Buying ‘China-made’ dreams in Nepal

With China taking greater interest in the internal politics of Nepal and funding several projects in the country, all eyes in the neighbourhood are on the Himalayan nation. However, as most of the projects awarded to Chinese firms lie in limbo and mired in cost-overrun, it is time that Kathmandu pauses to evaluate its engagement with the dragon Nestled in between two gigantic neighbours—India and China—Nepal has often found itself mired in situations when it has to juggle the primacy it places on its ties with the two nations. The Himalayan nation’s allegiance to either an assertive New Delhi and an imperial Beijing has shifted back and forth over the years depending on the powers that be. There is much talk lately about China and its funding to infrastructure projects that would ostensibly bring prosperity to Nepal. A new narrative spun by sections of Nepalese society and political class says that the renewed Chinese support would reduce Nepal’s dependence on its southern neighbour, India. This narrative received an increased impetus subsequent to Madhesi blockade in 2015 against the newly promulgated Constitution that was perceived to be non-inclusive and anti-federalism. The protest movement was projected as India-sponsored ‘economic blockade’ by a section of Nepalese media holding anti-India views and also the then ruling political party, Communist Party of Nepal, (CPN (UML). The attempts thereafter have been to cosy-up to China to attract investments and provide them as much space as they want in Nepalese economy, society and also polity. But one must pause to see the direction the Nepalese are headed in? Are these efforts to seek investment from China actually resulting in robust infrastructure in Nepal, except few cosmetic changes here and there? All major China-supported projects in the Himalayan nation are mired in cost overrun, delayed implementation and compromise on quality, like the West Seti, the Kulekhani 3 and the Budhigandaki hydropower projects. Various projects taken over by Chinese companies continue to remain incomplete as neither the agreed terms and conditions were honoured by the Chinese nor the time schedule on which they were supposed to be completed. A study of the actual on-the-ground situation in each of the five mega infrastructure projects funded by the Chinese is highly revelatory. It was not only the norms of awarding a project to a foreign entity that were flouted in favour of China but also major compromises were made in the process. Nepal’s impatience to grab Chinese investments has been fully exploited by China, which has been trying to wean them away from India’s sphere of influence like a pied piper. Take the example of West Seti hydropower project which was awarded to a sub-group of China, Three Gorges International Corporation (CTGC), in 2012 without any international bidding. The plan was to create a 750MW power generation unit but the project did not see any progress till 2016. Last year, after the Nepalese government threatened to scrap the deal, CTGC signed a framework agreement with Nepal Electricity Authority in January 2017 but the project still hangs in limbo because the agreement is yet to be ratified. The original idea of the project was to export the electricity generated from the plant to India. However, at the behest of China, Nepal eliminated that clause and inserted one which said that the electricity will be consumed domestically. It was not only the norms of awarding a project to a foreign entity that were flouted in favour of China but also major compromises were made in the process. Nepal’s impatience to grab Chinese investments has been fully exploited by China Another instance of delayed implementation relates to Kulekhani 3 hydropower project which has missed its fourth revised completion deadline set for July 2017. It was awarded to awarded to Chinese companies Zhejiang Jiahi and Sino Hydro in 2008 and was scheduled to be completed in 2012. The project faced not only time overruns but also cost escalation due to delays. The Initial estimated cost which was NR 2.43 billion is now doubled to NR 4.22 billion. The inordinate delay in the completion of the project prompted the National Planning Commission of Nepal to declare Kulekhani 3 a “troubled hydropower project.” The fate of Gautam Buddha Airport upgradation project, awarded to North West Civil Aviation Airport Construction Group of China in 2014, appears to be steeped in uncertainty just like the others. The airport infrastructure was to be improved as part of Nepal’s national pride project so that it could function as an international airport to serve the fast-rising business and industrial hub of Bhairahawa and facilitate international pilgrimage tourism to Lumbini, the birthplace of the Buddha. The upgradation work which was to be completed by December 2017 is now revised to be finished by 2019. Nepalese authorities have attributed slow progress on the project to very low bidding price and the weak cash flow situation in the Chinese company which grabbed the tender. Exasperated at the lack of desired progress, the Civil Aviation Authority of Nepal is contemplating measures to terminate the contract of the Chinese firm. They have also hired international contract management experts to resume the upgradation work at the airport. After remaining