It gave me a chance to read for once and not have to think about the technical merits about the subject material being discussed. At first I thought it might just be a pure photography book, nothing but pictures and little text. When it arrived I realized that there was a fair amount of story that surrounded the photos. The story about the Serengeti was very interesting and unique and an important part of the book. Book Title: Serengeti – The Eternal BeginningAuthor: Boyd NortonISBN: 978-1-55591-593-3Publisher: Fulcrum PublishingHard cover - 265 pages The ReviewOne of the first things that struck me about the book was the experience and depth that the author 'Boyd Norton' had with this area of Africa. While its not exactly clear, it appears that he has spent more than 25 years visiting and getting to know this area intimately about the life and its nuances in the Serengeti. The book is not only about Serengeti as its title suggests but also includes the Maswa Game Reserve, Ngorongoro conservation area, Loliondo Game controlled area and the Masai Mara National Reserve. As with most geographical features, country boundaries do not define its scope. This geographical area defines the coverage, for the most part of the 'Great Migration' that we are all familiar with. Where thousands upon thousand of larger African animals such as wildebeests are seen to run for thousands of miles in search of food as the weather patterns change. While the predators lie in wait for the dinner train to run by. The author discusses the parks earliest beginnings when there wasn’t a park but only early explorers and big game hunters. The early problems all countries seem to have when they want to control activities within an area with competing demands from indigenous groups, expanding farming base and other users of the resources. This is not only a book about the big and small animals that are found here but about the life of the people who have lived here for aeons. It’s interesting to read how the locals, weather patterns and what was a perfect balance of nature all worked together to create a faultless ecosystem. Especially when you think of this place as the dawn of human kind some 3-4 million year ago. The author’s writings give a life to photos that are not apparent directly from the photos. As an example, he discusses how certain rock outcrops called Gol Kopjes seems to have extra gravity that applies only to the big cats. In that whenever they arrive on these outcrops the local rock gravity immediately pulls them flat on the rock and holds them for hours. The book covers the Great Migration when it seems most of the life in the Serengeti is on the move and about the carnivores who patiently wait for their turn at the Great Feast or migration as we call it. The next chapter is about the lions which is a fascinating subculture all unto itself. Who gets to lead, who eats and who gets to live with the group. The 4th chapter is about Ngorongoro which is the remains of a huge volcano called a caldera, like a giant cauldron. In this crater life is different due to its unique ecosystem. The next chapter deals with creatures both large and small and how like any balanced system they each depend on each other for survival. From control of foliage to providing food for the predators. If your visit is to only see the big game animals then you are missing so much more about the life in the Serengeti. The next chapters deal each with the other more famous of the big African animals such as the Rhino, Leopard, Cheetah and Elephant and how they live and communicate. There are personal stories around each encounter with the wilds of Africa and this is what makes this book more exciting than just a lot of photos. There is Anna who can talk Rhino talk and the perception that they are stupid is really outdated. There are also the Acadia trees who can communicate when the giraffes arrive for dinner. Recommended Audience This book doesn’t tell you how to use your camera or take better photographs, which I'm glad as sometimes we just need stories that surround images we are seeing. Inspiration rarely comes from a technical how-to-manual. I now know that if I were to go on an African photo safari I would not expect or want a 1 or 2 day quick tour and photo-op around the park. The book has conveyed to me that there is just so much more to see and experience that it can only be fully appreciated if you take the time to watch and wait. When there, live in the flow of the Serengeti's life. If not for yourself then this would make a great gift for any friend who has been talking about going on an excursion to see those big game animals of Africa. I found every photo crisp and clear and representative of the its natural environment. The photos are taken as found and therefore the lighting is natural and not staged. Nor are the images manipulated to give extra punch as I am prone to do. All are color prints with no B&W. Since most of the animals by nature's design want to blend in with their surrounding there is not always a lot of high contrast, but they all stand out in the photos. The photos are all of the caliber that you would find in the National Geographic magazine. The front cover image is representative of the quality of images whether they are up close or distant landscapes contained within the book. Full Article books

