Iconic music producer Quincy Jones passed away just yesterday. He would have seen GPS. Does Helen Mirren not think this is as sad of a loss, because at least he died with the immense satisfaction of having watched a little dot that represented his car being driven towards a destination, thus making it a full and worthwhile life? Helen Mirren has yet to comment. Trash Pandas In the News A Sam's Club in Maryland had to shut down last week due to a racoon breaking into their bakery. A statement from the store read "after attempts to locate the raccoon were unsuccessful, Sam's Club representatives were advised best practices to properly capture the animal." Which means that during the initial attempts to capture the animal, they had no idea what the best practices were, and it must have been so funny. Imagine that raccoon's surprise to learn that there is a way to eat croissants, pre-trash? Going back to garbage pastries after that has got to be a bummer. Anyway, as the kids say, "in da clerb we all fam," but the clerb is a Sam's Club. Sam's Clerb, as it were. Okay, I just realized there's a bookshelf in my office that I haven't yet reorganized and if I don't do that right now it will make it so college-educated women in Michigan won't turn out in numbers, which makes a lot of sense when you think about it. The first step is the formation of a PvNMT–myristoyl–CoA binary complex that can bind to peptides. Understanding how inhibitors prevent protein binding will facilitate the development of PvNMT as a viable drug target. NMTs are secreted in all life stages of malarial parasites, making them attractive targets, unlike current antimalarials that are only effective during the plasmodial erythrocytic stages. The 2.3 Å resolution crystal structure of the ternary complex of PvNMT with myristoyl-CoA and a novel inhibitor is reported. One asymmetric unit contains two monomers. The structure reveals notable differences between the PvNMT and human enzymes and similarities to other plasmodial NMTs that can be exploited to develop new antimalarials. Full Article text

Soft modes were suggested to entropically drive such pore breathing through enhanced vibrational dynamics at high temperatures. In this work, oxygen K-edge resonant X-ray emission spectroscopy of the MIL-53(Al) MOF was performed to selectively probe the electronic perturbation accompanying pore breathing dynamics at the ligand carboxylate site for metal–ligand interaction. It was observed that the temperature-induced vibrational dynamics involves switching occupancy between antisymmetric and symmetric configurations of the carboxylate oxygen lone pair orbitals, through which electron density around carboxylate oxygen sites is redistributed and metal–ligand interactions are tuned. PMM offers a very powerful tool for comprehensive electronic structure analyses in real and momentum spaces. In this work, a VUV beam with variable polarization in the normal-incidence geometry was obtained at the same sample position as the soft X-ray beam from BL6U by branching the VUV beamline BL7U. The valence electronic structure of the Au(111) surface was measured using horizontal and vertical linearly polarized (s-polarized) light excitations from BL7U in addition to horizontal linearly polarized (p-polarized) light excitations from BL6U. Such highly symmetric photoemission geometry with normal incidence offers direct access to atomic orbital information via photon polarization-dependent transition-matrix-element analysis. Orbital angular momentum (OAM) beams hold substantial promise in various domains such as ultra-high-capacity optical communication, rotating body detection, optical tweezers, laser processing, super-resolution imaging etc. Hence, the advancement of OAM beam-generation technology and the enhancement of its technical proficiency and characterization capabilities are of paramount importance. These endeavours will not only facilitate the use of OAM beams in the aforementioned sectors but also extend the scope of applications in diverse fields related to OAM beams. At the FERMI Free-Electron Laser (Trieste, Italy), OAM beams are generated either by tailoring the emission process on the undulator side or, in most cases, by coupling a spiral zone plate (SZP) in tandem with the refocusing Kirkpatrick–Baez active optic system (KAOS). To provide a robust and reproducible workflow to users, a Hartmann wavefront sensor (WFS) is used for both optics tuning and beam characterization. KAOS is capable of delivering both tightly focused and