Sara is a journalist who wholeheartedly throws herself into her work as a journalist for a newspaper in Juarez. Mexico however is a country where journalists sometimes do their work under threats to themselves and their families and going to the police is not always a good idea. Sara soon finds that if she wants to pursue this particular story she could be putting the lives of herself, her younger brother Emiiano and their mother in danger.

Emiliano is a soccer star at his high school and in addition to this he is also a member of a school group called Jiparis who do hikes through the desert in order to build character in the young men, some of whom were involved in unsavory activities before joining the group.

One part of the Jipari pledge goes , "I will be honest with myself and others". This is easier said than done especially in a city like Juarez. One of the characters tells Emiliano, "everything is a spiderweb" and the speed with which he is enveloped in said web is astounding. Emiliano tells himself that he wants to help his family and friends out but are those his real reasons? Like any teen he wants to be seen as cool and there is also the small matter of a girl he wants to impress.

The first part of the book is told from each character's viewpoint and the author weaves the tale together in a very credible way showing how circumstances

Make no mistake, Disappeared is not a peaches and cream, hunky dory teen novel. It is a gritty and very realistic novel with a ripped from the headlines quality to it. The city of Juarez and the violence there doesn't dominate the headlines as it did a few years ago, but Stork's novel is a timely reminder that evil still exists and that it takes many people working in tandem to defeat it.

With his swift seizure of Crimea, Vladimir Putin looked to be playing a strong hand in the stand-off over Ukraine’s future. But recent events have shown the brittleness of his power in the face of international condemnation and the calm determination of Ukrainians.

On 3 March, 14 members of the UN Security Council denounced the 15th member, Russia, in unprecedentedly strong terms for the violation of Ukraine’s territorial integrity and use of military intimidation. Even China followed suit.

The Russian ambassador, Vitaly Churkin, who is used to getting his way in the Security Council, was dumbfounded. With surprising confidence, Churkin had asked for the emergency discussion over Ukraine. Each of his arguments was swiftly dismissed as inacceptable with regard to international law, or in bad faith. He got his way with a shameful nyet to Security Council resolutions on Syria, but not here.

The Russian state has been facing growing criticism from many governments and multilateral organizations since it launched an armed incursion into Crimea. NATO, the OSCE, the EU and the Council of Europe have condemned Russia’s resort to military force in Crimea. Sanctions are being discussed very seriously. And the economic and financial backlash is hurting the Russian currency, treasury and major corporations. The Kremlin has stumbled on international legal norms, which it wrongly believed it could interpret in its own free manner, with the support of China.

On 4 March, President Putin chose to express himself on Ukraine, at last. He looked nervous even though he was addressing a small and carefully selected group of young journalists for a ‘discussion-like press conference’. He told an odd story of the war he had threatened everyone with, but had never intended to wage. He repeated arguments that Churkin had already lost in New York the day before. And, with his never-abating desire to rewrite recent history, he condemned both Ukraine’s independence and the Orange Revolution of 2004.

He kept changing his mind about deposed Ukrainian President Viktor Yanukovich’s position. He first said that Yanukovich was ‘politically dead’, but later justified Russian military ‘protection’ of Crimea’s population with Yanukovich’s supposed written request to Moscow on 1 March. Such a pretext is less convincing to the US and Europe by the day, just like Yanukovych’s use of a hastily passed anti-terrorism law to attempt to justify his order to shoot at civilian protesters on the Maidan. Today, Yanukovich is a former despot on the run. The Kremlin’s propaganda has backfired.

Negotiation is now beginning to reassert itself over confrontation. The Russian and Ukrainian governments have just renewed a fragile communication line. Kyiv and Simferopol are setting up a commission to discuss a common strategy out of the military standoff, and the status of the autonomous republic of Crimea in the Ukrainian state. The war scare is not quite over, but it now looks clear that Moscow bears the responsibility for raising the stakes all the way to the brink of armed struggle, with civilians as potential victims. Most powers, together with international organizations, agree that Russia’s behaviour has been dangerous and that the new interim Ukrainian government is legitimate.