ginni M8R tropomyosin mutation disrupts actin binding and filament regulation: The beginning affects the middle and end [Molecular Bases of Disease] By www.jbc.org Published On :: 2020-12-11T00:06:20-08:00 Dilated cardiomyopathy (DCM) is associated with mutations in cardiomyocyte sarcomeric proteins, including α-tropomyosin. In conjunction with troponin, tropomyosin shifts to regulate actomyosin interactions. Tropomyosin molecules overlap via tropomyosin–tropomyosin head-to-tail associations, forming a continuous strand along the thin filament. These associations are critical for propagation of tropomyosin's reconfiguration along the thin filament and key for the cooperative switching between heart muscle contraction and relaxation. Here, we tested perturbations in tropomyosin structure, biochemistry, and function caused by the DCM-linked mutation, M8R, which is located at the overlap junction. Localized and nonlocalized structural effects of the mutation were found in tropomyosin that ultimately perturb its thin filament regulatory function. Comparison of mutant and WT α-tropomyosin was carried out using in vitro motility assays, CD, actin co-sedimentation, and molecular dynamics simulations. Regulated thin filament velocity measurements showed that the presence of M8R tropomyosin decreased calcium sensitivity and thin filament cooperativity. The co-sedimentation of actin and tropomyosin showed weakening of actin-mutant tropomyosin binding. The binding of troponin T's N terminus to the actin-mutant tropomyosin complex was also weakened. CD and molecular dynamics indicate that the M8R mutation disrupts the four-helix bundle at the head-to-tail junction, leading to weaker tropomyosin–tropomyosin binding and weaker tropomyosin–actin binding. Molecular dynamics revealed that altered end-to-end bond formation has effects extending toward the central region of the tropomyosin molecule, which alter the azimuthal position of tropomyosin, likely disrupting the mutant thin filament response to calcium. These results demonstrate that mutation-induced alterations in tropomyosin–thin filament interactions underlie the altered regulatory phenotype and ultimately the pathogenesis of DCM. Full Article

A three-week event featuring that format starts tomorrow, November 12. But there’s a twist: you won’t be able to select Kiriko or Sombra, or battle it out with an additional player on each side on Push maps just yet. That’s because in Overwatch 2’s first real taste of 6v6, Blizzard is taking us all the way back to the beginning with a limited-time mode called Overwatch: Classic. You will be able to experience Overwatch almost exactly as it was upon its May 2016 debut. That means you can choose from the first 21 heroes, who all have their original kits and abilities. That means Hanzo loses his Lunge jump but regains his dreaded Scatter Arrow, Bastion and Torbjorn are vastly different than they are now and Cassidy's Flashbang once again stun locks enemies for a moment. Symmetra reverts to being a support who can teleport allies almost anywhere on the map from the spawn room, while Mercy can will once again bring five dead teammates back to life. Ultimate abilities will charge up faster too. In addition, just like in Overwatch for a brief period at the very beginning, there are initially no limits on hero selection. So if you and your teammates want to run with a composition of four Winstons and two Lucios, have at it. However, this will only apply for the first few days, after which Blizzard will apply the single hero limit rule for the rest of the event. Games will take place under the Quick Play ruleset, rather than the Competitive format. The original 12 maps will be available too — including the assault maps that Blizzard retired from the main modes during the transition to Overwatch 2. While assault maps are still available in the Arcade and custom games, you'll once again be dealing with the notorious choke points of the otherwise gorgeous Hanamura, Temple of Anubis and Volskaya Industries. Blizzard Entertainment Things won't be exactly as they were in May 2016, however. Original maps that have seen major reworks over the years — Dorado, Numbani, Route 66 and Watchpoint: Gibraltar — will appear as they are in the current game. You'll only be able to use the original default Overwatch skins and no, there are no loot boxes. The user interface remains the same too, which hopefully means the ping system will still be in place. Blizzard doesn't plan for this to be a one-and-done deal. There will be other Overwatch: Classic events in the future, focusing on various moments in the game's history, like the infamous triple-tank, triple-support GOATS meta. This limited-time mode is also separate from the other 6v6 tests Blizzard plans to run in the coming months as it looks to measure players' interest in that format and garner feedback. There's a good chance that this limited-time mode will bring some lapsed players back into the mix, even just for a sip of nostalgia. I first played Overwatch several months after its debut, so it'll be fun to see roughly how the game felt at the very beginning. I will be instalocking Mei every match so I can remember what it's like to freeze an opponent before giving them a cheeky wave and firing an icicle into their skull. President Recep Tayyip Erdogan has responded with an iron fist to last Friday’s failed military coup attempt in Turkey by detaining, dismissing or suspending, so far, 60,000 military officers, police and intelligence officials, judges, teachers, academics and civil servants, and imposing a widespread travel ban and a three-month state of emergency. He is vowing to reintroduce the death penalty, abolished in 2004 as part of reforms required for opening EU accession negotiations.This uncompromising approach in the post-coup period will have profound negative implications on Turkey’s domestic politics, security and foreign policy in the foreseeable future to the detriment of its stability and prosperity.Fractured politicsErdogan’s indifference to the unprecedented