Lasso Meets Horseshoe: A Survey

Abstract:
The goal of this paper is to contrast and survey the major advances in two of the most commonly used high-dimensional techniques, namely, the Lasso and horseshoe regularization. Lasso is a gold standard for predictor selection while horseshoe is a state-of-the-art Bayesian estimator for sparse signals. Lasso is fast and scalable and uses convex optimization whilst the horseshoe is nonconvex. Our novel perspective focuses on three aspects: (i) theoretical optimality in high-dimensional inference for the Gaussian sparse model and beyond, (ii) efficiency and scalability of computation and (iii) methodological development and performance.

Their project engaged the community and accelerated research by providing a level playing field for comparing the performance of a priori specified algorithms. DHSSC identified themes concerning characteristics of the DGP, properties of the estimators, and inference. We discuss these themes in the context of targeted learning.

The Right Temporoparietal Junction Is Causally Associated with Embodied Perspective-taking

A prominent theory claims that the right temporoparietal junction (rTPJ) is especially associated with embodied processes relevant to perspective-taking. In the present study, we use high-definition transcranial direct current stimulation to provide evidence that the rTPJ is causally associated with the embodied processes underpinning perspective-taking. Eighty-eight young human adults were stratified to receive either rTPJ or dorsomedial PFC anodal high-definition transcranial direct current stimulation in a sham-controlled, double-blind, repeated-measures design. Perspective-tracking (line-of-sight) and perspective-taking (embodied rotation) were assessed using a visuo-spatial perspective-taking task that required understanding what another person could see or how they see it, respectively. Embodied processing was manipulated by positioning the participant in a manner congruent or incongruent with the orientation of an avatar on the screen. As perspective-taking, but not perspective-tracking, is influenced by bodily position, this allows the investigation of the specific causal role for the rTPJ in embodied processing. Crucially, anodal stimulation to the rTPJ increased the effect of bodily position during perspective-taking, whereas no such effects were identified during perspective-tracking, thereby providing evidence for a causal role for the rTPJ in the embodied component of perspective-taking. Stimulation to the dorsomedial PFC had no effect on perspective-tracking or taking. Therefore, the present study provides support for theories postulating that the rTPJ is causally involved in embodied cognitive processing relevant to social functioning.

SIGNIFICANCE STATEMENT The ability to understand another's perspective is a fundamental component of social functioning. Adopting another perspective is thought to involve both embodied and nonembodied processes. The present study used high-definition transcranial direct current stimulation (HD-tDCS) and provided causal evidence that the right temporoparietal junction is involved specifically in the embodied component of perspective-taking. Specifically, HD-tDCS to the right temporoparietal junction, but not another hub of the social brain (dorsomedial PFC), increased the effect of body position during perspective-taking, but not tracking. This is the first causal evidence that HD-tDCS can modulate social embodied processing in a site-specific and task-specific manner.

Contribution of NPY Y5 Receptors to the Reversible Structural Remodeling of Basolateral Amygdala Dendrites in Male Rats Associated with NPY-Mediated Stress Resilience

Endogenous neuropeptide Y (NPY) and corticotrophin-releasing factor (CRF) modulate the responses of the basolateral amygdala (BLA) to stress and are associated with the development of stress resilience and vulnerability, respectively. We characterized persistent effects of repeated NPY and CRF treatment on the structure and function of BLA principal neurons in a novel organotypic slice culture (OTC) model of male rat BLA, and examined the contributions of specific NPY receptor subtypes to these neural and behavioral effects. In BLA principal neurons within the OTCs, repeated NPY treatment caused persistent attenuation of excitatory input and induced dendritic hypotrophy via Y5 receptor activation; conversely, CRF increased excitatory input and induced hypertrophy of BLA principal neurons. Repeated treatment of OTCs with NPY followed by an identical treatment with CRF, or vice versa, inhibited or reversed all structural changes in OTCs. These structural responses to NPY or CRF required calcineurin or CaMKII, respectively. Finally, repeated intra-BLA injections of NPY or a Y5 receptor agonist increased social interaction, a validated behavior for anxiety, and recapitulated structural changes in BLA neurons seen in OTCs, while a Y5 receptor antagonist prevented NPY's effects both on behavior and on structure. These results implicate the Y5 receptor in the long-term, anxiolytic-like effects of NPY in the BLA, consistent with an intrinsic role in stress buffering, and highlight a remarkable mechanism by which BLA neurons may adapt to different levels of stress. Moreover, BLA OTCs offer a robust model to study mechanisms associated with resilience and vulnerability to stress in BLA.

