How they solve each problem is a function of their skill, talent, knowledge & time. The resultant solution will resolve the problem with greater or lesser complexity in the design. Thus, I find thinking about how (and more importantly where) complexity is handled, a useful way to evaluate a design. For this I use the time-honored technique of having three levels: Level Zero (Below this level, the problem is not solved) The programmer has provided a solution. However, the interface is a mine-field; click things in the wrong order, the program crashes. Or it works great, but you need to reread the documentation Every. Single. Time. Or the resultant surrounding workflow is a Rube Goldberg device. However it surfaces, the programmer has placed the complexity on the user. Level One The interface is well thought out and reasonably intuitive. Controls work as expected and the solution is robust - even bulletproof. The solution not only fits the larger context, it improves it. The code itself however, is hard to change (or even understand). It is not well-organized; Or is, but full of tricky, interleaved logic. Or over-engineered, over-patterned etc. Here, the programmer has placed the complexity on the maintainer.[1] Level Two The burden of managing complexity is on the current programmer. The person writing the code takes the time and employs the talent and skill necessary to find an elegant solution that reduces the code to the minimum complexity needed to solve the problem and does so in such a way that is understandable (and changeable) later: The user gains the benefits of a level 1 design, while maintainers are left with clean code to change.[2] Implications Level zero code is common; it's the default for new programmers and an uncomfortable amount of commercial software. The thing of it is: Too often this sort of code is derided (including by me) when there is both a de facto and a cost-benefit rationale for managing complexity this way. Short-term projects are one example; software written for one's own use is another. Level 1 software can be very valuable and enduring. I tend to equate this type of code with the Hacker ethic in all senses of that term. And I don't mean that pejoratively - we all use hacked together solutions every day and much of the world's technology infrastructure is built on it. It is a practical and stable design level. Having the skill (and the time) to write level 2 code is a rare and wonderful thing. Amidst the hyperbole, aspiring to be such a programmer is at the heart of the "Software as Craft" movement and is a worthwhile goal for anyone who aspires to be a professional programmer. I don't believe level 2 is inherently better than a level 1 (or level zero!) - it's about context - however, I think that for regularly edited and changed code (i.e. much IT software), this level of skill is what's implicitly expected (if not gotten) by the customer. Ironically, the time needed is often the first thing that goes as a non-technical customer can only assess code quality based on level 1 considerations and so pushes for faster results because everything "looks" OK. Only later - when their investment can't be changed without major overhaul - do they realize there is something wrong. And so the cycle continues... Summary This model is handy in several situations including: Judging the quality of an actual solution; choosing among different solutions to a given problem; estimating - and even when to stop refactoring. And while I don't believe that all code should be worked until it exhibits level 2 quality, it is what I want others to think of the code I leave behind. And it is what I hope to encounter in theirs. That said, being a successful professional programmer requires (among many other things) the ability to write all three levels of code, and the judgment to know when each is appropriate to use. [1] This may well be the same person who wrote the code. The essential characteristic of a level 1 design is that complexity is put off to future efforts, not the current one. Thus level 1 designs tend to be high in technical debt. [2]How the programmer achieves this is a matter of personal preference and technique. I do not subscribe to the notion that any particular set of programming practices provides this - or inhibit it by their absence. We then tie this definition to complexity, highlighting a number of important implications for the informing science transdiscipline. Background As informing science increasingly incorporates concepts of fitness and complexity in its research stream, rigorous discussion and definition of both terms is essential to effective communication. Methodology Our analysis consists principally of a synthesis of past work in the informing science field that incorporates concepts from evolutionary biology, economics and management. Contribution We provide a rigorous approach to defining fitness and introduce the construct “extrinsic complexity”, as a measure of the amount of information required to predict fitness, to more fully differentiate this form of complexity from other complexity constructs. We draw a number of conclusions regarding how behaviors under low and high extrinsic complexity will differ. Findings High extrinsic complexity environments are likely to produce behaviors that include resistance to change, imitation, turbulence and inequality. Recommendations for Practitioners As extrinsic complexity grows, effective search for problem solutions will increasingly dominate employing recommended solutions of “best practices”. Recommendation for Researchers As extrinsic complexity grows, research tools that rely on decomposing individual effects and hypothesis