political unity against the coup is, regretfully, a missed opportunity to dilute the deepening polarization and divisiveness bedeviling Turkish politics. His determination to use the putsch to consolidate political power in the presidency and to erode or eliminate the secular character of the Turkish state by means of a new constitution will widen the ideological and ethnic divide between, respectively, secular and conservative Turks and Turks and Kurds. Just a few months ago, Ismail Kahramam, speaker of the Turkish parliament and Erdogan ally, exhorted that ‘secularism cannot feature in the new [religious] constitution’.His policies and rhetoric, in other words, will undermine even more the almost imperceptible presence of ‘interpersonal trust’ in Turkish society - the willingness of one party to rely on the actions of another party – seen as incongruent with a robust polity and cohesive society. According to a 2010 OECD survey Turkey’s levels of interpersonal trust are considerably lower than OECD averages and it stands out among the 20 surveyed countries as the only one where higher educational attainment correlates with lower feelings of trust. That posture can only breed even more discord and mistrust between the different segments of the Turkish electorate and entrench personality-based and top-down politics, the root cause of political turmoil in Turkey.Diminished state capacityTurkey’s NATO partners fear that the purges of experienced military and security personnel have the potential to diminish its capability to thwart the threat posed by Islamic State of Iraq and Syria (ISIS) and other militant groups and to better manage its long and porous borders with Syria and Iraq. Thus far, Turkish authorities have incarcerated nearly one-third of Turkey’s senior military commanders and more than 7,000 police and intelligence officials. This constitutes a major loss of expertise and institutional memory at a time of heightening security challenges. After all, Turkey witnessed 14 bomb attacks over the last year, many of them carried out by ISIS or the separatist Kurdistan Workers’ Party (PKK).Similarly, the removal of tens of thousands of school teachers, both in private and state schools, university academics and education ministry officials will severely disrupt the provision of adequate educational services to enable future generations to succeed in an increasingly complex global economic environment. This ‘cleansing’ operation did not spare even the elite and renowned state and private universities considered bastions of liberalism and cosmopolitan values in Turkey.In all probability, the government’s replacements of key staff with less qualified loyalists will rupture the institutional integrity and professionalism of the military establishment and the state institutions. Such a hollowing out process was already underway prior to the coup but post-coup decision-making has greatly accelerated the speed. Sadly, under the best case scenario, it will take Turkey years, if not decades, to restore a modicum of rule of law and public services’ delivery at pre-coup standards to which the Turkish citizenry have been accustomed.Foreign policy challengesErdogan’s endorsement of the death penalty might signal the end of Turkey’s (already nearly non-existent) EU accession prospects and a more troubled relationship with Europe and the US. He was, before the coup, a prickly and challenging partner for the US and NATO to handle, a recalcitrant member of the US-led anti-ISIS coalition and vociferously against the US cooperation with PKK-affiliated Syrian Kurdish fighters targeting ISIS in northern Syria. After the coup, he will probably become more disagreeable to US and European foreign policy and security objectives.His disagreeability will probably extend to Turkey’s deal with the EU to stem the flow of Syrian migrants across the Aegean Sea and Greece into mainland Europe, which looks increasingly unsustainable. A pugnacious Erdogan may utilize the forthcoming EU refusal to abolish visas for Turkish travellers to the Schengen borderless zone by end-October to wring out more concessions from an Erdogan-sceptical Europe. Despite their exasperation, they should decipher from his rapprochement with Israel and Russia that he tends to compromise with muscular diplomacy as opposed to diplomatic niceties. Turkey will be so convulsed and self-absorbed by internal political machinations and its security and military capabilities so compromised that it cannot afford to deploy sizeable assets to promote regime change in Damascus. Syrian President Bashar al-Assad and his Russian and Iranian backers are, naturally, the prime beneficiaries while the armed largely Sunni opposition are the biggest losers. Arguably, Assad must now feel very secure in power and confident that he will enlarge his territorial acquisitions at the expense of the Sunni groups. Equally, the Syrian Kurds will seek to strengthen and, perhaps, extend the quasi-autonomous zone along the Turkey−Syria border commensurate with Turkey’s declining influence in the Syrian quagmire.Europe’s lessonTurkey is a bitter testimony to the ill-effects of sacrificing progressive values to political expediency, fear and interests. German Chancellor Angela Merkel and former French president Nicolas Sarkozy demonstrated a lack of strategic foresight by stymying Turkey’s desire to join the EU in 2005. Had the EU engaged Turkey in a credible accession process, however arduous it may have been, the coup would probably have never occurred. Turkish political leaders would have been forced to implement deeper and wider reforms to strengthen democracy, secularism, human rights and a functioning market economy.
