lms The N-Acetylglucosaminidase LytB of Streptococcus pneumoniae Is Involved in the Structure and Formation of Biofilms [Genetics and Molecular Biology] By aem.asm.org Published On :: 2020-05-05T08:00:35-07:00 The N-acetylglucosaminidase LytB of Streptococcus pneumoniae is involved in nasopharyngeal colonization and is responsible for cell separation at the end of cell division; thus, lytB mutants form long chains of cells. This paper reports the construction and properties of a defective pneumococcal mutant producing an inactive LytB protein (LytBE585A). It is shown that an enzymatically active LytB is required for in vitro biofilm formation, as lytB mutants (either lytB or producing the inactive LytBE585A) are incapable of forming substantial biofilms, despite that extracellular DNA is present in the biofilm matrix. Adding small amounts (0.5 to 2.0 μg/ml) of exogenous LytB or some LytB constructs restored the biofilm-forming capacity of lytB mutants to wild-type levels. The LytBE585A mutant formed biofilm more rapidly than lytB mutants in the presence of LytB. This suggests that the mutant protein acted in a structural role, likely through the formation of complexes with extracellular DNA. The chain-dispersing capacity of LytB allowed the separation of daughter cells, presumably facilitating the formation of microcolonies and, finally, of biofilms. A role for the possible involvement of LytB in the synthesis of the extracellular polysaccharide component of the biofilm matrix is also discussed. IMPORTANCE It has been previously accepted that biofilm formation in S. pneumoniae must be a multigenic trait because the mutation of a single gene has led to only to partial inhibition of biofilm production. In the present study, however, evidence that the N-acetylglucosaminidase LytB is crucial in biofilm formation is provided. Despite the presence of extracellular DNA, strains either deficient in LytB or producing a defective LytB enzyme formed only shallow biofilms. Full Article
lms Inactivation of Pseudomonas aeruginosa Biofilms by 405-Nanometer-Light-Emitting Diode Illumination [Physiology] By aem.asm.org Published On :: 2020-05-05T08:00:35-07:00 Biofilm formation by Pseudomonas aeruginosa contributes to its survival on surfaces and represents a major clinical threat because of the increased tolerance of biofilms to disinfecting agents. This study aimed to investigate the efficacy of 405-nm light-emitting diode (LED) illumination in eliminating P. aeruginosa biofilms formed on stainless steel coupons under different temperatures. Time-dependent killing assays using planktonic and biofilm cells were used to determine the antimicrobial and antibiofilm activities of LED illumination. We also evaluated the effects of LED illumination on the disinfectant susceptibility, biofilm structure, extracellular polymeric substance (EPS) structure and composition, and biofilm-related gene expression of P. aeruginosa biofilm cells. Results showed that the abundance of planktonic P. aeruginosa cells was reduced by 0.88, 0.53, and 0.85 log CFU/ml following LED treatment for 2 h compared with untreated controls at 4, 10, and 25°C, respectively. For cells in biofilms, significant reductions (1.73, 1.59, and 1.68 log CFU/cm2) were observed following LED illumination for 2 h at 4, 10, and 25°C, respectively. Moreover, illuminated P. aeruginosa biofilm cells were more sensitive to benzalkonium chloride or chlorhexidine than untreated cells. Scanning electron microscopy and confocal laser scanning microscopic observation indicated that both the biofilm structure and EPS structure were disrupted by LED illumination. Further, reverse transcription-quantitative PCR revealed that LED illumination downregulated the transcription of several genes associated with biofilm formation. These findings suggest that LED illumination has the potential to be developed as an alternative method for prevention and control of P. aeruginosa biofilm contamination. IMPORTANCE Pseudomonas aeruginosa can form biofilms on medical implants, industrial equipment, and domestic surfaces, contributing to high morbidity and mortality rates. This study examined the antibiofilm activity of 405-nm light-emitting diode (LED) illumination against mature biofilms formed on stainless steel coupons. We found that the disinfectant susceptibility, biofilm structure, and extracellular polymeric substance structure and composition were disrupted by LED illumination. We then investigated the transcription of several critical P. aeruginosa biofilm-related genes and analyzed the effect of illumination temperature on the above characteristics. Our results confirmed that LED illumination could be developed into an effective and safe method to counter P. aeruginosa biofilm contamination. Further research will be focused on the efficacy and application of LED illumination for elimination of complicated biofilms in the environment. Full Article

With current therapies, the 5-yr overall survival (OS) for children with unilateral favorable histology WT is greater than 85%. The prognosis is worse, however, for the roughly 15% of patients who relapse, with only 50%–80% OS reported in those with recurrence. Herein, we describe the extended and detailed clinical course of a rare case of a child with recurrent, pulmonary metastatic, favorable histology WT harboring a BRAF V600E mutation. The BRAF V600E mutation, commonly found in melanoma and other cancers, and previously undescribed in WT, has recently been reported by our group in a subset of epithelial-predominant WT. This patient, who was included in that series, presented with unilateral, stage 1, favorable histology WT and was treated with standard chemotherapy. Following the completion of therapy, the patient relapsed with pulmonary metastatic disease, that then again recurred despite an initial response to salvage chemotherapy and radiation. Next-generation sequencing (NGS) on the metastatic pulmonary nodule revealed a BRAF V600E mutation. After weighing the therapeutic options, a novel approach with dual BRAF/MEK inhibitor combination therapy was initiated. Complete radiographic response was observed following 4 months of therapy with dabrafenib and trametinib. At 12 months following the start of BRAF/MEK combination treatment, the patient continues with a complete response and has experienced minimal treatment-related side effects. This represents the first case, to our knowledge, of effective treatment with BRAF/MEK molecularly targeted therapy in a pediatric Wilms tumor patient. Full Article