mired in troubled relating to funding problems, Pokhara Regional International Airport witnessed some progress in August 2017 when the Chinese company, which was awarded the contract, started the construction work. The project, which was formulated at the cost of NR 22 billion, will be financed by China’s Export and Import Bank under a loan agreement with Nepal government. The airport, which was to be ready by July 2020, is now planned to be completed by July 2021 on paper at least. Whether it will happen or not remains to be seen. Likewise, Solar Power Project has run into controversy with the PAC directing the government to scrap the contract awarded to a Chinese company. The committee contended that the decision was against procurement law. On May 16, China’s Risen Co Energy Limited had bagged the contract from the Nepal Electricity Authority (NEA) management at the price of approximately NR 3.88 billion, despite the fact that the bidder ranked fourth in the financial proposal. An English daily in Nepal quoted PAC chairman Dor Prasad Upadhyaya as saying, “There (were) other bidders with equally good technical qualifications as that of the selected bidder, but the contract was given to this company in a premeditated way.” The committee has now asked the authorities to start a fresh tender to install solar power plants in the premises of Kulekhani reservoir in Makwanpur district and Devighat Hydropower Project in Nuwakot district. The project is already one and a half year behind its schedule and the NEA has stated that it could be abandoned if the World Bank decides to withdraw from financing due to overrun. The Budhi Gandaki hydropower project, awarded in June 2017 to China Gezhouba Group Corporation without any international bidding, has also run into troubles. A joint meeting of the parliament’s Agriculture and Water Resources Committee (AWRC) and Finance Committee has instructed the government to scrap the contract awarded to the Chinese firm to build the 1,200-megawatt hydroelectric project because the tender was awarded without bidding in violation of the Public Procurement Act. An English daily quoted Prakash Jwala, chairperson of the Finance Committee, as saying that the government made a blunder by awarding the project to a firm whose track record has been abysmal. He had said, “This project was handed over to a controversial Chinese company whose track record is so bad in Nepal by flouting various laws like Public Procurement Act. The intention behind selecting this Chinese firm is only to hold the project. How can a company, which could not build even a 30-megawatt project (Chameliya), build such a mega project?” Opposition to the project has also come up from political parties citing provisions in the Nepalese Constitution which ensures compulsory local investment in the development of natural resources. Experts are of the view that if the Chinese company proceeds with the development of the project, the cost is bound to inflate. Taking into account this viewpoint, the PAC of Nepal has summoned the concerned government officials involved in the deal. Opposition to the project has also come up from political parties citing provisions in the Nepalese Constitution which ensures compulsory local investment in the development of natural resources After the two-month-long standoff with India in Doklam plateau, China has accelerated its engagement with Nepal. The country’s foreign minister Wang Yi has promised to take the ties between Kathmandu and Beijing to a new level and the sentiment got a major boost when Nepal’s deputy prime minister Krishna Bahadur Mahara visited Beijing in September first week and met several high-level officials, including Premier Li Keqiang. To show that China approved of Nepal’s neutral stand during the Doklam standoff, Li had also said, “China is ready to provide support within its due capacity toNepal’s economic and social development.” Because of the growing bonhomie between the two nations, quite obviously, the developments in Nepal are being watched keenly by the neighbourhood. With the Left parties coming together to form a pre-poll alliance in October first week, the rumour mill is churning various kinds of news as to how the tieup will impact India and China. While some experts allege India’s role in forging the alliance, others claim that the tieup had received Beijing’s nod. The opinion is divided over how it will benefit or harm India’s interests. Considering that New Delhi has had its share of troubles with the CPN (UML) chief K.P. Sharma Oli in the past, a unity government under Oli’s leadership will be in Beijing’s favour. However, Nepal needs to tread the ground carefully. While seeking closeness to and investments from Beijing, Nepal will have to seek answers to a few questions: Why is China interested in Nepal? By helping Nepal, is Beijing serving its own nationalistic interests? Could the funds coming in from China land Nepal in a debt trap like Sri Lanka (which was forced to lease its Hambantota port to China for 99 years in order to pay back the debt)? Nepalese must ask themselves what are the commonalities between China and Nepal, in terms of religion, language, food, dress, culture and identity? China, as of now, is like a new toy being held in high esteem. Once its glitter is gone and it shows its true nature, it might be too late for Nepal.

