WASHINGTON, D.C. -- The National Association of Broadcasters announced the promotions of four employees in its Government Relations, Technology and Legal and Regulatory Affairs departments to senior vice president, effective July 1.

Improving the situation of Roma in Ukraine by enhancing co-operation between Roma civil society organizations and national and regional authorities was the focus of a roundtable meeting organized by the OSCE Office for Democratic Institutions and Human Rights (ODIHR) and the Ministry of Culture of Ukraine in Kyiv on 24 November 2015.

Some 40 participants from Ukrainian national and regional authorities, Roma civil society and the international community, discussed the challenges faced by Roma in the areas of education, employment, housing and policing.

“Further efforts are needed to implement a strong anti-discrimination approach in all policies targeting the integration and protection of Roma and, in particular, of Roma women,” said Mirjam Karoly, ODIHR Senior Adviser on Roma and Sinti Issues. “Measurable progress at the local level can only be achieved if policies are needs-based and have concrete objectives that are matched with measurable indicators and realistic budgets.”

Andriy Vitrenko, Deputy Minister of Culture of Ukraine for European Integration, stated that the Ministry was tasked in 2013to facilitate the implementation of the Strategy on the Protection and Integration of the Roma National Minority into Ukrainian Society up to 2020. “In order to achieve the goals set out in the Strategy, close collaboration among a number of ministries at the national level and good co-ordination with regional and local level authorities are needed.”

During the meeting, the participants emphasized that many Roma lack personal documents, hindering them from exercising their fundamental, social and political rights.

Volodomyr Kondur, Chairperson of the Roma Coalition, said: “We welcome the adoption of national and regional policy documents addressing the problems Roma face, but we want to see effective and sustainable results and ask for ensuring involvement of the Roma civil society in all stages of policy implementation and evaluation.”

The roundtable meeting was organized as a follow-up to ODIHR’s Situation Assessment Report on Roma in Ukraine and the Impact of the Current Crises and in line with the 2003 OSCE Action Plan on Roma and Sinti.

Akt3 regulates mitochondrial content in endothelial cells through the inhibition of PGC-1α nuclear localization and is also required for angiogenesis. However, whether there is a direct link between mitochondrial function and angiogenesis is unknown. Here we show that Akt3 depletion in primary endothelial cells results in decreased uncoupled oxygen consumption, increased fission, decreased membrane potential, and increased expression of the mitochondria-specific protein chaperones, HSP60 and HSP10, suggesting that Akt3 is required for mitochondrial homeostasis. Direct inhibition of mitochondrial homeostasis by the model oxidant paraquat results in decreased angiogenesis, showing a direct link between angiogenesis and mitochondrial function. Next, in exploring functional links to PGC-1α, the master regulator of mitochondrial biogenesis, we searched for compounds that induce this process. We found that, sildenafil, a phosphodiesterase 5 inhibitor, induced mitochondrial biogenesis as measured by increased uncoupled oxygen consumption, mitochondrial DNA content, and voltage-dependent anion channel protein expression. Sildenafil rescued the effects on mitochondria by Akt3 depletion or pharmacological inhibition and promoted angiogenesis, further supporting that mitochondrial homeostasis is required for angiogenesis. Sildenafil also induces the expression of PGC-1 family member PRC and can compensate for PGC-1α activity during mitochondrial stress by an Akt3-independent mechanism. The induction of PRC by sildenafil depends upon cAMP and the transcription factor CREB. Thus, PRC can functionally substitute during Akt3 depletion for absent PGC-1α activity to restore mitochondrial homeostasis and promote angiogenesis. These findings show that mitochondrial homeostasis as controlled by the PGC family of transcriptional activators is required for angiogenic responses.

On December 3, a federal appeals court ruled against one of the FDA’s untouchable restrictions on industry—thou shalt not promote the off-label use of pharmaceutical products. An industry that is little interested in constitutional law suddenly finds itself talking about the First Amendment and whether, and on what grounds, the case will be appealed. Meantime, the court’s decision left FDA Matters torn between cheering and booing. Patients are poorly served if their doctor is prescribing drugs without being able to tap into all sources of relevant knowledge. However, permitting off-label promotion undercuts the incentive for companies to thoroughly investigate the safety and efficacy of a drug for a second or third use.

Improving Enrollment in Pediatric Clinical Trials

Logistically, ethically, and emotionally, involving children in medical research is greatly different from the same research in adults. Some of the toughest clinical trials I've worked on, across a number of therapeutic areas, have been pediatric ones. They challenge you to come up with different approaches to introducing and explaining clinical research – approaches that have to work for doctors, kids, and parents simultaneously.

On Thursday June 27, Don Sickler, one of my team members, will be chairing a session titled “Parents as Partners: Engaging Caregivers for Pediatric Trials”. It should be a good session.

Joining Don are 2 people I've had the pleasure of working with in the past. Both of them combine strong knowledge of clinical research with a massive amount of positive energy and enthusiasm (no doubt a big part of what makes them successful).

However, they also differ in one key aspect: what they work on. One of them – Tristen Moors from Hyperion Therapeutics - works on an ultra-rare condition, Urea Cycle Disorder, a disease affecting only a few hundred children every year. On the other hand, Dr. Ann Edmunds is an ENT working in a thriving private practice. I met her because she was consistently the top enroller in a number of trials relating to tympanostomy tube insertion. Surgery to place “t-tubes” is one of the most common and routine outpatients surgeries there is, with an estimated half million kids getting tubes each year.

Each presents a special challenge: for rare conditions, how do you even find enough patients? For routine procedures, how do you convince parents to complicate their (and their children’s) lives by signing up for a multi-visit, multi-procedure trial?

Ann and Tristen have spent a lot of time tackling these issues, and should have some great advice to give.
