ios Chrome for iOS Gets Google Lens Update, Shopping Insights and More By www.macrumors.com Published On :: Tue, 12 Nov 2024 09:00:00 PST Google today announced several new features for the Chrome browser that's available on the iPhone and the iPad, with the new additions aimed at providing users with more intuitive ways for finding information and completing tasks. Google Lens now supports searching with images and text at the same time, rather than search being limited to just an image. Users can add words to a visual query to refine results and conduct more complex searches. Google Drive and Google Photos users can now save content from the web directly to those services from Chrome, freeing up on-device storage space. To save a file to Google Drive from Chrome, tap on the Google Drive option. Saving an image to Photos from Chrome can be done by long pressing on the image and then selecting the Save in Google Photos option. For U.S. users, Chrome on iOS is gaining Shopping Insights, which are designed to better surface deals. If Chrome has Shopping Insights for a product that a user is searching for, there will be a "Good Deal Now" notification in the address bar. The feature requires signing into Chrome and toggling on "Make Searches and Browsing Better." When viewing a map of an address in Chrome, users will soon be able to tap an underlined address and see a more detailed mini-map of the location directly in the browser without having to swap over to Google Maps. The public betas come a day after Apple provided developers with new betas. Public beta testers can download the updates from the Settings app on each device after opting into the beta through Apple's public beta testing website. Note that Apple has also released public betas for watchOS 11.2, tvOS 18.2, and the latest HomePod software. iOS 18.2, iPadOS 18.2, and macOS Sequoia introduce the next Apple Intelligence features, including the first image generation capabilities. The update adds Image Playground, a new app for creating images based on text descriptions. You can enter anything you want, though Apple will suggest costumes, locations, items, and more to add to an image. There are options to create characters that resemble your friends and family, and you can choose a photo for Image Playground to use as inspiration to create a related image. Elements added to Image Playground creations are previewed, and there is a preview history so you can undo a change and go back to a prior version. While Image Playground is a standalone app, it is also integrated into Messages, Notes, Freeform, and more. Image Playground does not make photorealistic images and is instead limited to animation or illustration styles. The update also adds Genmoji, which are customizable emoji characters that you can create based on descriptions and phrases. Like Image Playground creations, you can base them on your friends and family, with the data pulled from the People album in Photos. You can also make characters using basic elements, and you'll get multiple Genmoji suggestions to choose from. You can create Genmoji using the emoji keyboard. Genmoji are limited to iOS 18.2 and iPadOS 18.2 right now, and will be coming to macOS Sequoia later. Siri in iOS 18.2, iPadOS 18.2, and macOS Sequoia 15.2 has ChatGPT integration. If Siri is unable to provide an answer to a question, Siri will hand the request over to ChatGPT, though Siri will need user permission first. ChatGPT will answer the question and relay the information back through Siri. ChatGPT can be used to create content from scratch, including text and images. No account is required to use ChatGPT integration, and Apple and OpenAI do not store requests. If you have an iPhone 16, there's a Visual Intelligence feature in iOS 18.2 that provides information about what's around you. Open up the camera and point it at a restaurant to get reviews, or point it at an item to search Google for it. Some other Visual Intelligence capabilities include reading text out loud, detecting phone numbers and addresses to add them to Contacts, copying text, and summarizing text. Apple added Writing Tools in iOS 18.1, but in iOS 18.2, you can more freely describe the tone or content change that you want to make, such as adding more action words, or turning an email into a poem. Apple Intelligence now supports localized English in Australia, Canada, New Zealand, South Africa, Ireland, and the UK in addition to U.S. English. Wait List If you've already been testing Apple Intelligence and are opted in, you will have access to Writing Tools, ChatGPT integration, and Visual Intelligence automatically. There is a secondary waiting list for early access to use Genmoji, Image Playground, and Image Wand. You can sign up to get access in Image Playground or in the areas where you access Genmoji or Image Wand. When you request access, you are added to a wait list for all three capabilities and you'll get a notification when the features are available for you to use. Apple will roll out access over time. Availability and Compatibility The public betas are available on all devices, but the Apple Intelligence features require a device capable of Apple Intelligence. We believe that Apple is likely planning to release iOS 18.2 on Monday, December 9, based on a recent notice that British carrier EE provided to its customers. In the notice, EE said it will be discontinuing an iPhone feature