WHAT IS IMPLIED IN EVANGELICAL FAITH:

1. It implies an intellectual perception of the things, facts, and truths believed. No one can believe that which he does not understand.
2. Evangelical faith implies the appropriation of the truths of the Gospel to ourselves. It implies an acceptance of Christ as our wisdom, righteousness, sanctification, and redemption. The soul that truly believes in Christ, believes that He tasted death for every man, and of course for it. It apprehends Christ as the Saviour of the world, as offered to all, and embraces and receives Him for itself. It appropriates His atonement, and His resurrection and His intercession, and His promises to itself. Christ is thus presented in the Gospel not only as the Saviour of the world, but also to the individual acceptance of men. He saves the world no further than He saves individuals. Evangelical faith implies a personal acceptance and appropriation of Christ to meet the necessities of the individual soul.
3. Faith is a state of committal to Christ, and of course it implies that the soul will be unreseveredly yielded up to Him, in all His relations to it, so far and so fast as these are apprehended by the intellect.
4. Evangelical faith implies an evangelical life. This would not be true if faith were merely an intellectual state or exercise. But since, as we have seen, faith is of the heart, since it consists in the committal of the will to Christ, it follows, by a law of necessity, that the life will correspond with faith. Let this be kept in perpetual remembrance.
5. Evangelical faith implies repentance towards God, that is, a turning from sin to God.
6. Evangelical faith implies a renunciation of self-righteousness. It is impossible for one to embrace Christ as the Saviour of the soul, any further than he renounces all hope or expectation of being saved by his own works, or righteousness.
7. Of course it implies peace of mind. In Christ the soul finds its full and present salvation. It has found its resting-place in Christ, and rests in profound peace under the shadow of the Almighty.
8. It implies hope, as soon as the believing soul considers what is conveyed by the Gospel, that is, a hope of eternal life in and through Christ.
9. It implies joy in God and in Christ. St. Peter speaks of joy as the unfailing accompaniment of faith, as resulting from it.
10. Present evangelical faith implies a state of present sinlessness [covered by the blood of Jesus (1 John 1:7)]. Observe: faith is the yielding and committal of the whole will, and of the whole being to Christ. This, and nothing short of this, is evangelical faith. But this comprehends and implies the whole of present, true obedience to Christ. This is the reason why faith is spoken of as the condition, and as it were, the only condition, of salvation. It really implies all virtue.

When contemplated as an attribute of love, it is only a branch of sanctification. When contemplated in the wider sense of universal conformity of will to the will of God, it is then synonymous with entire present sanctification. Contemplated in either light, its existence in the heart must be inconsistent with present sin there. Monitoring farmland biodiversity across Europe: It could cost less than you think

How can we monitor Europe-wide farmland biodiversity so that it makes sense to farmers, is ecologically credible and scientifically sound and can be implemented for a reasonable price? Two new studies answer these questions.

First, stakeholders were asked, which indicators provided best "value for money" for their purpose. Habitat, plant species and farm management indicators ranked highest. Wild bees, earthworms and spiders as important providers of ecosystem services came next. Together they form a minimum set of indicators which provides non-redundant information and which can make dominant changes in farmland biodiversity visible.

Researchers from the FP7 funded EU projects "Biodiversity Indicators for European Farming Systems (BioBio)" and "Building the European Biodiversity Observation Network (EU BON)", then developed cost estimates for nine monitoring scenarios and the authors conclude that a continent-wide farmland biodiversity monitoring scheme would require only a modest share of the Common Agricultural Policy (CAP) budget (2014-2020).

Cost assessments showed that the farmland biodiversity monitoring scenarios require 0·01% - 0·74% of the total CAP budget and 0·04% - 2·48% of the CAP budget specifically allocated to environmental targets. With 30% of the CAP devoted to environmental targets (more than 120 billion EURO), investing in a monitoring process seems a logical choice given these results. The researchers provide a framework for individual countries to start farmland biodiversity monitoring, building towards a coherent European picture.

The studies were published in the Journal of Applied Ecology and the Journal of Environmental Management.

"Despite scientific proof that monitoring increases the (cost) efficiency of policy measures, monitoring rarely gets included in policy programme budgets. We identified that the cost are not as high as feared. To further facilitate implementation, the study provides stepping stones to build a European monitoring scheme, offering a choice in indicators and using regions as a unit of trend analysis," explains Dr. Ilse Geijzendorffer, the lead author of the Journal of Applied Ecology paper.

Seeded fibrils of the germline variant of human {lambda}-III immunoglobulin light chain FOR005 have a similar core as patient fibrils with reduced stability [Molecular Biophysics]

Systemic antibody light chains (AL) amyloidosis is characterized by deposition of amyloid fibrils derived from a particular antibody light chain. Cardiac involvement is a major risk factor for mortality. Using MAS solid-state NMR, we studied the fibril structure of a recombinant light chain fragment corresponding to the fibril protein from patient FOR005, together with fibrils formed by protein sequence variants that are derived from the closest germline (GL) sequence. Both analyzed fibril structures were seeded with ex-vivo amyloid fibrils purified from the explanted heart of this patient. We find that residues 11-42 and 69-102 adopt β-sheet conformation in patient protein fibrils. We identify arginine-49 as a key residue that forms a salt bridge to aspartate-25 in the patient protein fibril structure. In the germline sequence, this residue is replaced by a glycine. Fibrils from the GL protein and from the patient protein harboring the single point mutation R49G can be both heterologously seeded using patient ex-vivo fibrils. Seeded R49G fibrils show an increased heterogeneity in the C-terminal residues 80-102, which is reflected by the disappearance of all resonances of these residues. By contrast, residues 11-42 and 69-77, which are visible in the MAS solid-state NMR spectra, show 13Cα chemical shifts that are highly like patient fibrils. The mutation R49G thus induces a conformational heterogeneity at the C terminus in the fibril state, whereas the overall fibril topology is retained. These findings imply that patient mutations in FOR005 can stabilize the fibril structure.