broad spots, with independent control over vertical and horizontal magnification. This study explores a novel non-conventional `near collimation' operational mode aimed at generating beams with OAM that employs the use of a lithographically manufactured SZP to achieve this goal. The article evaluates the mirror's performance through Hartmann wavefront sensing, offers a discussion of data analysis methodologies, and provides a quantitative analysis of these results with ptychographic reconstructions. bit Biophysical and structural study of La Crosse virus endonuclease inhibition for the development of new antiviral options By journals.iucr.org Published On :: 2024-04-24 The large Bunyavirales order includes several families of viruses with a segmented ambisense (−) RNA genome and a cytoplasmic life cycle that starts by synthesizing viral mRNA. The initiation of transcription, which is common to all members, relies on an endonuclease activity that is responsible for cap-snatching. In La Crosse virus, an orthobunyavirus, it has previously been shown that the cap-snatching endonuclease resides in the N-terminal domain of the L protein. Orthobunyaviruses are transmitted by arthropods and cause diseases in cattle. However, California encephalitis virus, La Crosse virus and Jamestown Canyon virus are North American species that can cause encephalitis in humans. No vaccines or antiviral drugs are available. In this study, three known Influenza virus endonuclease inhibitors (DPBA, L-742,001 and baloxavir) were repurposed on the La Crosse virus endonuclease. Their inhibition was evaluated by fluorescence resonance energy transfer and their mode of binding was then assessed by differential scanning fluorimetry and microscale thermophoresis. Finally, two crystallographic structures were obtained in complex with L-742,001 and baloxavir, providing access to the structural determinants of inhibition and offering key information for the further development of Bunyavirales endonuclease inhibitors. New treatments are needed because the currently used drug, ivermectin, is contraindicated in pregnant women and those co-infected with Loa loa. The Seattle Structural Genomics Center for Infectious Disease (SSGCID) produced, crystallized and determined the apo structure of N-terminally hexahistidine-tagged O. volvulus macrophage migration inhibitory factor-1 (His-OvMIF-1). OvMIF-1 is a possible drug target. His-OvMIF-1 has a unique jellyfish-like structure with a prototypical macrophage migration inhibitory factor (MIF) trimer as the `head' and a unique C-terminal `tail'. Deleting the N-terminal tag reveals an OvMIF-1 structure with a larger cavity than that observed in human MIF that can be targeted for drug repurposing and discovery. Removal of the tag will be necessary to determine the actual biological oligomer of OvMIF-1 because size-exclusion chomatographic analysis of His-OvMIF-1 suggests a monomer, while PISA analysis suggests a hexamer stabilized by the unique C-terminal tails. Complex 1 crystallizes in the P21/n space group with two crystallographically independent but chemically identical [Pr(CB6)(NO3)(H2O)5]2+ complex cations, four nitrate counter-anions and 19.12 interstitial water molecules per asymmetric unit. The nonacoordinated PrIII in 1 are located in the PrO9 coordination environment formed by two carbonyl O atoms from bidentate cucurbit[6]uril units, two oxygen atoms from the bidentate nitrate anion and five water molecules. Considering the differences in Pr—O bond distances and O—Pr—O angles in the coordination spheres, the coordination polyhedrons of the two PrIII atoms can be described as distorted spherical capped square antiprismatic and muffin polyhedral. The multi-component solvated Lopinavir crystal, systematic name (2S)-N-[(2S,4S,5S)-5-[2-(2,6-dimethylphenoxy)acetamido]-4-hydroxy-1,6-diphenylhexan-2-yl]-3-methyl-2-(2-oxo-1,3-diazinan-1-yl)butanamide–ethane-1,2-diol–water (8/3/7) 8C37H48N4O5·3C2H6O2·7H2O, was prepared using evaporative methods. The crystalline material obtained from this experimental synthesis was characterized and elucidated by single-crystal X-ray diffraction (SC-XRD). The crystal structure is unusual in that the unit cell contains 18 molecules. The stoichiometric ratio of this crystal is eight Lopinavir molecules [8(C37H48N4O5)], three ethane-1,2-diol molecules [3(C2H6O2)] and seven water molecules [7(H2O)]. (WULF:NASDAQ) with a price target of US$7.50. The report follows TeraWulf's October production update and insights into its HPC capacity expansion