The coordination of the ligands with respect to the central atom in the complex bromido­tricarbon­yl[diphen­yl(pyridin-2-yl)phosphane-κ2N,P]rhenium(I) chloro­form disolvate, [ReBr(C17H14NP)(CO)3]·2CHCl3 or [κ2-P,N-{(C6H5)2(C5H5N)P}Re(CO)3Br]·2CHCl3, (I·2CHCl3), is best described as a distorted octa­hedron with three carbonyls in a facial conformation, a bromide atom, and a biting P,N-di­phenyl­pyridyl­phosphine ligand. Hirshfeld surface analysis shows that C—Cl⋯H inter­actions contribute 26%, the distance of these inter­actions are between 2.895 and 3.213 Å. The reaction between I and piperidine (C5H11N) at 313 K in di­chloro­methane leads to the partial decoord­ination of the pyridyl­phosphine ligand, whose pyridyl group is replaced by a piperidine mol­ecule, and the complex bromido­tricarbon­yl[diphen­yl(pyridin-2-yl)phosphane-κP](piperidine-κN)rhenium(I), [ReBr(C5H11N)(C17H14NP)(CO)3] or [P-{(C6H5)2(C5H5N)P}(C5H11N)Re(CO)3Br] (II). The mol­ecule has an intra­molecular N—H⋯N hydrogen bond between the non-coordinated pyridyl nitro­gen atom and the amine hydrogen atom from piperidine with D⋯A = 2.992 (9) Å. Thermogravimetry shows that I·2CHCl3 losses 28% of its mass in a narrow range between 318 and 333 K, which is completely consistent with two solvating chloro­form mol­ecules very weakly bonded to I. The remaining I is stable at least to 573 K. In contrast, II seems to lose solvent and piperidine (12% of mass) between 427 and 463 K, while the additional 33% loss from this last temperature to 573 K corresponds to the release of 2-pyridyl­phosphine. The contribution to the scattering from highly disordered solvent mol­ecules in II was removed with the SQUEEZE routine [Spek (2015). Acta Cryst. C71, 9-18] in PLATON. The stated crystal data for Mr, μ etc. do not take this solvent into account.

In each of the products (RS)-3-(4-chloro­phen­yl)-5-[4-(prop-2-yn­yloxy)phen­yl]-4,5-di­hydro­pyrazole-1-carbo­thio­amide, C19H16ClN3OS, (IV), (RS)-3-(4-bromo­phen­yl)-5-[4-(prop-2-yn­yloxy)phen­yl]-4,5-di­hydro­pyrazole-1-carbo­thio­amide, C19H16BrN3OS, (V), and (RS)-3-(4-meth­oxy­phen­yl)-5-[4-(prop-2-yn­yloxy)phen­yl]-4,5-di­hydro­pyrazole-1-carbo­thio­amide, C20H19N3O2S, (VI), the reduced pyrazole ring adopts an envelope conformation with the C atom bearing the 4-prop-2-yn­yloxy)phenyl substituent, which occupies the axial site, displaced from the plane of the four ring atoms. Compounds (IV) and (V) are isomorphous and their mol­ecules are linked into chains of edge-fused rings by a combination of N—H⋯S and C—H⋯S hydrogen bonds. The mol­ecules of (VI) are linked into sheets by a combination of N—H⋯S, N—H⋯N and C—H⋯π(arene) hydrogen bonds. Comparisons are made with the structures of some related compounds.

The two new tetra­kis-substituted pyrazines, 1,1',1'',1'''-(pyrazine-2,3,5,6-tetra­yl) tetra­kis­(N,N-di­methyl­methanamine), C16H32N6, (I) and N,N',N'',N'''-[pyrazine-2,3,5,6-tetra­yltetra­kis­(methyl­ene)]tetra­kis­(N-methyl­aniline), C36H40N6, (II), both crystallize with half a mol­ecule in the asymmetric unit; the whole mol­ecules are generated by inversion symmetry. There are weak intra­molecular C—H⋯N hydrogen bonds present in both mol­ecules and in (II) the pendant N-methyl­aniline rings are linked by a C—H⋯π inter­action. The degredation product, N,N'-[(6-phenyl-6,7-di­hydro-5H-pyrrolo­[3,4-b]pyrazine-2,3-di­yl)bis(methyl­ene)]bis­(N-methyl­aniline), C28H29N5, (III), was obtained several times by reacting (II) with different metal salts. Here, the 6-phenyl ring is almost coplanar with the planar pyrrolo­[3,4-b]pyrazine unit (r.m.s. deviation = 0.029 Å), with a dihedral angle of 4.41 (10)° between them. The two N-meth­yl­aniline rings are inclined to the planar pyrrolo­[3,4-b]pyrazine unit by 88.26 (10) and 89.71 (10)°, and to each other by 72.56 (13)°. There are also weak intra­molecular C—H⋯N hydrogen bonds present involving the pyrazine ring and the two N-methyl­aniline groups. In the crystal of (I), there are no significant inter­molecular contacts present, while in (II) mol­ecules are linked by a pair of C—H⋯π inter­actions, forming chains along the c-axis direction. In the crystal of (III), mol­ecules are linked by two pairs of C—H⋯π inter­actions, forming inversion dimers, which in turn are linked by offset π–π inter­actions [inter­centroid distance = 3.8492 (19) Å], forming ribbons along the b-axis direction.