A lathe system for micrometre-sized cylindrical sample preparation at room and cryogenic temperatures

A simple two-spindle based lathe system for the preparation of cylindrical samples intended for X-ray tomography is presented. The setup can operate at room temperature as well as under cryogenic conditions, allowing the preparation of samples down to 20 and 50 µm in diameter, respectively, within minutes. Case studies are presented involving the preparation of a brittle biomineral brachiopod shell and cryogenically fixed soft brain tissue, and their examination by means of ptychographic X-ray computed tomography reveals the preparation method to be mainly free from causing artefacts. Since this lathe system easily yields near-cylindrical samples ideal for tomography, a usage for a wide variety of otherwise challenging specimens is anticipated, in addition to potential use as a time- and cost-saving tool prior to focused ion-beam milling. Fast sample preparation becomes especially important in relation to shorter measurement times expected in next-generation synchrotron sources.

Crystal structures of two isomeric 2-aryl-3-phenyl-1,3-thia­zepan-4-ones

The crystal of 6-(3-nitro­phen­yl)-7-phenyl-5-thia-7-aza­spiro­[2.6]nonan-8-one (1), C19H18N2O3S, has monoclinic (P21/n) symmetry while that of its isomer 6-(4-nitro­phen­yl)-7-phenyl-5-thia-7-aza­spiro­[2.6]nonan-8-one (2), has ortho­rhom­bic (Pca21) symmetry: compound 1 has two mol­ecules, A and B, in the asymmetric unit while 2 has one. In all three mol­ecules, the seven-membered thia­zepan ring exhibits a chair conformation with Q2 and Q3 values (Å) of 0.521 (3), 0.735 (3) and 0.485 (3), 0.749 (3) in 1 and 0.517 (5), 0.699 (5) in 2. In each structure, the phenyl rings attached to adjacent atoms of the thia­zepan ring have inter­planar angles ranging between 41 and 47°. Except for the nitro groups, the three mol­ecules have similar conformations when overlayed in pairs. Both crystal structures are consolidated by C—H⋯O hydrogen bonds.

The `super acid' BF3H2O stabilized by 1,4-dioxane: new preparative aspects and the crystal structure of BF3H2O·C4H8O2

Highly Brønsted-acidic boron trifluoride monohydrate, a widely used `super acid-catalyst', is a colourless fuming liquid that releases BF3 at room temperature. Com­pared to the liquid com­ponents, i.e. boron trifluoride monohydrate and 1,4-dioxane, their 1:1 adduct, BF3H2O·C4H8O2, is a solid with pronounced thermal stability (m.p. 401–403 K). The crystal structure of the long-time-stable easy-to-handle and weighable com­pound is reported along with new preparative aspects and the results of 1H, 11B, 13C and 19F spectroscopic investigations, particularly documenting its high Brønsted acidity in aceto­nitrile solution. The remarkable stability of solid BF3H2O·C4H8O2 is attributed to the chain structure established by O—H⋯O hydrogen bonds of exceptional strength {O2⋯H1—O1 [O⋯O = 2.534 (3) Å] and O1—H1⋯O3i [2.539 (3) Å] in the concatenating unit >O2⋯H1—O1—H2⋯O3i<}, taking into account the mol­ecular (non-ionic) character of the structural moieties. Indirectly, this structural feature documents the outstanding acidification of the H2O mol­ecule bound to BF3 and reflects the super acid nature of BF3H2O. In detail, the C22(7) zigzag chain system of hydrogen bonding in the title structure is characterized by the double hydrogen-bond donor and double (κO,κO') hydrogen-bond acceptor functionality of the aqua ligand and dioxane molecule, respectively, the almost equal strength of both hydrogen bonds, the approximatety linear arrangement of the dioxane O atoms and the two neighbouring water O atoms. Furthermore, the approximately planar arrangement of B, F and O atoms in sheets perpendicular to the c axis of the ortho­rhom­bic unit cell is a characteristic structural feature.

Structure refinement of (NH4)3Al2(PO4)3 prepared by ionothermal synthesis in phospho­nium based ionic liquids – a redetermination

After crystallization during ionothermal syntheses in phospho­nium-containing ionic liquids, the structure of (NH4)3Al2(PO4)3 [tri­ammonium dialuminum tris­(phosphate)] was refined on the basis of powder X-ray diffraction data from a synchrotron source. (NH4)3Al2(PO4)3 is a member of the structural family with formula A3Al2(PO4)3, where A is a group 1 element, and of which the NH4, K, and Rb forms were previously known. The NH4 form is isostructural with the K form, and was previously solved from single-crystal X-ray data when the material (SIZ-2) crystallized from a choline-containing eutectic mixture [Cooper et al. (2004). Nature, 430, 1012–1017]. Our independent refinement incorporates NH4 groups and shows that these NH4 groups are hydrogen bonded to framework O atoms present in rings containing 12 T sites in a channel along the c-axis direction. We describe structural details of (NH4)3Al2(PO4)3 and discuss differences with respect to isostructural forms.

