Curing HIV infection will require the elimination of a reservoir of infected CD4+ T cells that persists despite HIV-specific cytotoxic T cell (CTL) responses. Although viral latency is a critical factor in this persistence, recent evidence also suggests a role for intrinsic resistance of reservoir-harboring cells to CTL killing. This resistance may have contributed to negative outcomes of clinical trials, where pharmacologic latency reversal has thus far failed to drive reductions in HIV reservoirs. Through transcriptional profiling, we herein identified overexpression of the prosurvival factor B cell lymphoma 2 (BCL-2) as a distinguishing feature of CD4+ T cells that survived CTL killing. We show that the inducible HIV reservoir was disproportionately present in BCL-2hi subsets in ex vivo CD4+ T cells. Treatment with the BCL-2 antagonist ABT-199 was not sufficient to drive reductions in ex vivo viral reservoirs when tested either alone or with a latency-reversing agent (LRA). However, the triple combination of strong LRAs, HIV-specific T cells, and a BCL-2 antagonist uniquely enabled the depletion of ex vivo viral reservoirs. Our results provide rationale for novel therapeutic approaches targeting HIV cure and, more generally, suggest consideration of BCL-2 antagonism as a means of enhancing CTL immunotherapy in other settings, such as cancer.

An updated Disaster Resilience Scorecard to help cities and local government agencies improve their preparedness and reduce risks from disasters was launched today at the Global Platform for Disaster Risk Reduction. There was a time – before I sank my teeth into the board game hobby and before I took on the demands of being a stepmom to three children - that I planned frequent trips for my friends circle. Each month would see us jet setting off to a different part of the United States (or a different part of the world) in search of adventure and great cuisine. I would pour myself into the research project that proper trip planning requires and create a custom itinerary for our group, working to ensure we caught all the highlights of our destination. This post is about the one I worked up for our last trip to some of the great Western National Parks in the United States.  I share it here with the hope that you might find the itinerary useful in planning your trip out west.

On March 31, the volunteer computing part of SETI@home will stop distributing work and will go into hibernation.

We're doing this for two reasons:

1) Scientifically, we're at the point of diminishing returns; basically, we've analyzed all the data we need for now.

2) It's a lot of work for us to manage the distributed processing of data. We need to focus on completing the back-end analysis of the results we already have, and writing this up in a scientific journal paper.

However, SETI@home is not disappearing. The web site and the message boards will continue to operate. We hope that other UC Berkeley astronomers will find uses for the huge computing capabilities of SETI@home for SETI or related areas like cosmology and pulsar research. If this happens, SETI@home will start distributing work again. We'll keep you posted about this.

If you're currently running SETI@home on your computer, we encourage you to attach to other BOINC-based projects as well. Or use Science United and sign up to do astronomy. You can stay attached to SETI@home, of course, but you won't get any jobs until we find new applications.

We're extremely grateful to all of our volunteers for supporting us in many ways during the past 20 years. Without you there would be no SETI@home. We're excited to finish up our original science project, and we look forward to what comes next.
