Inhibition of glycosphingolipid biosynthesis reverts multidrug resistance by differentially modulating ABC transporters in chronic myeloid leukemias [Cell Biology]

Multidrug resistance (MDR) in cancer arises from cross-resistance to structurally- and functionally-divergent chemotherapeutic drugs. In particular, MDR is characterized by increased expression and activity of ATP-binding cassette (ABC) superfamily transporters. Sphingolipids are substrates of ABC proteins in cell signaling, membrane biosynthesis, and inflammation, for example, and their products can favor cancer progression. Glucosylceramide (GlcCer) is a ubiquitous glycosphingolipid (GSL) generated by glucosylceramide synthase, a key regulatory enzyme encoded by the UDP-glucose ceramide glucosyltransferase (UGCG) gene. Stressed cells increase de novo biosynthesis of ceramides, which return to sub-toxic levels after UGCG mediates incorporation into GlcCer. Given that cancer cells seem to mobilize UGCG and have increased GSL content for ceramide clearance, which ultimately contributes to chemotherapy failure, here we investigated how inhibition of GSL biosynthesis affects the MDR phenotype of chronic myeloid leukemias. We found that MDR is associated with higher UGCG expression and with a complex GSL profile. UGCG inhibition with the ceramide analog d-threo-1-(3,4,-ethylenedioxy)phenyl-2-palmitoylamino-3-pyrrolidino-1-propanol (EtDO-P4) greatly reduced GSL and monosialotetrahexosylganglioside levels, and co-treatment with standard chemotherapeutics sensitized cells to mitochondrial membrane potential loss and apoptosis. ABC subfamily B member 1 (ABCB1) expression was reduced, and ABCC-mediated efflux activity was modulated by competition with nonglycosylated ceramides. Consistently, inhibition of ABCC-mediated transport reduced the efflux of exogenous C6-ceramide. Overall, UGCG inhibition impaired the malignant glycophenotype of MDR leukemias, which typically overcomes drug resistance through distinct mechanisms. This work sheds light on the involvement of GSL in chemotherapy failure, and its findings suggest that targeted GSL modulation could help manage MDR leukemias.

Our R&D team focused on performance improvements in the 3.2 release, which was shipped last August.  Based on our suite of performance benchmarks, we achieved an overall 10x performance boost.  This comes from two fundamental improvements: an overall 3.5x boost in single processor performance, and an overall 2.8x performance boost from using four CPUs in a multiprocessor configuration. 

Your mileage may vary, of course.  Our test suite includes a variety of designs and processes - LVS testcases as well as DRC.  We typically noticed the most significant performance improvement on large designs that previously ran for many hours. 

This is a maintenance upgrade from the previous release, so there's no risk if you want to download the latest version just to kick the tires.  It will probably save you some time.

Darrell Issa Appears to Flee to Building Roof to Avoid Protesters

Rep. Darrell Issa (R-Calif.) was seen taking refuge on the roof of his office building in Vista, California, Tuesday, taking photos of angry constituents who had gathered below to protest the congressman's voting record. The incident comes before a much-anticipated town hall meeting this Saturday at San Juan Hills High School, where the nine-term congressman is expected to face a hostile crowd because of his support for various Trump administration policies, including the Republican plan to repeal and replace Obamacare.

Democrat Mike Levin, an environmental lawyer who recently announced his bid to challenge Issa in 2018, shared an image of the congressman appearing to avoid demonstrators on social media, where it was roundly mocked.

Others saw his retreating to a rooftop as reminiscent of Michael Scott, Steve Carrell's character in The Office who memorably took to the roof in the episode titled "Safety Training."

Mira Rajput tries her hand at embroidery with Misha, 'Little hands made the heart with a lot of excitement'

Shahid Kapoor and Mira Rajput tied the knot on July 7, 2015, in a private ceremony and since then their love has grown stronger. Mira is the daughter of Delhi-based businessman Vikramaditya Rajput and Bela Rajput. Shahid Kapoor and Mira Rajput became proud parents to baby girl Misha on August 26, 2016. Shahid Kapoor and Mira Rajput welcomed their second child, a baby boy on September 5, 2018, and the duo named their new-born Zain Kapoor.

Misha and Zain are one of the most adorable star kids of B-town. The cute little kiddos are the apple of everyone's eye. Just like other star babies, both the kids, particularly Misha has several fan clubs dedicated to her. Shahid and Mira often treat their fans with lovey-dovey pictures of their kids on Instagram.

During the coronavirus lockdown, Mira tried her hands at embroidery and even took some help from daughter Misha. Mira shared a picture of an embroidered heart and wrote, "Big love with my little love. With a hand drawn pattern and help on the turns, little hands made the heart with a lot of excitement and filled with a whole lot of love!" Take a look at the picture here:

The star wife keeps sharing snippets of her family quarantine time on her Instagram profile. She recently gave a shoutout to everyone for staying indoors. She wrote an elaborate open letter which read, "Love them. Hug them. Ask them how they feel and let them cry if they're not okay. Kiss them. Paint with them and let their clothes and the walls get dirty. Watch their favourite cartoon with them. Snuggle up with them. Give them extra ketchup if they ask for it that day. The Sandeep Reddy Vanga directorial was the story of a brilliant doctor who takes the self-destructive path when the love of his life is forced to marry another man. It earned over Rs 250 crore and became one of the highest grosser of 2019.

As of now, Shahid has signed the Hindi remake of the Telugu film Jersey, which is the story of a cricketer and how he battles the politics within the game and his inner demons. The film is all set to release on August 28, 2020.