A germline PAF1 paralog complex ensures cell type-specific gene expression [Research Papers]

Animal germline development and fertility rely on paralogs of general transcription factors that recruit RNA polymerase II to ensure cell type-specific gene expression. It remains unclear whether gene expression processes downstream from such paralog-based transcription is distinct from that of canonical RNA polymerase II genes. In Drosophila, the testis-specific TBP-associated factors (tTAFs) activate over a thousand spermatocyte-specific gene promoters to enable meiosis and germ cell differentiation. Here, we show that efficient termination of tTAF-activated transcription relies on testis-specific paralogs of canonical polymerase-associated factor 1 complex (PAF1C) proteins, which form a testis-specific PAF1C (tPAF). Consequently, tPAF mutants show aberrant expression of hundreds of downstream genes due to read-in transcription. Furthermore, tPAF facilitates expression of Y-linked male fertility factor genes and thus serves to maintain spermatocyte-specific gene expression. Consistently, tPAF is required for the segregation of meiotic chromosomes and male fertility. Supported by comparative in vivo protein interaction assays, we provide a mechanistic model for the functional divergence of tPAF and the PAF1C and identify transcription termination as a developmentally regulated process required for germline-specific gene expression.

ITPR1-associated spinocerebellar ataxia with craniofacial features--additional evidence for germline mosaicism [RESEARCH ARTICLE]

Inositol 1,4,5-triphosphate receptor type 1 (ITPR1) is an endoplasmic reticulum–bound intracellular inositol triphosphate receptor involved in the regulation of intracellular calcium. Pathogenic variants in ITPR1 are associated with spinocerebellar ataxia (SCA) types 15/16 and 29 and have recently been implicated in a facial microsomia syndrome. In this report, we present a family with three affected individuals found to have a heterozygous missense c.800C > T (predicted p.Thr267Met) who present clinically with a SCA29-like syndrome. All three individuals presented with varying degrees of ataxia, developmental delay, and apparent intellectual disability, as well as craniofacial involvement—an uncommon finding in patients with SCA29. The variant was identified using clinical exome sequencing and validated with Sanger sequencing. It is presumed to be inherited via parental germline mosaicism. We present our findings to provide additional evidence for germline mosaic inheritance of SCA29, as well as to expand the clinical phenotype of the syndrome.

Clinical and functional analysis of the germline TP53 p.K164E acetylation site variant [RESEARCH ARTICLE]

TP53 plays a critical role as a tumor suppressor by controlling cell cycle progression, DNA repair, and apoptosis. Post-translational modifications such as acetylation of specific lysine residues in the DNA binding and carboxy-terminus regulatory domains modulate its tumor suppressor activities. In this study, we addressed the functional consequences of the germline TP53 p.K164E (NM_000546.5: c.490A>G) variant identified in a patient with early-onset breast cancer and a significant family history of cancer. K164 is a conserved residue located in the L2 loop of the p53 DNA binding domain that is post-translationally modified by acetylation. In silico, in vitro, and in vivo analyses demonstrated that the glutamate substitution at K164 marginally destabilizes the p53 protein structure but significantly impairs sequence-specific DNA binding, transactivation, and tumor cell growth inhibition. Although p.K164E is currently considered a variant of unknown significance by different clinical genetic testing laboratories, the clinical and laboratory-based findings presented here provide strong evidence to reclassify TP53 p.K164E as a likely pathogenic variant.

Synchronous T-lymphoblastic lymphoma and neuroblastoma in a 3-yr-old with novel germline SMARCA4 and EZH2 variants [RAPID CANCER COMMUNICATION]

T-lymphoblastic lymphoma (T-LLy) is the most common lymphoblastic lymphoma in children and often presents with a mediastinal mass. Lymphomatous suprarenal masses are possible but rare. Here, we discuss the case of a previously healthy 3-yr-old male who presented with mediastinal T-LLy with bilateral suprarenal masses. Following initial treatment, surgical biopsy of persisting adrenal masses revealed bilateral neuroblastoma (NBL). A clinical genetics panel for germline cancer predisposition did not identify any pathogenic variants. Combination large panel (864 genes) profiling analysis in the context of a precision oncology study revealed two novel likely pathogenic heterozygous variants: SMARCA4 c.1420-1G > T p.? and EZH2 c.1943G > C p.(Ile631Phefs*44). Somatic analysis revealed potential second hits/somatic variants in EZH2 (in the T-LLy) and a segmental loss in Chromosome 19p encompassing SMARCA4 (in the NBL). Synchronous cancers, especially at a young age, warrant genetic evaluation for cancer predisposition; enrollment in a precision oncology program assessing germline and tumor DNA can fulfill that purpose, particularly when standard first-line genetic testing is negative and in the setting of tumors that are not classic for common cancer predisposition syndromes.