Structures of substrate- and product-bound forms of a multi-domain copper nitrite reductase shed light on the role of domain tethering in protein complexes

Copper-containing nitrite reductases (CuNiRs) are found in all three kingdoms of life and play a major role in the denitrification branch of the global nitro­gen cycle where nitrate is used in place of di­oxy­gen as an electron acceptor in respiratory energy metabolism. Several C- and N-terminal redox domain tethered CuNiRs have been identified and structurally characterized during the last decade. Our understanding of the role of tethered domains in these new classes of three-domain CuNiRs, where an extra cytochrome or cupredoxin domain is tethered to the catalytic two-domain CuNiRs, has remained limited. This is further compounded by a complete lack of substrate-bound structures for these tethered CuNiRs. There is still no substrate-bound structure for any of the as-isolated wild-type tethered enzymes. Here, structures of nitrite and product-bound states from a nitrite-soaked crystal of the N-terminal cupredoxin-tethered enzyme from the Hyphomicrobium denitrificans strain 1NES1 (Hd1NES1NiR) are provided. These, together with the as-isolated structure of the same species, provide clear evidence for the role of the N-terminal peptide bearing the conserved His27 in water-mediated anchoring of the substrate at the catalytic T2Cu site. Our data indicate a more complex role of tethering than the intuitive advantage for a partner-protein electron-transfer complex by narrowing the conformational search in such a combined system.

Structure of the N-terminal domain of ClpC1 in complex with the antituberculosis natural product ecumicin reveals unique binding interactions

The biological processes related to protein homeostasis in Mycobacterium tuberculosis, the etiologic agent of tuberculosis, have recently been established as critical pathways for therapeutic intervention. Proteins of particular interest are ClpC1 and the ClpC1–ClpP1–ClpP2 proteasome complex. The structure of the potent antituberculosis macrocyclic depsipeptide ecumicin complexed with the N-terminal domain of ClpC1 (ClpC1-NTD) is presented here. Crystals of the ClpC1-NTD–ecumicin complex were monoclinic (unit-cell parameters a = 80.0, b = 130.0, c = 112.0 Å, β = 90.07°; space group P21; 12 complexes per asymmetric unit) and diffracted to 2.5 Å resolution. The structure was solved by molecular replacement using the self-rotation function to resolve space-group ambiguities. The new structure of the ecumicin complex showed a unique 1:2 (target:ligand) stoichiometry exploiting the intramolecular dyad in the α-helical fold of the target N-terminal domain. The structure of the ecumicin complex unveiled extensive interactions in the uniquely extended N-terminus, a critical binding site for the known cyclopeptide complexes. This structure, in comparison with the previously reported rufomycin I complex, revealed unique features that could be relevant for understanding the mechanism of action of these potential antituberculosis drug leads. Comparison of the ecumicin complex and the ClpC1-NTD-L92S/L96P double-mutant structure with the available structures of rufomycin I and cyclomarin A complexes revealed a range of conformational changes available to this small N-terminal helical domain and the minor helical alterations involved in the antibiotic-resistance mechanism. The different modes of binding and structural alterations could be related to distinct modes of action.

An efficient synthesis of 1-aryl­isochromeno[3,4-d][1,2,3]triazol-5(1H)-ones, involving the diazo­tization of 3-amino-4-aryl­amino-1H-isochromen-1-ones in weakly acidic solution, has been developed and the spectroscopic characterization and crystal structures of four examples are reported. The mol­ecules of 1-phenyl­isochromeno[3,4-d][1,2,3]triazol-5(1H)-one, C15H9N3O2, (I), are linked into sheets by a combination of C—H⋯N and C—H⋯O hydrogen bonds, while the structures of 1-(2-methyl­phen­yl)isochromeno[3,4-d][1,2,3]triazol-5(1H)-one, C16H11N3O2, (II), and 1-(3-chloro­phen­yl)isochromeno[3,4-d][1,2,3]triazol-5(1H)-one, C15H8ClN3O2, (III), each contain just one hydrogen bond which links the mol­ecules into simple chains, which are further linked into sheets by π-stacking inter­actions in (II) but not in (III). In the structure of 1-(4-chloro­phen­yl)isochromeno[3,4-d][1,2,3]triazol-5(1H)-one, (IV), isomeric with (III), a combination of C—H⋯O and C—H⋯π(arene) hydrogen bonds links the mol­ecules into sheets. When com­pound (II) was exposed to a strong acid in methanol, qu­anti­tative conversion occurred to give the ring-opened transesterification product methyl 2-[4-hy­droxy-1-(2-methyl­phen­yl)-1H-1,2,3-triazol-5-yl]benzoate, C17H15N3O3, (V), where the mol­ecules are linked by paired O—H⋯O hydrogen bonds to form centrosymmetric dimers.

