The Amateur Traveler talks to Italophile Ira Bernstein who returns to the show to talk about day trips in Tuscany that you can take from Florence. We talk about visiting the walled city of Siena and its famous bareback horse race the palio, viewing the many towers of San Gimignano, climbing the leaning tower of Pisa (after you take the obligatory picture of someone holding it up), and walking on the walls of Lucca. Each town has its own character and attractions. Arezzo has a jewelry museum and the home of the “Father of Humanism” Petrarch. Fiesole is a rich enclave, a suburb of Florence with views of the city and a roman theatre. Montepulciano and Montalcino are both known for their wines. Ira talks about some of the events that the locals enjoy like the palios, flea markets and flag throwing competitions. So even if you just get as far out of Florence as Piazzale Michelangelo there are many places to explore in Tuscany.

Kara tells us Miami is not as it is pictured in TV shows like Miami Vice and Burn Notice (except perhaps that it is filled with beautiful people). Miami is known for its beach life and Kara tells us where the daring can go to bare all or where you can find a quiet spot away from the tourists. Increasingly Miami is known for its fashion district and its food scene. The summers are hot but offer some of the best bargains on restaurants, hotels and spas… if you can deal with the humidity. Miami defines hot in all senses of the word. So whether you want to see snowbirds or eat stone crabs, Miami might be a place you should check out.

I have the life that I have because I've made a lot of weird decisions, and they've worked out well.


Not gonna lie to you. I'm a huge nerd. Surprised?

Yep. I can recite big chunks of The Big Lebowski from memory. I can argue for an hour on the merits of Dick York over Dick Sargent. And, I can—and frequently do—catch myself thinking Catwoman, Batgirl, Princess Leia, and Emma Peel should have a light-hearted pillow fight that ends with an hour of genial french-kissing.

Pretty much like you, probably. I dunno, maybe your version includes Kitty Pryde. Po-tay-to/Po-taht-o, right?

Perhaps most saliently, by virtue of having spent a solid 2,399 days as a Fake Productivity Guru, I have been provided with an unquestionably Janusian monkey's paw of a gift; I now know a lot about workflows. Nerdy, nerdy workflows.

I can tell you a few things that almost always work, I can tell you a handful of things that almost never work, and—best or worst of all—I can tell you thousands of things that might work. Sometimes. Maybe. Kinda. For some people. For now.

And, at the risk of gay-marrying my arrogance to my hypocrisy, I can tell you that I also know enough about the unholy diarrhea of potential options for Theoretical Productivity to share two big patterns:

  1. Getting your workflow right matters.
  2. Getting your workflow right to the exclusion of the actual work is a fool's game.

But. Managing to get the most useful and most elegant and least fiddly mix of 1 and 2 right is super-hard. Especially for nerds. Especially for me.


So, as I type this today, I believe there can be no greater testament to these claims—or, at least, no greater place to test the veracity of these claims for yourself—than in this TWO AND A HALF HOUR-long interview for Mac Power Users.

It is reeeeeeeeally nerdy. Almost intolerably nerdy. Just…overwhelmingly nerdy.

But, man, is it ever really good, and really fat with the most insanely granular details of How I Work.

Lo, even these 928.5 days after officially retiring from productivity pr0n, my desire to not "vend stroke material for your joyless addiction to puns about procrastination and systems for generating more taxonomically satisfying meta-work" is tempered by a (widely under-reported) practical streak.

Yes: I continue to despise empty advice about rearranging deck chairs on The Titanic. But, yes: I do also still very much enjoy talking about how all the tips and tricks can or can't work in the context of work you care about. That matters. It really does.


So. Here goes. A one hundred and forty six minute-long, Joyce-ian amble through the Big Stuff and the Little Stuff. David and Katie were very patient.

How I name text files. Why I break iOS apps. Why I love the letter "x." Why I won't row out to islands any more. How a 115,000 word book manuscript is "like a house full of confederate money." How "The Cloud" broke in New Zealand. How I use MultiMarkdown, Scrivener, TextExpander, OmniFocus, TextMate, Notational Velocity, Dropbox, and an explosive combination of Elements, Notesy, Nebulous, Simplenote, CF Outliner, iThoughts, Instacast, Good Reader, and wow wow wow.

How I try not to fiddle—how I sometimes succeed and often don't. But, how I try.

Anyhow. There you go. A perfectly nerdy bookend to last year's first Magnum Opus MPU interview on these same topics, Mac Power Users Episode 46 is just insanely nerdy. And, what have you.

I hope you like it. I hope it's useful. I hope you don't use it to replace real work.

And, as ever, I really hope Batgirl starts having more sexy pillow fights.

Prince Harry Opens the Stavros Niarchos Foundation Floor

Prince Harry visited Chatham House on 15 June to open the Stavros Niarchos Foundation Floor.