plans. Aftahi highlighted the company's HPC capacity development, stating, "WULF gained better insight into its supply chain for delivering ~72.5MW of HPC capacity by 2Q25, with the potential to generate ~US$90M in annualized revenue with US$60M+ in profit (exit run rate). Its 2.5MW concept building is now complete, and its 20MW facility is on schedule for 1Q25." Regarding October's production metrics, the analyst noted, "WULF mined 150 BTC in October, up 7% m/m on a comparable basis to only Lake Mariner in September (140 BTC). The ending hash rate is now 8.1 EH/s (vs. 10) from the removal of Nautilus." He added that "machine efficiency is now 22 J/TH vs. the prior combined rate of ~24.6 J/TH." The analyst observed operational improvements, commenting, "Avg. operating hash rate was ~6.8 or ~84%, slightly ahead of prior months (although reported in aggregate). Part of this is being driven by the accelerated replacement of older miners at Lake Mariner." The report noted that upon reinstallation of damaged miners at Lake Mariner, the hash rate would increase to approximately 8.7 EH/s, though power cost per BTC increased about 5% month over month. Roth MKM's valuation methodology is based on an EBITDA multiple approach. Aftahi explained, "We apply a ~24x multiple to our projected FY25 adj. EBITDA of ~US$120M to arrive at our price target of US$7.50." The analyst outlined several risk factors, including Bitcoin price volatility, regulatory changes, unproven new business segments, capital requirements, weather impacts, competition, power contracts, and reliance on third-party suppliers. In conclusion, Roth MKM's maintenance of their Buy rating and US$7.50 price target reflects confidence in TeraWulf's operational execution and growth potential in both Bitcoin mining and HPC capacity. Hinkle's "The Evanesced" was inspired by the #SayHerName movement against police violence, as well as Los Angeles's Grim Sleeper serial killer. Hinkle depicts black women in the nude, twisting and writhing, as though they're sinking back into the canvas. Or are they reemerging from it? Deputy Director of the California African American Museum Naima Keith says Hinkle's exhibit looks at the "historical present," the way in which history still affects us today, harkening back to slavery and Jim Crow. Keith says the main issue Hinkle is addressing is the invisibility of black women, especially those who are abused or in danger. Hinkle was particularly inspired by the South LA serial killer "The Grim Sleeper." He is accused of murdering over one hundred women from the 1980's onward, until being captured in 2007. Many of his victims were women of color according to the Los Angeles Police Department. "He had been killing prostitutes and runaways and drug addicted women," says Keith, noting that some saw these deaths as occupational hazards. Most of Hinkle's subjects in the paintings and sketches in "The Evanesced" are clearly nude. This was a deliberate choice to showcase femininity, according to Keith. She says: She’s talking about being women... There’s love, there’s joy, there’s pain. All things we experience as all women... But [nudity], I think, allows us to focus on the female form, not necessarily get caught up on what they are wearing or what they’re doing. In the artwork, viewers can see that every face, body, and hair style is completely unique to each sketch or painting. Keith says this helps the viewer appreciate the diversity amongst women of color. She says: You have women that are smiling. You have women that are looking at you- you know- lovingly, shyly. Not every one, not every image in the show is about negativity, disappearance, or sadness. There is a bit of celebration. There’s interaction between multiple women. That’s what makes the body of work so interesting: it’s not just seeing women of color through one lens. There’s the possibility of seeing them through, like I said, disappearance, and also the freedom to have a wide range of emotions. There is one painting that continues to draw Naima Keith back to it. It is called "Uproot 2017" and it features a feminine figure with three exposed breasts. She says this painting speaks to her about motherhood and the connection women have with their changing bodies. Keith says: I asked Kenyatta why she depicts women with multiple [extra] breasts and we had a conversation about being moms. Kenyatta and I are both mothers of young children... As moms, we just kinda talked about how things aren't what they used to be, in terms of where they used to be.
