channels, which may influence the pathological consequences of this mutation in retinal photoreceptors. Full Article

Harvey was awarded the 50,000-pound ($64,000) prize for what she has called a "space pastoral" about six orbiting astronauts, which she began writing during COVID-19 pandemic lockdowns. The confined characters loop through 16 sunrises and 16 sunsets over the course of a day, trapped in one another's company and transfixed by the globe's ever-changing vistas. "To look at the Earth from space is like a child looking into a mirror and realizing for the first time that the person in the mirror is herself," said Harvey, who researched her novel by reading books by astronauts and watching the space station's live camera. "What we do to the Earth we do to ourselves." She said the novel "is not exactly about climate change, but implied in the view of the Earth is the fact of human-made climate change." She dedicated the prize to everyone who speaks "for and not against the Earth, for and not against the dignity of other humans, other life." "All the people who speak for and call for and work for peace — this is for you," she said. Writer and artist Edmund de Waal, who chaired the five-member judging panel, called "Orbital" a "miraculous novel" that "makes our world strange and new for us." Gaby Wood, chief executive of the Booker Prize Foundation, noted that "in a year of geopolitical crisis, likely to be the warmest year in recorded history," the winning book was "hopeful, timely and timeless." Harvey, who has written four previous novels and a memoir about insomnia, is the first British writer since 2020 to win the Booker. The prize is open to English-language writers of any nationality and has a reputation for transforming writers' careers. Previous winners include Ian McEwan, Margaret Atwood, Salman Rushdie and Hilary Mantel. De Waal praised the "crystalline" writing and "capaciousness" of Harvey's succinct novel — at 136 pages in its U.K. paperback edition, one of the shortest-ever Booker winners. "This is a book that repays slow reading," he said. He said the judges spent a full day picking their winner and came to a unanimous conclusion. Harvey beat five other finalists from Canada, the United States, Australia and the Netherlands, chosen from among 156 novels submitted by publishers. American writer Percival Everett had been the bookies' favorite to win with "James," which reimagines Mark Twain's "Huckleberry Finn" from the point of view of its main Black character, the enslaved man Jim. The other finalists were American writer Rachel Kushner's spy story "Creation Lake"; Canadian Anne Michaels' poetic novel "Held"; Charlotte Wood's Australian saga "Stone Yard Devotional"; and "The Safekeep" by Yael van der Wouden, the first Dutch author to be shortlisted for the Booker. Harvey is the first female Booker winner since 2019, though one of five women on this year's shortlist, the largest number in the prize's 55-year history. De Waal said issues such as the gender or nationality of the authors were "background noise" that did not influence the judges. "There was absolutely no question of box ticking or of agendas or of anything else. It was simply about the novel," he said before the awards ceremony at Old Billingsgate, a grand former Victorian fish market in central London. Founded in 1969, the Booker Prize is open to novels originally written in English published in the U.K. or Ireland. Last year's winner was Irish writer Paul Lynch for post-democratic dystopia "Prophet Song." Lynch handed Harvey her Booker trophy at the ceremony, warning her that her life was about to change dramatically because of the Booker publicity boost.