His Royal Highness met with young fellows from the Queen Elizabeth II Academy for Leadership in International Affairs and discussed the important role that the next generation of leaders must play in bringing about positive change in their communities. 

The Academy was formally launched by Her Majesty The Queen at Chatham House in 2014.

Prince Harry also contributed to the first scenario exercise held in the institute’s new simulation centre, which explored how to respond to a humanitarian emergency that required landmine clearance, drawing on the prince’s work in the field of landmine eradication.

The opening of the Stavros Niarchos Foundation Floor marks a significant moment in the modern history of the institute and is a core component of the Chatham House Second Century Initiative, which aims to strengthen the institute’s capacity to innovate and meet the growing demand for its research in the lead-up to its centenary in 2020.

The extension includes a series of new facilities, including the Asfari Centre for Academy Fellows, the simulation centre, new meeting spaces and a media studio, which will ensure that Chatham House can continue to contribute to building a sustainably secure, prosperous and just world over the coming decades.

Sea ice is one of the least understood components of our climate. Naturally its abundance or scarcity is a telling sign of climate change, but sea ice is also an important actor in change as well, insulating the ocean and reflecting sunlight. A branch of mathematics called percolation theory helps explain how salt water travels through sea ice, a process that is crucial both to the amount of sea ice present and to the microscopic communities that sustain polar ecosystems. By taking samples, doing on-site experiments, and then incorporating the data into models of porous materials, mathematicians are working to understand sea ice and help refine climate predictions. Using probability, numerical analysis, and partial differential equations, researchers have recently shown that the permeability of sea ice is similar to that of some sedimentary rocks in the earth.s crust, even though the substances are otherwise dissimilar. One major difference between the two is the drastic changes in permeability of sea ice, from total blockage to clear passage, that occur over a range of just a few degrees. This difference can have a major effect on measurements by satellite, which provide information on the extent and thickness of sea ice. Results about sea ice will not only make satellite measurements more reliable, but they can also be applied to descriptions of lung and bone porosity, and to understanding ice on other planets. Banks to ease cashflow pressure

The banking sector today launched a scheme to alleviate the cashflow pressure faced by corporate customers in light of the economic challenges brought about by the COVID-19 outbreak.

 

Jointly announced by the Monetary Authority and the Banking Sector SME Lending Coordination Mechanism, the Pre-approved Principal Payment Holiday Scheme is estimated to cover more than 80% of all corporate borrowers in Hong Kong.

 

All corporate customers that have an annual sales turnover of $800 million or less and with no outstanding loan payments overdue for more than 30 days are eligible for the scheme.

 

Eligible customers’ loan principal payments due within a six-month period between May 1 and October 31 this year will be pre-approved for deferment.

 

The loans’ principal payments, including revolving facilities, will generally be deferred by six months, whereas trade facilities, given their short-term nature, will be deferred by three months.

 

The scheme does not cover syndicated loans or loans used for financing purchases of shares or other financial assets.

 

Banks will begin informing eligible customers of the pre-approval under the scheme as soon as practicable. Eligible customers do not need to apply. They only need to contact the bank within 14 days of the bank’s notice to confirm the detailed arrangements.

 

The authority has issued a circular requesting all banks to participate in the scheme and has received support from all 11 major lenders in the co-ordination mechanism.

 

It will monitor the scheme’s operation and maintain close dialogue with banks.

(Florida Museum of Natural History) Florida and Georgia archaeologists have discovered the location of Fort San Antón de Carlos, home of one of the first Jesuit missions in North America. A Quantitative Tri-fluorescent Yeast Two-hybrid System: From Flow Cytometry to In cellula Affinities [Technological Innovation and Resources]

We present a technological advancement for the estimation of the affinities of Protein-Protein Interactions (PPIs) in living cells. A novel set of vectors is introduced that enables a quantitative yeast two-hybrid system based on fluorescent fusion proteins. The vectors allow simultaneous quantification of the reaction partners (Bait and Prey) and the reporter at the single-cell level by flow cytometry. We validate the applicability of this system on a small but diverse set of PPIs (eleven protein families from six organisms) with different affinities; the dissociation constants range from 117 pm to 17 μm. After only two hours of reaction, expression of the reporter can be detected even for the weakest PPI. Through a simple gating analysis, it is possible to select only cells with identical expression levels of the reaction partners. As a result of this standardization of expression levels, the mean reporter levels directly reflect the affinities of the studied PPIs. With a set of PPIs with known affinities, it is straightforward to construct an affinity ladder that permits rapid classification of PPIs with thus far unknown affinities. Conventional software can be used for this analysis. To permit automated analysis, we provide a graphical user interface for the Python-based FlowCytometryTools package.