The priority, now that armed violence is abating, is quick and robust support to the Ukrainian economy and society. And, as a necessary corollary, Western governments will have to devote time to helping the Russian president save face and stay quiet behind the Kremlin walls.

Structures of the MHC-I molecule BF2*1501 disclose the preferred presentation of an H5N1 virus-derived epitope [Protein Structure and Folding]

Lethal infections by strains of the highly-pathogenic avian influenza virus (HPAIV) H5N1 pose serious threats to both the poultry industry and public health worldwide. A lack of confirmed HPAIV epitopes recognized by cytotoxic T lymphocytes (CTLs) has hindered the utilization of CD8+ T-cell–mediated immunity and has precluded the development of effectively diversified epitope-based vaccination approaches. In particular, an HPAIV H5N1 CTL-recognized epitope based on the peptide MHC-I–β2m (pMHC-I) complex has not yet been designed. Here, screening a collection of selected peptides of several HPAIV strains against a specific pathogen-free pMHC-I (pBF2*1501), we identified a highly-conserved HPAIV H5N1 CTL epitope, named HPAIV–PA123–130. We determined the structure of the BF2*1501–PA123–130 complex at 2.1 Å resolution to elucidate the molecular mechanisms of a preferential presentation of the highly-conserved PA123–130 epitope in the chicken B15 lineage. Conformational characteristics of the PA123–130 epitope with a protruding Tyr-7 residue indicated that this epitope has great potential to be recognized by specific TCRs. Moreover, significantly increased numbers of CD8+ T cells specific for the HPAIV–PA123–130 epitope in peptide-immunized chickens indicated that a repertoire of CD8+ T cells can specifically respond to this epitope. We anticipate that the identification and structural characterization of the PA123–130 epitope reported here could enable further studies of CTL immunity against HPAIV H5N1. Such studies may aid in the development of vaccine development strategies using well-conserved internal viral antigens in chickens.

Polarization of protease-activated receptor 2 (PAR-2) signaling is altered during airway epithelial remodeling and deciliation [Immunology]

Protease-activated receptor 2 (PAR-2) is activated by secreted proteases from immune cells or fungi. PAR-2 is normally expressed basolaterally in differentiated nasal ciliated cells. We hypothesized that epithelial remodeling during diseases characterized by cilial loss and squamous metaplasia may alter PAR-2 polarization. Here, using a fluorescent arrestin assay, we confirmed that the common fungal airway pathogen Aspergillus fumigatus activates heterologously-expressed PAR-2. Endogenous PAR-2 activation in submerged airway RPMI 2650 or NCI–H520 squamous cells increased intracellular calcium levels and granulocyte macrophage–colony-stimulating factor, tumor necrosis factor α, and interleukin (IL)-6 secretion. RPMI 2650 cells cultured at an air–liquid interface (ALI) responded to apically or basolaterally applied PAR-2 agonists. However, well-differentiated primary nasal epithelial ALIs responded only to basolateral PAR-2 stimulation, indicated by calcium elevation, increased cilia beat frequency, and increased fluid and cytokine secretion. We exposed primary cells to disease-related modifiers that alter epithelial morphology, including IL-13, cigarette smoke condensate, and retinoic acid deficiency, at concentrations and times that altered epithelial morphology without causing breakdown of the epithelial barrier to model early disease states. These altered primary cultures responded to both apical and basolateral PAR-2 stimulation. Imaging nasal polyps and control middle turbinate explants, we found that nasal polyps, but not turbinates, exhibit apical calcium responses to PAR-2 stimulation. However, isolated ciliated cells from both polyps and turbinates maintained basolateral PAR-2 polarization, suggesting that the calcium responses originated from nonciliated cells. Altered PAR-2 polarization in disease-remodeled epithelia may enhance apical responses and increase sensitivity to inhaled proteases.