known as "Wi-Fi calling on other supported iCloud-connected devices" on December 9. The feature enables users to make Wi-Fi calls on devices like the iPad and Mac, even when their iPhone is not nearby. The second developer beta of iOS 18.2 last week came bundled with a new EE carrier settings version numbered 60.5.2, up from 59.5.1 on the first iOS 18.2 beta and iOS 18.1. The new carrier settings version removed the "Add Wi-Fi Calling For Other Devices" toggle from the Wi-Fi Calling section in the Settings app on iPhones used by EE customers, in line with the carrier announcing that it will stop offering the feature next month. Nonetheless, the regioselectivity control of the epoxide ring opening by EHs remains challenging. Alp1U is an α/β-fold EH that exhibits poor regioselectivity in the epoxide hydrolysis of fluostatin C (compound 1) and produces a pair of stereoisomers. Herein, we established the absolute configuration of the two stereoisomeric products and determined the crystal structure of Alp1U. A Trp-186/Trp-187/Tyr-247 oxirane oxygen hole was identified in Alp1U that replaced the canonical Tyr/Tyr pair in α/β-EHs. Mutation of residues in the atypical oxirane oxygen hole of Alp1U improved the regioselectivity for epoxide hydrolysis on 1. The single site Y247F mutation led to highly regioselective (98%) attack at C-3 of 1, whereas the double mutation W187F/Y247F resulted in regioselective (94%) nucleophilic attack at C-2. Furthermore, single-crystal X-ray structures of the two regioselective Alp1U variants in complex with 1 were determined. These findings allowed insights into the reaction details of Alp1U and provided a new approach for engineering regioselective epoxide hydrolases. Full Article

We have recently shown that biosynthesis of Ruminococcin C (RumC), a novel ribosomally synthesized and posttranslationally modified peptide (RiPP) produced by the commensal bacterium Ruminococcus gnavus, requires two radical SAM enzymes (RumMC1 and RumMC2) catalyzing the formation of four Cα-thioether bridges. These bridges, which are essential for RumC's antibiotic properties against human pathogens such as Clostridium perfringens, define two hairpin domains giving this sactipeptide (sulfur-to-α-carbon thioether–containing peptide) an unusual architecture among natural products. We report here the biochemical and spectroscopic characterizations of RumMC2. EPR spectroscopy and mutagenesis data support that RumMC2 is a member of the large family of SPASM domain radical SAM enzymes characterized by the presence of three [4Fe-4S] clusters. We also demonstrate that this enzyme initiates its reaction by Cα H-atom abstraction and is able to catalyze the formation of nonnatural thioether bonds in engineered peptide substrates. Unexpectedly, our data support the formation of a ketoimine rather than an α,β-dehydro-amino acid intermediate during Cα-thioether bridge LC–MS/MS fragmentation. Finally, we explored the roles of the leader peptide and of the RiPP precursor peptide recognition element, present in myriad RiPP-modifying enzymes. Collectively, our data support a more complex role for the peptide recognition element and the core peptide for the installation of posttranslational modifications in RiPPs than previously anticipated and suggest a possible reaction intermediate for thioether bond formation. ios Novel fluorescent GPCR biosensor detects retinal equilibrium binding to opsin and active G protein and arrestin signaling conformations [Molecular Biophysics] By www.jbc.org Published On :: 2020-12-18T00:06:18-08:00 Rhodopsin is a canonical class A photosensitive G protein–coupled receptor (GPCR), yet relatively few pharmaceutical agents targeting this visual receptor have been identified, in part due to the unique characteristics of its light-sensitive, covalently bound retinal ligands. Rhodopsin becomes activated when light isomerizes 11-cis-retinal into an agonist, all-trans-retinal (ATR), which enables the receptor to activate its G protein. We have previously demonstrated that, despite being covalently bound, ATR can display properties of equilibrium binding, yet how this is accomplished is unknown. Here, we describe a new approach for both identifying compounds that can activate and attenuate rhodopsin and testing the hypothesis that opsin binds retinal in equilibrium. Our method uses opsin-based fluorescent sensors, which directly report the formation of active receptor conformations by detecting the binding of G protein or arrestin fragments that have been fused onto the receptor's C terminus. We show that these biosensors can be used to monitor equilibrium binding of the agonist, ATR, as well as the noncovalent binding of β-ionone, an antagonist for G protein activation. Finally, we use these novel biosensors to observe ATR release from an activated, unlabeled receptor and its subsequent transfer to the sensor in real time. Taken together, these data support the retinal equilibrium binding hypothesis. The approach we describe should prove directly translatable to other GPCRs, providing a new tool for ligand discovery and mutant characterization. Full Article