Crystal structure, Hirshfeld analysis and a mol­ecular docking study of a new inhibitor of the Hepatitis B virus (HBV): ethyl 5-methyl-1,1-dioxo-2-{[5-(pentan-3-yl)-1,2,4-oxa­diazol-3-yl]meth­yl}-2H-1,2,6-thia­diazine-4-carboxyl­a

The title compound, C15H22N4O5S, was prepared via alkyl­ation of 3-(chloro­meth­yl)-5-(pentan-3-yl)-1,2,4-oxa­diazole in anhydrous dioxane in the presence of tri­ethyl­amine. The thia­diazine ring has an envelope conformation with the S atom displaced by 0.4883 (6) Å from the mean plane through the other five atoms. The planar 1,2,4-oxa­diazole ring is inclined to the mean plane of the thia­diazine ring by 77.45 (11)°. In the crystal, mol­ecules are linked by C—H⋯N hydrogen bonds, forming chains propagating along the b-axis direction. Hirshfeld surface analysis and two-dimensional fingerprint plots have been used to analyse the inter­molecular contacts present in the crystal. Mol­ecular docking studies were use to evaluate the title compound as a potential system that inter­acts effectively with the capsid of the Hepatitis B virus (HBV), supported by an experimental in vitro HBV replication model.

Structure of a push–pull olefin prepared by ynamine hydro­boration with a borandiol ester

N-[(Z)-2-(2H-1,3,2-Benzodioxaborol-2-yl)-2-phenyl­ethen­yl]-N-(propan-2-yl)aniline, C23H22BNO2, contains a C=C bond that is conjugated with a donor and an acceptor group. An analysis that included similar push–pull olefins revealed that bond lengths in their B—C=C—N core units correlate with the perceived acceptor and donor strength of the groups. The two phenyl groups in the mol­ecule are rotated with respect to the plane that contains the BCCN atoms, and are close enough for significant π-stacking. Definite characterization of the title compound demonstrates, for the first time in a reliable way, that hydro­boration of ynamines with borandiol esters is feasible. Compared to olefin hydro­boration with borane, the ynamine substrate is activated enough to undergo reaction with the less active hydro­boration reagent catecholborane.

X-ray diffraction using focused-ion-beam-prepared single crystals

High-quality single-crystal X-ray diffraction measurements are a prerequisite for obtaining precise and reliable structure data and electron densities. The single crystal should therefore fulfill several conditions, of which a regular defined shape is of particularly high importance for compounds consisting of heavy elements with high X-ray absorption coefficients. The absorption of X-rays passing through a 50 µm-thick LiNbO3 crystal can reduce the transmission of Mo Kα radiation by several tens of percent, which makes an absorption correction of the reflection intensities necessary. In order to reduce ambiguities concerning the shape of a crystal, used for the necessary absorption correction, a method for preparation of regularly shaped single crystals out of large samples is presented and evaluated. This method utilizes a focused ion beam to cut crystals with defined size and shape reproducibly and carefully without splintering. For evaluation, a single-crystal X-ray diffraction study using a laboratory diffractometer is presented, comparing differently prepared LiNbO3 crystals originating from the same macroscopic crystal plate. Results of the data reduction, structure refinement and electron density reconstruction indicate qualitatively similar values for all prepared crystals. Thus, the different preparation techniques have a smaller impact than expected. However, the atomic coordinates, electron densities and atomic charges are supposed to be more reliable since the focused-ion-beam-prepared crystal exhibits the smallest extinction influences. This preparation technique is especially recommended for susceptible samples, for cases where a minimal invasive preparation procedure is needed, and for the preparation of crystals from specific areas, complex material architectures and materials that cannot be prepared with common methods (breaking or grinding).

A cryo-EM grid preparation device for time-resolved structural studies

Structural biology generally provides static snapshots of protein conformations that can provide information on the functional mechanisms of biological systems. Time-resolved structural biology provides a means to visualize, at near-atomic resolution, the dynamic conformational changes that macromolecules undergo as they function. X-ray free-electron-laser technology has provided a powerful tool to study enzyme mechanisms at atomic resolution, typically in the femtosecond to picosecond timeframe. Complementary to this, recent advances in the resolution obtainable by electron microscopy and the broad range of samples that can be studied make it ideally suited to time-resolved approaches in the microsecond to millisecond timeframe to study large loop and domain motions in biomolecules. Here we describe a cryo-EM grid preparation device that permits rapid mixing, voltage-assisted spraying and vitrification of samples. It is shown that the device produces grids of sufficient ice quality to enable data collection from single grids that results in a sub-4 Å reconstruction. Rapid mixing can be achieved by blot-and-spray or mix-and-spray approaches with a delay of ∼10 ms, providing greater temporal resolution than previously reported mix-and-spray approaches.