SIGNIFICANCE STATEMENT Within the basolateral amygdala (BLA), neuropeptide Y (NPY) is associated with buffering the neural stress response induced by corticotropin releasing factor, and promoting stress resilience. We used a novel organotypic slice culture model of BLA, complemented with in vivo studies, to examine the cellular mechanisms associated with the actions of NPY. In organotypic slice cultures, repeated NPY treatment reduces the complexity of the dendritic extent of anxiogenic BLA principal neurons, making them less excitable. NPY, via activation of Y5 receptors, additionally inhibits and reverses the increases in dendritic extent and excitability induced by the stress hormone, corticotropin releasing factor. This NPY-mediated neuroplasticity indicates that resilience or vulnerability to stress may thus involve neuropeptide-mediated dendritic remodeling in BLA principal neurons.

Ventral Hippocampal Input to the Prelimbic Cortex Dissociates the Context from the Cue Association in Trace Fear Memory

The PFC, through its high degree of interconnectivity with cortical and subcortical brain areas, mediates cognitive and emotional processes in support of adaptive behaviors. This includes the formation of fear memories when the anticipation of threat demands learning about temporal or contextual cues, as in trace fear conditioning. In this variant of fear learning, the association of a cue and shock across an empty trace interval of several seconds requires sustained cue-elicited firing in the prelimbic cortex (PL). However, it is unknown how and when distinct PL afferents contribute to different associative components of memory. Among the prominent inputs to PL, the hippocampus shares with PL a role in both working memory and contextual processing. Here we tested the necessity of direct hippocampal input to the PL for the acquisition of trace-cued fear memory and the simultaneously acquired contextual fear association. Optogenetic silencing of ventral hippocampal (VH) terminals in the PL of adult male Long-Evans rats selectively during paired trials revealed that direct communication between the VH and PL during training is necessary for contextual fear memory, but not for trace-cued fear acquisition. The pattern of the contextual memory deficit and the disruption of local PL firing during optogenetic silencing of VH-PL suggest that the VH continuously updates the PL with the current contextual state of the animal, which, when disrupted during memory acquisition, is detrimental to the subsequent rapid retrieval of aversive contextual associations.

SIGNIFICANCE STATEMENT Learning to anticipate threat from available contextual and discrete cues is crucial for survival. The prelimbic cortex is required for forming fear memories when temporal or contextual complexity is involved, as in trace fear conditioning. However, the respective contribution of distinct prelimbic afferents to the temporal and contextual components of memory is not known. We report that direct input from the ventral hippocampus enables the formation of the contextual, but not trace-cued, fear memory necessary for the subsequent rapid expression of a fear response. This finding dissociates the contextual and working-memory contributions of prelimbic cortex to the formation of a fear memory and demonstrates the crucial role for hippocampal input in contextual fear learning.

Basigin Associates with Integrin in Order to Regulate Perineurial Glia and Drosophila Nervous System Morphology

The Drosophila nervous system is ensheathed by a layer of outer glial cells, the perineurial glia, and a specialized extracellular matrix, the neural lamella. The function of perineurial glial cells and how they interact with the extracellular matrix are just beginning to be elucidated. Integrin-based focal adhesion complexes link the glial membrane to the extracellular matrix, but little is known about integrin's regulators in the glia. The transmembrane Ig domain protein Basigin/CD147/EMMPRIN is highly expressed in the perineurial glia surrounding the Drosophila larval nervous system. Here we show that Basigin associates with integrin at the focal adhesions to uphold the structure of the glia-extracellular matrix sheath. Knockdown of Basigin in perineurial glia using RNAi results in significant shortening of the ventral nerve cord, compression of the glia and extracellular matrix in the peripheral nerves, and reduction in larval locomotion. We determined that Basigin is expressed in close proximity to integrin at the glial membrane, and that expression of the extracellular integrin-binding domain of Basigin is sufficient to rescue peripheral glial compression. We also found that a reduction in expression of integrin at the membrane rescues the ventral nerve cord shortening, peripheral glial compression, and locomotor phenotypes, and that reduction in the integrin-binding protein Talin can partially rescue glial compression. These results identify Basigin as a potential negative regulator of integrin in the glia, supporting proper glial and extracellular matrix ensheathment of the nervous system.