The coronavirus pandemic has forced many people to work from home, and that includes employees of the federal government. The numbers vary by agency, but at the Social Security Administration, some 53,000 workers are doing so.

Social Security field offices are closed. But the shutdown hasn't stopped the agency from processing claims for new benefits and appeals of benefit denials. And according to statistics that the SSA sent its workers, the agency has been doing so at a faster pace than before.

"Telework is proving a great boon to the service Social Security provides to the American people," says Ralph deJuliis, who works at the SSA's office in Tulsa, Okla. "We are getting the checks to people faster and quicker."

DeJuliis is president of the American Federation of Government Employees Council 220, which represents many Social Security workers. And he says he hopes the SSA will continue allowing employees to work from home.

Telework, he says, is "good for the employees, good for the public. We've got the work done. We kept the public out of harm's way because, let's face it, we deal with mostly people who are old or disabled. They are at the highest risk."

According to deJuliis, the SSA has found that its backlog of pending cases has fallen by 11% since March 23, when the agency instituted wide-scale telework, and that calls from recipients are answered more quickly.

Isabel Sawhill, a senior fellow at the Brookings Institution, says it's not surprising that productivity is up.

"Actually, there are studies that have been done, including studies in government agencies — small-scale studies, to be sure — but they have shown that productivity does rise when people get to work from home," she says.

Jeff Neal, a former head of human resources at the Department of Homeland Security, says it's not surprising that people become more productive when they work from home.

"The really good workers might be sitting there at their home desk, wherever that is," he says. "And they're supposed to stop at 5 o'clock, and they look at their watch or their computer and they realize it's 7 o'clock and they've still been working, because they get into things and they start getting stuff done and they just keep on going."

It's unclear how many federal workers across the government are teleworking. According to the most recent statistics, from two years ago, 42% of the some 2.1 million government employees were eligible to telework, although only about half of those were in fact working from home.

The Trump administration had been hostile to teleworking, Neal says, because in its view it sees it as a benefit to federal workers. But Neal says it's also a benefit to taxpayers.

"If people view it as what it really is, which is something that is in the interest of the federal government to have, then they would continue it because it helps them hire. It helps them retain people," he says. "And most importantly, it helps them remain operational during a national emergency. So it's a very good thing."

And Sawhill at Brookings says she expects teleworking will continue to increase both in government and the private sector after the coronavirus crisis ends.

"This experience has showed us that we can get work done at home and that we can meet people's needs, the public's needs, by doing so," she says. "That doesn't mean there aren't lots of downsides. But overall, I think this is a trend that is going to accelerate sharply as a result of this recent experience."

The federal government has not given any guidance as to when it expects all federal workers to return to their offices.

The SSA issued a statement saying it continues to monitor the COVID-19 situation across the nation, promising that when it does reopen offices, it will provide a safe environment for the people it serves and its employees.

A moment Hollywood's been waiting a while for will take place... in Hollywood. 

A ceremony is planned for Thursday morning at the Chinese Theater where Governor Jerry Brown will sign the "California Film and Television Job Retention and Promotion Act" into law.

The bill - also known  as AB 1839 — will more than triple the funding for California's film and television production tax credit program. 

The push to expand and enhance the tax credit program has been going on for more than a year. In August of 2013, Los Angeles Mayor Eric Garcetti used the term "state of emergency" to characterize the flight of film and television production to other states and countries. Garcetti is expected to speak at the ceremony. 

Los Angeles-area Assemblymen Mike Gatto and Raul Bocanegra are also expected to be on hand. They introduced AB 1839 in February and moved it strategically through the legislature in Sacramento. While there were few vocal opponents of expanding the tax credit program, the big question was by how much. Many supporters hoped to see the annual pot raised from the current $100 million to at least $400 million, but an exact dollar amount wasn't specified until very late in the legislative process.

In April, the state Legislative Analyst's Office released its hard look at the current tax credit program, pointing out that the state is only getting back 65 cents in tax revenues for every dollar it’s spending on the film and TV subsidy.  The bill to expand the program kept moving.

California's magic number turned out to be $330 million dollars, not as high as chief rival New York State's $420 million per year, but still more than triple California's current offering. Along with the extra cash, AB 1839 also changes the way the tax credit program will be administered.   Rather than using a one-day lottery to determine which productions receive the credit, the state will measure the projects based on their potential to create jobs.   A project that overestimates that potential could be penalized.

A recent study has described a simple method to screen the toxicity of materials used in consumer products. Using utility meter products as examples, the study found, for example, that stainless steel and polyvinyl chloride (PVC) had high toxicity potentials and suggests less toxic, but equally effective and priced, alternatives that could be used instead.

Researchers undertook life-cycle assessments of products including detergents, soaps and toilet cleaners, and compared their environmental footprints looking at different types of environmental damage.

Low-carbon product design: look at the parts to improve the whole

Researchers have developed a method to identify the parts that contribute the most to a product's greenhouse gas (GHG) emissions and evaluate alternative design solutions. When applied to a LCD television, it showed that GHG emissions could be cut by 36 per cent by using alternative parts to those currently used.