It was the first film screening in the city organised by SteppinOut Movie Nights, which has presence across Chennai, Delhi and Bangalore. Ojal Kulkarni, the brand head for the firm, says they chose Novotel for its cool vibe and plan a movie night at the venue every two months. "Yes, I am picky when it comes to choosing films. I try to be part of films that I would want to watch. I also try to pick different roles from what I have done. I started with '1920'. I am very fortunate, the actresses in our industry for years don't get a chance to do what I got to do in my debut," Adah told IANS. Adah made her Bollywood debut 2008 with the film "1920". She was later seen in films like "Hum Hai Raahi Car Ke", the "Commando" franchise, "Bypass Road" and "Phhir". She also worked in the southern film industry. Adah says she likes to surprise the audience. "I try with each film to do something different because the audience also waits to see 'what different is Adah Sharma going to do' and if I do the same then they are like 'anyone can do that!'. So nowadays even if it is a short film like I just did 'Tindey', 'moh' or a music video... I try to pick something that is different from what I have done and others have done as well," she said. The film revolves around actor Naveen Kasturia's character falling in love with and marrying Sharma's character, only to realise she was biologically a man who became a woman after surgery. Portraying as a thriller, the video shows a man being chased by a masked man dressing like a ghost. As the man reaches at the doorstep of his house, he stops to remove his shoes and socks before stepping in, which customary in Indian households. "I loved shooting in India. The people were incredible. I didn't realise how popular the Marvel films were in India. So that was, at times, overwhelming," Hemsworth told IANS during a special video call before the release of his Netflix movie. "There was such positivity and enthusiasm for all of us and our crew… During shooting, we had hundreds and thousands of people on certain days lining up on the streets and up on top of bridges and in buildings, you know, after every takes, cheering and applauding. I've never experienced anything like it. And as I said, I have such fond memories of India and the people there," he added while recounting his time in India. Hemsworth shot for the Netflix film in India in 2018. He shot in Ahmedabad and Mumbai. The "Thor" star was scheduled to be in Mumbai on March 16 to promote the film that has been shot in India. The promotional tour was called off due to the global coronavirus outbreak. Earlier in an interview to IANS, Hemsworth, whose daughter is named India, revealed the reason behind naming his daughter after the country. "My wife (Elsa Pataky) spent a lot of time in India and that was where the name originally came from," Hemsworth said in that interview, adding: "I love the place and the people. It was sort of intimidating as it was exciting because there were so many people… We felt like rock stars in a stadium. But the warmth and support we got while shooting there, no matter how disruptive we were to their daily lives, it was so good. " Originally titled "Dhaka", the film also stars Indian actors Randeep Hooda, Pankaj Tripathi, Priyanshu Painyulli, and Rudraksh Jaiswal. It revolves around Tyler Rake (essayed by Hemsworth), a fearless, black market mercenary, who embarks on the most deadly extraction of his career when he is enlisted to rescue the kidnapped son of an imprisoned international crime lord. The film, directed by Sam Hargrave, will release on Netflix on April 24. Talking about working for a streaming platform for the first time, Hemsworth said: "It's my first time working on a streaming platform. I was excited to do it, strangely enough, you know, it's probably the best place to be at this point. "We put in, you know, the same amount of commitment and passion and enthusiasm as we would with anything else, whether it was a cinema release or a streaming space. I am beyond proud of the film. From the very beginning, I loved the script. I love the story that we were able to tell and the opportunity to shoot action, unlike anything I'd certainly been a part of before." Talking about his upcoming Netflix action film, Hargrave told IANS: "It seems that there's not as much space in the cinemas for these kinds of mid-budget action movies. It seems that it's mostly the super big-budget superhero movies or the kind of the smaller stuff." Hargrave is glad that his first movie will reach a wider audience through Netflix. "Netflix has carved out space for itself for movies like these to exist, which I think is really great… Because there are a lot of great stories out there… These kinds of movies, now, have a place to be seen, be viewed and be enjoyed," he said. With "Extraction", Hargrave is graduating from being Chris Evans' stunt double in "Captain America: The Winter Soldier" for fight scenes and stunt coordinator in "Captain America: Civil War" to a feature director. Hargrave was an additional second unit director in "Avengers: Infinity War". Hargrave continued: "I think that's a really great thing for audiences around the world. I'm happy that my first time directing (feature film) come out on a platform like Netflix where so many people can see this.