SIGNIFICANCE STATEMENT The glial cells and extracellular matrix play important roles in supporting and protecting the nervous system, but the interactions between these components have not been well characterized. Our study identified expression of a conserved Ig superfamily protein, Basigin, at the glial membrane of Drosophila where it associates with the integrin-based focal adhesion complexes to ensure proper ensheathment of the CNS and PNS. Loss of Basigin in the glia results in an overall compression of the nervous system due to integrin dysregulation, which causes locomotor defects in the animals. This underlies the importance of glia-matrix communication for structural and functional support of the nervous system.

Data contradicts Harvard professor's assertions about homeschooling

Denver Newsroom, May 7, 2020 / 05:29 pm (CNA).- A Notre Dame sociologist is using data to challenge a Harvard Law professor’s assertions that homeschooling is “dangerous”, and detrimental to society.

The controversy stems from a recent paper by professor Elizabeth Bartholet in which she calls for a presumptive ban on homeschooling in the United States.

Bartholet, as quoted in a Harvard Magazine piece based on her paper, points to unspecified “surveys of homeschoolers” to assert that “up to 90 percent” of homeschooling families are “driven by conservative Christian beliefs, and seek to remove their children from mainstream culture.”

“Some” homeschooling parents are “‘extreme religious ideologues’ who question science and promote female subservience and white supremacy,” she writes.

David Sikkink, associate professor in the Department of Sociology at the University of Notre Dame, analyzed surveys of homeschooling families— including a 2016 government survey—  and found that these families are not overwhelmingly Christian nor religious, and are not as universally closed-off to the outside world as Bartholet asserts.

In the analysis Sikkink conducted, just 16% of homeschooling parents said they were homeschooling primarily for religious reasons. The number one reason homeschooling parents cited was a concern about school environment, such as safety, drugs, or negative peer pressure.

Eleven percent of parents reported homeschooling because their child has special needs.

While approximately half of the homeschooling parents surveyed mentioned religion as a factor in their decision to homeschool, Sikkink notes that the parents who cited religion as a reason were, on the whole, more highly educated than those parents who did not.

In terms of Bartholet’s assertion that some homeschooling parents “believe that women should be totally subservient to men and educated in ways that promote such subservience,” Sikkink’s analysis did not find evidence that religious households oppose higher education for girls.

Among the homeschooling families in the survey who use a religious curriculum, there was no difference in their self-reported educational expectations— i.e., what education level they expected their children to reach—  for their male children vs. their female children.

Several past studies have shown that homeschool students typically outperform their public and private school counterparts on things like standardized tests and college performance. A 2016 study from the National Council on Measurement in Education showed that, when adjusted for demographic factors, homeschool students were on par academically with their demographically-similar peers.

Moreover, the data Sikkink analyzed suggests that after family background and demographic controls are accounted for, about 64% of homeschoolers “completely agree” that they have much in life to be thankful for, compared to 53% of public schoolers.

On feelings of helplessness, or lack or goals or direction in life, homeschoolers do not substantially differ from their public school counterparts, the analysis suggests.

In the Arizona Law Review, Bartholet argues that while homeschool children may perform as well as their peers on standardized tests or in college, they are also often isolated from their peers and denied experiences and exposures that would make them more productive citizens.

Bartholet claims in her article that “a very large proportion of homeschooling parents are ideologically committed to isolating their children from the majority culture and indoctrinating them in views and values that are in serious conflict with that culture.”

“Isolated families,” she asserts, “constitute a significant part of the homeschooling world.”

In contrast, Sikkink’s analysis found that among the schooling groups surveyed, homeschooling families had the highest level of “community involvement” of all school sectors.

“Community involvement” activities included attending sporting events, attending concerts, going to the zoo or aquarium, going to a museum, going to a library, visiting a bookstore, or attending an event sponsored by a community, religious, or ethnic group.

Homeschooling graduates are almost identical to their public school counterparts in likelihood to vote in federal and local elections, Sikkink found.

Furthermore, the total number of volunteer and community service hours for homeschooling graduates is very similar to or slightly higher than public school graduates, the analysis found.

Bartholet asserts that some homeschoolers “engage in homeschooling to promote racist ideologies and avoid racial intermingling.”

