OSCE facilitates consolidation workshop on Regulatory Reform Roadmap for Armenia

The finalization of a Regulatory Reform Roadmap for Armenia was the focus of a workshop co-organized by the OSCE Office for Democratic Institutions and Human Rights (OSCE/ODIHR), the Ministry of Justice of Armenia and the OSCE Office in Yerevan on 27 and 28 May 2016 in Tsaghkadzor, Armenia.

The workshop provided an opportunity for participants from the executive branch, the legislature, civil society and international organizations to exchange views on how to improve the overall process of lawmaking in the country. Topics discussed included policy-making and legislative planning, public consultations and regulatory impact assessment, as well as monitoring and evaluation of legislation.

The purpose of the workshop was to discuss recommendations for reform from previous thematic lawmaking workshops, conducted in Armenia in 2015, and from OSCE/ODIHR’s 2014 report on the “Comprehensive Assessment of the Legislative Procedure in Armenia”. These recommendations were consolidated in a draft Regulatory Reform Roadmap with concrete action points in early 2016, and this draft provided a basis for workshop discussions.

"The recommendations in the draft Regulatory Reform Roadmap focus on identifying workable solutions and were developed by key Armenian stakeholders during a series of discussions," said Alice Thomas, Chief of ODIHR’s Legislative Support Unit. "The draft Roadmap thus constitutes a home-grown plan for action to address weaknesses identified in the Armenian lawmaking system during discussions at our 2015 workshops."

Arsen Mkrtchyan, First Deputy Minister of Justice of Armenia, said: "We have already begun initiating reforms as indicated in the draft Roadmap. This workshop will help us finalize this document, which will be very useful in helping us plan and implement additional reform processes to make our legislative process less burdensome and more planned, transparent, and participatory."

The consolidating workshop begins the final stage of the process of drafting a Regulatory Reform Roadmap for Armenia, as envisaged by the Memorandum of Understanding signed between OSCE/ODIHR and the Ministry of Justice in 2014. Once ready, the Roadmap will form the basis for future reforms of the legislative process in the country.

Unelected Power: Finding Legitimacy in Central Banking and the Regulatory State

A novel stress-inducible CmtR-ESX3-Zn2+ regulatory pathway essential for survival of Mycobacterium bovis under oxidative stress [Microbiology]

Reactive oxygen species (ROS) are an unavoidable host environmental cue for intracellular pathogens such as Mycobacterium tuberculosis and Mycobacterium bovis; however, the signaling pathway in mycobacteria for sensing and responding to environmental stress remains largely unclear. Here, we characterize a novel CmtR-Zur-ESX3-Zn2+ regulatory pathway in M. bovis that aids mycobacterial survival under oxidative stress. We demonstrate that CmtR functions as a novel redox sensor and that its expression can be significantly induced under H2O2 stress. CmtR can physically interact with the negative regulator Zur and de-represses the expression of the esx-3 operon, which leads to Zn2+ accumulation and promotion of reactive oxygen species detoxication in mycobacterial cells. Zn2+ can also act as an effector molecule of the CmtR regulator, using which the latter can de-repress its own expression for further inducing bacterial antioxidant adaptation. Consistently, CmtR can induce the expression of EsxH, a component of esx-3 operon involved in Zn2+ transportation that has been reported earlier, and inhibit phagosome maturation in macrophages. Lastly, CmtR significantly contributes to bacterial survival in macrophages and in the lungs of infected mice. Our findings reveal the existence of an antioxidant regulatory pathway in mycobacteria and provide novel information on stress-triggered gene regulation and its association with host–pathogen interaction.

The heptameric structure of the flagellar regulatory protein FlrC is indispensable for ATPase activity and disassembled by cyclic-di-GMP [Protein Structure and Folding]

The bacterial enhancer-binding protein (bEBP) FlrC, controls motility and colonization of Vibrio cholerae by regulating the transcription of class-III flagellar genes in σ54-dependent manner. However, the mechanism by which FlrC regulates transcription is not fully elucidated. Although, most bEBPs require nucleotides to stimulate the oligomerization necessary for function, our previous study showed that the central domain of FlrC (FlrCC) forms heptamer in a nucleotide-independent manner. Furthermore, heptameric FlrCC binds ATP in “cis-mediated” style without any contribution from sensor I motif 285REDXXYR291 of the trans protomer. This atypical ATP binding raises the question of whether heptamerization of FlrC is solely required for transcription regulation, or if it is also critical for ATPase activity. ATPase assays and size exclusion chromatography of the trans-variants FlrCC-Y290A and FlrCC-R291A showed destabilization of heptameric assembly with concomitant abrogation of ATPase activity. Crystal structures showed that in the cis-variant FlrCC-R349A drastic shift of Walker A encroached ATP-binding site, whereas the site remained occupied by ADP in FlrCC-Y290A. We postulated that FlrCC heptamerizes through concentration-dependent cooperativity for maximal ATPase activity and upon heptamerization, packing of trans-acting Tyr290 against cis-acting Arg349 compels Arg349 to maintain proper conformation of Walker A. Finally, a Trp quenching study revealed binding of cyclic-di-GMP with FlrCC. Excess cyclic-di-GMP repressed ATPase activity of FlrCC through destabilization of heptameric assembly, especially at low concentration of protein. Systematic phylogenetic analysis allowed us to propose similar regulatory mechanisms for FlrCs of several Vibrio species and a set of monotrichous Gram-negative bacteria.