testing become increasingly unreliable. Impact on Society We raise concerns about society’s continuing investment in academic research that discounts the extrinsic complexity of the domains under study. Examples of these consequences are discussed at the individual level (e.g., deskilling, barriers to advancement), the task level (e.g., perpetuation of past practices), as well as broader consequences that may result from the need to function in an environment that is more extrinsically complex (e.g., erosion of predictable causality, shortened time horizons, inequality, tribalism). We conclude by reflecting on the implications of attempting to manage or limit increases of complexity. Contribution: Shows how many unintended consequences of IT could be attributed to growing complexity. Findings: We find that these three forms of complexity feed into one another resulting in a positive feedback loop that we term the Complexity Cycle. As examples, we analyze ChatGPT, blockchain and quantum computing, through the lens of the complexity cycle, speculating how experienced complexity can lead to greater intrinsic complexity in task performance through the incorporation of IT which, in turn, increases the extrinsic complexity of the economic/technological environment. Recommendations for Practitioners: Consider treating increasing task complexity as an externality that should be considered as new systems are developed and deployed. complexity The complexity and dynamics of the tissue glycoproteome associated with prostate cancer progression [Research] By www.mcponline.org Published On :: 2020-10-30T10:35:20-07:00 The complexity and dynamics of the immensely heterogeneous glycoproteome of the prostate cancer (PCa) tumour micro-environment remain incompletely mapped, a knowledge gap that impedes our molecular-level understanding of the disease. To this end, we have used sensitive glycomics and glycoproteomics to map the protein-, cell- and tumour grade-specific N- and O-glycosylation in surgically-removed PCa tissues spanning five histological grades (n = 10/grade) and tissues from patients with benign prostatic hyperplasia (n = 5). Quantitative glycomics revealed PCa grade-specific alterations of the oligomannosidic-, paucimannosidic- and branched sialylated complex-type N-glycans, and dynamic remodelling of the sialylated core 1- and core 2-type O-glycome. Deep quantitative glycoproteomics identified ~7,400 unique N-glycopeptides from 500 N-glycoproteins and ~500 unique O-glycopeptides from nearly 200 O-glycoproteins. With reference to a recent Tissue and Blood Atlas, our data indicate that paucimannosidic glycans of the PCa tissues arise mainly from immune cell-derived glycoproteins. Further, the grade-specific PCa glycosylation arises primarily from dynamics in the cellular makeup of the PCa tumour microenvironment across grades involving increased oligomannosylation of prostate-derived glycoproteins and decreased bisecting GlcNAcylation of N-glycans carried by the extracellular matrix proteins. Further, elevated expression of several oligosaccharyltransferase subunits and enhanced N-glycoprotein site occupancy were observed associated with PCa progression. Finally, correlations between the protein-specific glycosylation and PCa progression were observed including increased site-specific core 2-type O-glycosylation of collagen VI. In conclusion, integrated glycomics and glycoproteomics have enabled new insight into the complexity and dynamics of the tissue glycoproteome associated with PCa progression generating an important resource to explore the underpinning disease mechanisms. We tackle these questions by first creating a pipeline to assess matched long- and short-read data using a variety of transcriptome statistics. We find that across data sets, algorithms, and technologies, matched short-read data detects ~30% more splice junctions, such that ~10%–30% of the splice junctions included at ≥20% by short reads are missed by long reads. In contrast, long reads detect many more intron-retention events and can detect full isoforms, pointing to the benefit of combining the technologies. We introduce MAJIQ-L, an extension of the MAJIQ software, to enable a unified view of transcriptome variations from both technologies and demonstrate its benefits. Our software can be used to assess any future long-read technology or algorithm and can be combined with short-read data for improved transcriptome analysis. Full Article

The crystal structure is based on a 3D titanosilicate framework consisting of trigonal prismatic titanosilicate (TPTS) clusters centered by Ce3+ in [9]-coordination. Four adjacent TPTS clusters are linked into four-membered rings within the (010) plane and connected via ribbons parallel to 101. The ribbons are organized into layers parallel to (010) and modulated along the a axis with a modulation wavelength of csinβ = 32.91 Å and an amplitude of ∼b/2 = 13.89 Å. The layers are linked by additional silicate tetrahedra. Na+, K+, Ba2+ and H2O groups occur in the framework cavities and have different occupancies and coordination environments. The crystal structure of ilmajokite can be separated into eight hierarchical levels: atoms, coordination polyhedra, TPTS clusters, rings, ribbons, layers, the framework and the whole structure. The information-based analysis allows estimation of the complexity of the structure as 8.468 bits per atom and 11990.129 bits per cell. According to this analysis, ilmajokite is the third-most complex mineral known to date after ewingite and morrisonite, and is the most complex mineral framework structure, comparable in complexity to paulingite-(Ca) (11 590.532 bits per cell).