In contrast: “The reality is that about 41% of homeschooled children are racial and ethnic minorities,” Sikkink writes.

“When asked about four closest friends, about 37% of young adult homeschoolers...mention someone of a different race or ethnicity—exactly the same as public schoolers.”

This diversity also extends to schooling practices— increasingly, Sikkink says, homeschooling adopts new forms, including “hybrids” that combine the benefits of home and institutional schooling.

“About 57 percent of homeschoolers are using some form of instruction outside the family,” Sikkink told CNA in an email.

“That includes using tutors, private or public schools, colleges or universities, or homeschooling coops. That percentage would be higher if we included those who reported obtaining curriculum from formal institutions, such as public schools.”

Moreover, about a third of homeschooling parents obtain their curriculum or books from a public school or school district.

“Altogether, 46% of homeschoolers have some pedagogical relationship with public schools,” Sikkink asserts.

Bartholet argues that homeschooling puts children at risk of abuse by their parents, while if children were in public schools, they would be among teachers who are mandatory reporters of any suspected abuse that may be taking place.

“The issue is, do we think that parents should have 24/7, essentially authoritarian control over their children from ages zero to 18? I think that’s dangerous,” Bartholet asserts in the Harvard Magazine piece.

“I think it’s always dangerous to put powerful people in charge of the powerless, and to give the powerful ones total authority.”

Sikkink says Bartholet’s image of a child confined to the home “24/7...from ages zero to 18” is not consistent with the data.

“When we look at the use of homeschooling for each year of the child's upbringing, we only find a small percentage that report that the child was homeschooled for all their years of schooling,” Sikkink told CNA in an email.

Many of these students are part-time public schoolers— about 25% of homeschoolers receive some instruction in public schools during their school-age careers, he wrote.

Homeschooling regulations vary widely by state. Sikkink told CNA he hopes future studies will examine the effects of state-level variation in regulation on homeschooling quality.

“The question of schooling oversight remains, of course, but it would be short-sighted not to keep homeschooling and other creative schooling options in the mix, including the hybrid models that cross sector boundaries,” Sikkink concludes.

 

Subsequent to the publication of this story, Sikkink told CNA he had revised his assessment of the percentage of homeschoolers using instruction outside the family, from 64% to 57%. The story has been updated to reflect that assessment.

Lessons learned during past pandemics - from a Catholic perspective

Coronavirus is only the latest iteration of an age-old human affliction. Even now, with the benefit of advanced medical science, our reaction – our confusion, our fear – is not so different from how our ancestors experienced recurrent and terrifying onslaughts of plague, cholera, and yellow fever across the ages. We can learn from the courage and ingenuity of those who travelled this road before us.
 
Consider the work of Dr. Carlos Finlay in Cuba. In 1880 he hypothesized, and then worked to prove his hypothesis, that yellow fever, a disease that regularly decimated coastal populations up and down the Americas, was spread by infected mosquitos. Those mosquitos came to our shores in the 17th century on African slave ships and attacked portal communities in the tropics as well as cities like New Orleans and Philadelphia. The resulting epidemics occurred with oppressive regularity in the summer months, to the people’s great dread, with mortality rates as high as 50 percent. The impact was tremendous – not only in the milllions of lives lost and the wretchedness this caused, but in economic gains and opportunities wiped out or delayed (the Panama Canal).
 
Connecting the transmission of the deadly virus to its source or vector was a decisive step forward in the long struggle against yellow fever. It preceded the development of a vaccine by more than 60 years. Here's how it happened: A young doctor, Carlos Finlay, returned to his home in Havana one night, exhausted, after caring for a Carmelite priest dying of yellow fever. Realizing he had forgotten to say his daily rosary, he sat in his armchair, sweating in the oppressive heat, fingering his beads and swatting at a bothersome mosquito. Suddenly, inspiration pierced his depression and weariness: Could the mosquito, like the one annoying him that moment, be transmitting the infection from person to person? If so, this was marvelous. One could not fight the brutal steamy summer air – the miasma – but one could fight mosquitos.
 
Inspiration, however, was not enough to proceed. Courage and even heroism would be needed to prove Finlay’s hypothesis. These were at hand, thanks to 57 young Jesuit priests and brothers who volunteered as experimental subjects. As each arrived from Spain to staff the Colegio de Belen, newly founded by Queen Isabel II of Spain, he was met by Finlay, carrying a test tube filled with mosquitos that had just fed on a patient sick with yellow fever. Taking their lives in their hands, these Jesuits allowed themselves to be bitten for the sake of their fellow human beings. Three died of the bite, but all 57 were willing to do the same.
 
