X-ray fluorescence analysis of metal distributions in cryogenic biological samples using large-acceptance-angle SDD detection and continuous scanning at the Hard X-ray Micro/Nano-Probe beamline P06 at PETRA III

A new Rococo 2 X-ray fluorescence detector was implemented into the cryogenic sample environment at the Hard X-ray Micro/Nano-Probe beamline P06 at PETRA III, DESY, Hamburg, Germany. A four sensor-field cloverleaf design is optimized for the investigation of planar samples and operates in a backscattering geometry resulting in a large solid angle of up to 1.1 steradian. The detector, coupled with the Xspress 3 pulse processor, enables measurements at high count rates of up to 106 counts per second per sensor. The measured energy resolution of ∼129 eV (Mn Kα at 10000 counts s−1) is only minimally impaired at the highest count rates. The resulting high detection sensitivity allows for an accurate determination of trace element distributions such as in thin frozen hydrated biological specimens. First proof-of-principle measurements using continuous-movement 2D scans of frozen hydrated HeLa cells as a model system are reported to demonstrate the potential of the new detection system.

XUV-driven plasma switch for THz: new spatio-temporal overlap tool for XUV–THz pump–probe experiments at FELs

A simple and robust tool for spatio-temporal overlap of THz and XUV pulses in in-vacuum pump–probe experiments is presented. The technique exploits ultrafast changes of the optical properties in semiconductors (i.e. silicon) driven by ultrashort XUV pulses that are probed by THz pulses. This work demonstrates that this tool can be used for a large range of XUV fluences that are significantly lower than when probing by visible and near-infrared pulses. This tool is mainly targeted at emerging X-ray free-electron laser facilities, but can be utilized also at table-top high-harmonics sources.

Picosecond pump–probe X-ray scattering at the Elettra SAXS beamline

A new setup for picosecond pump–probe X-ray scattering at the Austrian SAXS beamline at Elettra-Sincrotrone Trieste is presented. A high-power/high-repetion-rate laser has been installed on-site, delivering UV/VIS/IR femto­second-pulses in-sync with the storage ring. Data acquisition is achieved by gating a multi-panel detector, capable of discriminating the single X-ray pulse in the dark-gap of the Elettra hybrid filling mode. Specific aspects of laser- and detection-synchronization, on-line beam steering as well protocols for spatial and temporal overlap of laser and X-ray beam are also described. The capabilities of the setup are demonstrated by studying transient heat-transfer in an In/Al/GaAs superlattice structure and results are confirmed by theoretical calculations.

Correlative vibrational spectroscopy and 2D X-ray diffraction to probe the mineralization of bone in phosphate-deficient mice

Bone crystallite chemistry and structure change during bone maturation. However, these properties of bone can also be affected by limited uptake of the chemical constituents of the mineral by the animal. This makes probing the effect of bone-mineralization-related diseases a complicated task. Here it is shown that the combination of vibrational spectroscopy with two-dimensional X-ray diffraction can provide unparalleled information on the changes in bone chemistry and structure associated with different bone pathologies (phosphate deficiency) and/or health conditions (pregnancy, lactation). Using a synergistic analytical approach, it was possible to trace the effect that changes in the remodelling regime have on the bone mineral chemistry and structure in normal and mineral-deficient (hypophosphatemic) mice. The results indicate that hypophosphatemic mice have increased bone remodelling, increased carbonate content and decreased crystallinity of the bone mineral, as well as increased misalignment of crystallites within the bone tissue. Pregnant and lactating mice that are normal and hypophosphatemic showed changes in the chemistry and misalignment of the apatite crystals that can be related to changes in remodelling rates associated with different calcium demand during pregnancy and lactation.

Attorneys for Rick Bright, the government scientist who said he had been reassigned and subsequently filed a whistleblower complaint, say a government watchdog agrees that he should be reinstated to his post.

Bright was serving as director of the Biomedical Advanced Research and Development Authority, which is working on a vaccine to combat the coronavirus.

He said he was ousted from the position last month because he wanted to spend money on safe and vetted treatments for COVID-19 — not on ones without "scientific merit," such as hydroxychloroquine, the anti-malarial drug that President Trump and others had been touting.

Trump on Wednesday called Bright "a disgruntled employee who's trying to help the Democrats win an election."

Bright's attorneys say that the Office of Special Counsel, which hears whistleblower cases, determined there were "reasonable grounds" to believe that his removal was retaliatory and therefore prohibited.

Bright's attorneys say OSC plans to contact the Department of Health and Human Services to request that it put Bright's removal on hold for 45 days so the office can complete its investigation into the allegations.

The OSC said it "cannot comment on or confirm the status of open investigations."

In a statement to NPR, Caitlin Oakley, a spokesperson for HHS, said: "This is a personnel matter that is currently under review. However, HHS strongly disagrees with the allegations and characterizations in the complaint from Dr. Bright."

Electrochemical corrosion potential (ECP) probe assemblies may be used to monitor ECP of materials due to coolant chemistry in an operating nuclear reactor. Example embodiment assemblies include at least one ECP probe that detects ECP of potentially several different materials, a structural body providing a fluid flow path for the coolant over the ECP probes, and a signal transmitter that transmits or carries ECP data to an external receiver. The ECP probes may be of any number and/or type, so as to detect ECP for different component materials, including stainless steel, a zirconium alloys, etc. The ECP probes may further detect ECP due to ion concentration, pH, etc. The ECP data may be transmitted through wired or wireless signal transmitters. Example methods include installing and using example embodiment ECP probe assemblies in nuclear reactors and facilities.

In each split plunger the diagonal surface of the upper split plunger part exerts a transverse force to the diagonal surface of the lower split plunger part ensuring good electrical contact between the lower split plunger member part and the barrel wall.