Here, we review current understanding of the molecular mechanisms underlying hyaluronan and HAS2 regulation and the role of soluble proteoglycan in affecting autophagy and angiogenesis. Specifically, we assess the role of proteoglycan-evoked autophagy in regulating angiogenesis via the HAS2-hyaluronan axis and ATG9A, a novel HAS2 binding partner. We discuss extracellular hyaluronan biology and the post-transcriptional and post-translational modifications that regulate its main synthesizer, HAS2. We highlight the emerging group of proteoglycans that utilize outside-in signaling to modulate autophagy and angiogenesis in cancer microenvironments and thoroughly review the most up-to-date understanding of endorepellin signaling in vascular endothelia, providing insight into the temporal complexities involved. Considerable variability exists in the chain length and degree of saturation of both of these hydrophobic chains, and recent work has implicated ceramides with different LCBs and N-acyl chains in distinct biological processes; moreover, they may play different roles in disease states and possibly even act as prognostic markers. We now demonstrate that the half-life, or turnover rate, of ceramides containing diverse N-acyl chains is different. By means of a pulse-labeling protocol using stable-isotope, deuterated free fatty acids, and following their incorporation into ceramide and downstream SLs, we show that very-long-chain (VLC) ceramides containing C24:0 or C24:1 fatty acids turn over much more rapidly than long-chain (LC) ceramides containing C16:0 or C18:0 fatty acids due to the more rapid metabolism of the former into VLC sphingomyelin and VLC hexosylceramide. In contrast, d16:1 and d18:1 ceramides show similar rates of turnover, indicating that the length of the sphingoid LCB does not influence the flux of ceramides through the biosynthetic pathway. Together, these data demonstrate that the N-acyl chain length of SLs may not only affect membrane biophysical properties but also influence the rate of metabolism of SLs so as to regulate their levels and perhaps their biological functions. ios Radiosensitization by Kinase Inhibition Revealed by Phosphoproteomic Analysis of Pancreatic Cancer Cells [Research] By www.mcponline.org Published On :: 2020-10-01T00:05:25-07:00 Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive cancers and known for its extensive genetic heterogeneity, high therapeutic resistance, and strong variation in intrinsic radiosensitivity. To understand the molecular mechanisms underlying radioresistance, we screened the phenotypic response of 38 PDAC cell lines to ionizing radiation. Subsequent phosphoproteomic analysis of two representative sensitive and resistant lines led to the reproducible identification of 7,800 proteins and 13,000 phosphorylation sites (p-sites). Approximately 700 p-sites on 400 proteins showed abundance changes after radiation in all cell lines regardless of their phenotypic sensitivity. Apart from recapitulating known radiation response phosphorylation markers such as on proteins involved in DNA damage repair, the analysis uncovered many novel members of a radiation-responsive signaling network that was apparent only at the level of protein phosphorylation. These regulated p-sites were enriched in potential ATM substrates and in vitro kinase assays corroborated 10 of these. Comparing the proteomes and phosphoproteomes of radiosensitive and -resistant cells pointed to additional tractable radioresistance mechanisms involving apoptotic proteins. For instance, elevated NADPH quinine oxidoreductase 1 (NQO1) expression in radioresistant cells may aid in clearing harmful reactive oxygen species. Resistant cells also showed elevated phosphorylation levels of proteins involved in cytoskeleton organization including actin dynamics and focal adhesion kinase (FAK) activity and one resistant cell line showed a strong migration phenotype. Pharmacological inhibition of the kinases FAK by Defactinib and of CHEK1 by Rabusertib showed a statistically significant sensitization to radiation in radioresistant PDAC cells. Together, the presented data map a comprehensive molecular network of radiation-induced signaling, improves the understanding of radioresistance and provides avenues for developing radiotherapeutic strategies. The profiling of gene expression during spermatogenesis has been performed in previous studies, yet few studies have aimed to find new functional genes. Since there is a huge gap between the number of genes that are able to be quantified and the number of genes that can be characterized by phenotype screening in one assay, an efficient method to rank quantified genes according to phenotypic relevance is of great importance. We proposed to rank genes by the probability of their function in mammalian meiosis based on global protein abundance using machine learning. Here, nine types of germ cells focusing on continual substages of meiosis prophase I were isolated, and the corresponding proteomes were quantified by high-resolution mass spectrometry. By combining meiotic labels annotated from the MGI mouse knockout database and the spermatogenesis proteomics dataset, a supervised machine learning package, FuncProFinder, was developed to rank meiosis-essential candidates. Of the candidates whose functions were unannotated, four of ten genes with the top prediction scores, Zcwpw1, Tesmin, 1700102P08Rik and Kctd19, were validated as meiosis-essential genes by knockout mouse models. Therefore, mammalian meiosis-essential genes could be efficiently predicted based on the protein abundance dataset, which provides a paradigm for other functional gene mining from a related abundance dataset.