Subsequent experiments supported Finlay’s hypothesis. Although a vaccine to definitively eradicate the disease would not come for decades, Finlay’s insight helped man to co-exist safely with yellow fever until that time. The incidence of yellow fever in Cuba dropped precipitously through mosquito control. Standing water, a breeding ground for the noxious pests, was eliminated where possible or treated aggressively with insecticides where not. Panama, where tens of thousands of workers had already died of the disease while building the canal followed Cuba’s lead. The last Panama Canal worker to die of yellow fever came in 1906.
 
There are important lessons for us here -- first and foremost, lessons in resourcefulness and valor. 
 
Already, thousands of human minds are, today, tenaciously working to find a solution to Covid-19. They’re persisting without respite, persisting through depression and fatigue, to find a way forward. Just as Dr. Finlay did.
 
And, you can depend on it, inspiration is sure to strike again.
 
You can also see today the same kind of valor that animated the Jesuit volunteers who let the infected mosquitos bite them. You see it in the countless men and women who keep showing up for work at nursing homes or crowded food production lines. Their examples help us all to keep up and increase our courage so we can join them as we ease back into our normal daily lives.
 
As we face the moment when we too realize that we have no choice but to go back out into the world of work and personal interactions, we can take hope from contemplating our predecessors’ success in confronting yellow fever. Like us, they dreamed of a vaccine. But they didn’t lock themselves away until it was developed. They found a way to steel themselves and then to steal the deadly efficiency away from the virus that plagued them. A century later, we can do the same.

In this pilot study, we prospectively identified baseline risk factors for the development of seizures in individuals with ASD and also identified characteristics sensitive to seizure onset up to 6 years after enrollment in the Autism Speaks Autism Treatment Network.

METHODS:

Children with ASD and no history of seizures at baseline who either experienced onset of seizures after enrollment in the Autism Treatment Network or remained seizure free were included in the analysis.

RESULTS:

Among 472 qualifying children, 22 (4.7%) experienced onset of seizures after enrollment. Individuals who developed seizures after enrollment exhibited lower scores at baseline on all domains of the Vineland Adaptive Behavior Scales, greater hyperactivity on the Aberrant Behavior Checklist (25.4 ± 11.8 vs 19.2 ± 11.1; P = .018), and lower physical quality of life scores on the Pediatric Quality of Life Inventory (60.1 ± 24.2 vs 76.0 ± 18.2; P < .001). Comparing change in scores from entry to call-back, adjusting for age, sex, length of follow-up, and baseline Vineland II composite score, individuals who developed seizures experienced declines in daily living skills (–8.38; 95% confidence interval –14.50 to –2.50; P = .005). Adjusting for baseline age, sex, and length of follow-up, baseline Vineland II composite score was predictive of seizure development (risk ratio = 0.95 per unit Vineland II composite score, 95% confidence interval 0.92 to 0.99; P = .007).

CONCLUSIONS:

Individuals with ASD at risk for seizures exhibited changes in adaptive functioning and behavior.

Eosinophilic Pneumonia and Lymphadenopathy Associated With Vaping and Tetrahydrocannabinol Use

Idiopathic acute eosinophilic pneumonia is a rare and potentially life-threatening condition that is defined by bilateral pulmonary infiltrates and fever in the presence of pulmonary eosinophilia. It often presents acutely in previously healthy individuals and can be difficult to distinguish from infectious pneumonia. Although the exact etiology of idiopathic acute eosinophilic pneumonia remains unknown, an acute hypersensitivity reaction to an inhaled antigen is suggested, which is further supported by recent public health risks of vaping (electronic cigarette) use and the development of lung disease. In this case, a patient with a year-long history of vaping in conjunction with tetrahydrocannabinol cartridge use who was diagnosed with idiopathic acute eosinophilic pneumonia with associated bilateral hilar lymphadenopathy is described.

Chorioamnionitis is associated with preterm birth and an increased risk of common adverse outcomes of prematurity, including early-onset sepsis and neurodevelopmental impairment. The effect of chorioamnionitis on postnatal immune function and risk of late-onset infection is unknown.