Flow-induced reorganization of laminin-integrin networks within the endothelial basement membrane uncovered by proteomics [Research]

The vessel wall is continuously exposed to hemodynamic forces generated by blood flow. Endothelial mechanosensors perceive and translate mechanical signals via cellular signaling pathways into biological processes that control endothelial development, phenotype and function. To assess the hemodynamic effects on the endothelium on a system-wide level, we applied a quantitative mass spectrometry approach combined with cell surface chemical footprinting. SILAC-labeled endothelial cells were subjected to flow-induced shear stress for 0, 24 or 48 hours, followed by chemical labeling of surface proteins using a non-membrane permeable biotin label, and analysis of the whole proteome and the cell surface proteome by LC-MS/MS analysis. These studies revealed that of the >5000 quantified proteins 104 were altered, which were highly enriched for extracellular matrix proteins and proteins involved in cell-matrix adhesion. Cell surface proteomics indicated that LAMA4 was proteolytically processed upon flow-exposure, which corresponded to the decreased LAMA4 mass observed on immunoblot. Immunofluorescence microscopy studies highlighted that the endothelial basement membrane was drastically remodeled upon flow exposure. We observed a network-like pattern of LAMA4 and LAMA5, which corresponded to the localization of laminin-adhesion molecules ITGA6 and ITGB4. Furthermore, the adaptation to flow-exposure did not affect the inflammatory response to tumor necrosis factor α, indicating that inflammation and flow trigger fundamentally distinct endothelial signaling pathways with limited reciprocity and synergy. Taken together, this study uncovers the blood flow-induced remodeling of the basement membrane and stresses the importance of the subendothelial basement membrane in vascular homeostasis.

Empagliflozin Ameliorates Obesity-Related Cardiac Dysfunction by Regulating Sestrin2-Mediated AMPK-mTOR Signaling and Redox Homeostasis in High-Fat Induced Obese Mice

Sodium glucose co-transporter-2 inhibitors (SGLT2i) have favorable cardiovascular outcomes in diabetic patients. However, whether SGLT2i can improve obesity-related cardiac dysfunction is unknown. Sestrin2 is a novel stress-inducible protein that regulates AMPK-mTOR and suppresses oxidative damage. The aim of this study was to determine whether empagliflozin (EMPA) improves obesity-related cardiac dysfunction via regulating Sestrin2-mediated pathways in diet-induced obesity. C57BL/6J mice and Sestrin2 knockout mice were fed a high-fat diet (HFD) for 12 weeks and then treated with or without EMPA (10 mg/kg) for 8 weeks. Treating HFD-fed C57BL/6J mice with EMPA reduced body weight, whole-body fat, and improved metabolic disorders. Furthermore, EMPA improved myocardial hypertrophy/fibrosis and cardiac function, and reduced cardiac fat accumulation and mitochondria injury. Additionally, EMPA significantly augmented Sestrin2 levels, increased AMPK and eNOS phosphorylation, but inhibited Akt and mTOR phosphorylation. These beneficial effects were partially attenuated in HFD-fed Sestrin2 knockout mice. Intriguingly, EMPA treatment enhanced the Nrf2/HO-1-mediated oxidative stress response, suggesting antioxidant and anti-inflammatory activity. Thus, EMPA improved obesity-related cardiac dysfunction via regulating Sestrin2-mediated AMPK-mTOR signaling and maintaining redox homeostasis. These findings provide a novel mechanism for the cardiovascular protection of SGLT2i in obesity.

Integrated Pancreatic Blood Flow: Bi-Directional Microcirculation Between Endocrine and Exocrine Pancreas

The pancreatic islet is a highly-vascularized endocrine micro-organ. The unique architecture of rodent islets, a so-called core-mantle arrangement seen in 2D images, led researchers to seek functional implications for islet hormone secretion. Three models of islet blood flow were previously proposed, all based on the assumption that islet microcirculation occurs in an enclosed structure. Recent electrophysiological and molecular biological studies using isolated islets also presumed uni-directional flow. Using intravital analysis of the islet microcirculation in mice, we find that islet capillaries are continuously integrated to those in the exocrine pancreas, which makes the islet circulation rather open, not self-contained. Similarly in human islets, the capillary structure was integrated with pancreatic microvasculature in its entirety. Thus, islet microcirculation has no relation to islet cytoarchitecture, which explains its well-known variability throughout species. Furthermore, tracking fluorescent-labeled red blood cells at the endocrine-exocrine interface revealed bi-directional blood flow, with similar variability in blood flow speed in both the intra- and extra-islet vasculature. To date, the endocrine and exocrine pancreas have been studied separately by different fields of investigators. We propose that the open circulation model physically links both endocrine and exocrine parts of the pancreas as a single organ through the integrated vascular network.
