Probert's "Hat"


I have neglected to post about my decision to start muzzling Probert when he is with "the pack" because so many people react so negatively to it, but I'm ready so here it is. I have posted a few photos of Probert in his "hat" (we don't use the "m" word) on my personal Facebook page and have gotten some very mixed reactions. I understand that it is sometimes difficult to see a dog in a muzzle because it makes them "look mean" or for any other reason. For the first month after I bought Probert's muzzle, I couldn't even put it on him for any amount of time, but I finally dove in and now I am very used to seeing it on him and know that underneath, he is still my sweet little guy.

Probert has a history of redirecting in stressful situations (in the yard, something outside the fence usually) and biting Wrigley. He has done Wrigley some serious damage and in the interest of preventing that from happening (on the occasion when it does happen) and also in not isolating Probert from the pack, I bought him a muzzle. Since buying it several months ago, there have been two incidents when it has prevented him from doing any damage when attempting to bite Wrigley and the rest of the time, it allows him to interact with the pack like a normal dog and without my worrying about what "could" happen - so it's doing its job.

So, despite the "scary" exterior appearance of the muzzle, I have to say that it's one of the better decisions I have made for the health and happiness of all of my dogs. In the interest of looking a little less "scary" (and preventing further wall-gouging and painful leg-bashing from the heavy metal one) I just ordered Probert a new JAFCO clear flexi vinyl muzzle, so we'll see how that one works when it gets here. If anyone is interested, I will be happy to report back! In the meantime, please don't be too quick to judge a dog (or his person) by his hat!

Tracking isotopically labeled oxidants using boronate-based redox probes [Methods and Resources]

Reactive oxygen and nitrogen species have been implicated in many biological processes and diseases, including immune responses, cardiovascular dysfunction, neurodegeneration, and cancer. These chemical species are short-lived in biological settings, and detecting them in these conditions and diseases requires the use of molecular probes that form stable, easily detectable, products. The chemical mechanisms and limitations of many of the currently used probes are not well-understood, hampering their effective applications. Boronates have emerged as a class of probes for the detection of nucleophilic two-electron oxidants. Here, we report the results of an oxygen-18–labeling MS study to identify the origin of oxygen atoms in the oxidation products of phenylboronate targeted to mitochondria. We demonstrate that boronate oxidation by hydrogen peroxide, peroxymonocarbonate, hypochlorite, or peroxynitrite involves the incorporation of oxygen atoms from these oxidants. We therefore conclude that boronates can be used as probes to track isotopically labeled oxidants. This suggests that the detection of specific products formed from these redox probes could enable precise identification of oxidants formed in biological systems. We discuss the implications of these results for understanding the mechanism of conversion of the boronate-based redox probes to oxidant-specific products.

The Government has mechanisms to investigate complaints against civil servants impartially, Secretary for the Civil Service Patrick Nip said today.

 

Mr Nip made the remarks in response to media queries on recent allegations levelled against senior police officers.

 

He said: “There are established mechanisms to investigate every complaint in accordance with the procedures and rules. Depending on the circumstances and the case details, some are being handled by departments and bureaus. Some may be handled by the Civil Service Bureau.

 

“There are established mechanisms in dealing with that. Of course we would handle each and every complaint in a very serious and impartial manner.”

Chief Executive Carrie Lam today said law enforcement agencies are investigating cases where police officers are suspected of breaching property rules and will give a full account to society.

 

Speaking to reporters before the Executive Council meeting this morning, Mrs Lam said nobody is above the law.

 

“The law will be applied in the same manner regardless of the status, the background, the political affiliation of that particular person.

 

“As far as I am concerned, as the Chief Executive of Hong Kong and also as an individual, no law-breaking behaviour is acceptable. But it is not for me to stand here to judge each and every case because every case or every complaint has to be investigated and analysed by the law enforcement agencies.

 

“I am sure that they will do it as diligently as possible and will give a full account to society, especially given the recent concerns.”

Initial Clinical Results of a Novel Immuno-PET Theranostic Probe in HER2-negative Breast Cancer

Purpose: This prospective study evaluated the imaging performance of a novel immunological pretargeting positron-emission tomorgraphy (immuno-PET) method in patients with HER2-negative, carcinoembryonic antigen (CEA)-positive, metastatic breast cancer (BC), compared to computed tomography (CT), bone magnetic resonance imaging (MRI), and 18Fluorodeoxyglucose PET (FDG-PET). Patients and Methods: Twenty-three patients underwent whole-body immuno-PET after injection of 150 MBq 68Ga-IMP288, a histamine-succinyl-glycine peptide given following initial targeting of a trivalent anti-CEA, bispecific, anti-peptide antibody. The gold standards were histology and imaging follow-up. Tumor standard uptake values (SUVmax and SUVmean) were measured, and tumor burden analyzed using Total Tumor Volume (TTV) and Total Lesion Activity (TLA). Results: Total lesion sensitivity of immuno-PET and FDG-PET was 94.7% (1116/1178) and 89.6% (1056/1178), respectively. Immuno-PET had a somewhat higher sensitivity than CT and FDG-PET in lymph nodes (92.4% vs 69.7% and 89.4%, respectively) and liver metastases (97.3% vs 92.1% and 94.8%, respectively), whereas sensitivity was lower for lung metastases (48.3% vs 100% and 75.9%, respectively). Immuno-PET showed higher sensitivity than MRI and FDG-PET for bone lesions (95.8% vs 90.7% and 89.3%, respectively). In contrast to FDG-PET, immuno-PET disclosed brain metastases. Despite equivalent tumor SUVmax, SUVmean, and TTV, TLA was significantly higher with immuno-PET compared to FDG PET (P = 0.009). Conclusion: Immuno-PET using anti-CEA/anti-IMP288 bispecific antibody, followed by 68Ga-IMP288, is a potentially sensitive theranostic imaging method for HER2-negative, CEA-positive, metastatic BC patients, and warrants further research.