The resolution revolution in cryoEM requires high-quality sample preparation: a rapid pipeline to a high-resolution map of yeast fatty acid synthase

Single-particle electron cryo-microscopy (cryoEM) has undergone a `resolution revolution' that makes it possible to characterize megadalton (MDa) complexes at atomic resolution without crystals. To fully exploit the new opportunities in molecular microscopy, new procedures for the cloning, expression and purification of macromolecular complexes need to be explored. Macromolecular assemblies are often unstable, and invasive construct design or inadequate purification conditions and sample-preparation methods can result in disassembly or denaturation. The structure of the 2.6 MDa yeast fatty acid synthase (FAS) has been studied by electron microscopy since the 1960s. Here, a new, streamlined protocol for the rapid production of purified yeast FAS for structure determination by high-resolution cryoEM is reported. Together with a companion protocol for preparing cryoEM specimens on a hydrophilized graphene layer, the new protocol yielded a 3.1 Å resolution map of yeast FAS from 15 000 automatically picked particles within a day. The high map quality enabled a complete atomic model of an intact fungal FAS to be built.

Crystal structures of the Bacillus subtilis prophage lytic cassette proteins XepA and YomS

As part of the Virus-X Consortium that aims to identify and characterize novel proteins and enzymes from bacteriophages and archaeal viruses, the genes of the putative lytic proteins XepA from Bacillus subtilis prophage PBSX and YomS from prophage SPβ were cloned and the proteins were subsequently produced and functionally characterized. In order to elucidate the role and the molecular mechanism of XepA and YomS, the crystal structures of these proteins were solved at resolutions of 1.9 and 1.3 Å, respectively. XepA consists of two antiparallel β-sandwich domains connected by a 30-amino-acid linker region. A pentamer of this protein adopts a unique dumbbell-shaped architecture consisting of two discs and a central tunnel. YomS (12.9 kDa per monomer), which is less than half the size of XepA (30.3 kDa), shows homology to the C-terminal part of XepA and exhibits a similar pentameric disc arrangement. Each β-sandwich entity resembles the fold of typical cytoplasmic membrane-binding C2 domains. Only XepA exhibits distinct cytotoxic activity in vivo, suggesting that the N-terminal pentameric domain is essential for this biological activity. The biological and structural data presented here suggest that XepA disrupts the proton motive force of the cytoplasmatic membrane, thus supporting cell lysis.

Shake-it-off: a simple ultrasonic cryo-EM specimen-preparation device

Although microscopes and image-analysis software for electron cryomicroscopy (cryo-EM) have improved dramatically in recent years, specimen-preparation methods have lagged behind. Most strategies still rely on blotting microscope grids with paper to produce a thin film of solution suitable for vitrification. This approach loses more than 99.9% of the applied sample and requires several seconds, leading to problematic air–water interface interactions for macromolecules in the resulting thin film of solution and complicating time-resolved studies. Recently developed self-wicking EM grids allow the use of small volumes of sample, with nanowires on the grid bars removing excess solution to produce a thin film within tens of milliseconds from sample application to freezing. Here, a simple cryo-EM specimen-preparation device that uses components from an ultrasonic humidifier to transfer protein solution onto a self-wicking EM grid is presented. The device is controlled by a Raspberry Pi single-board computer and all components are either widely available or can be manufactured by online services, allowing the device to be constructed in laboratories that specialize in cryo-EM rather than instrument design. The simple open-source design permits the straightforward customization of the instrument for specialized experiments.

Successful sample preparation for serial crystallography experiments

Serial crystallography, at both synchrotron and X-ray free-electron laser light sources, is becoming increasingly popular. However, the tools in the majority of crystallization laboratories are focused on producing large single crystals by vapour diffusion that fit the cryo-cooled paradigm of modern synchrotron crystallography. This paper presents several case studies and some ideas and strategies on how to perform the conversion from a single crystal grown by vapour diffusion to the many thousands of micro-crystals required for modern serial crystallography grown by batch crystallization. These case studies aim to show (i) how vapour diffusion conditions can be converted into batch by optimizing the length of time crystals take to appear; (ii) how an understanding of the crystallization phase diagram can act as a guide when designing batch crystallization protocols; and (iii) an accessible methodology when attempting to scale batch conditions to larger volumes. These methods are needed to minimize the sample preparation gap between standard rotation crystallography and dedicated serial laboratories, ultimately making serial crystallography more accessible to all crystallographers.
