Chylomicronemia from GPIHBP1 autoantibodies [Reviews]

Some cases of chylomicronemia are caused by autoantibodies against glycosylphosphatidylinositol-anchored HDL binding protein 1 (GPIHBP1), an endothelial cell protein that shuttles LPL to the capillary lumen. GPIHBP1 autoantibodies prevent binding and transport of LPL by GPIHBP1, thereby disrupting the lipolytic processing of triglyceride-rich lipoproteins. Here, we review the "GPIHBP1 autoantibody syndrome" and summarize clinical and laboratory findings in 22 patients. All patients had GPIHBP1 autoantibodies and chylomicronemia, but we did not find a correlation between triglyceride levels and autoantibody levels. Many of the patients had a history of pancreatitis, and most had clinical and/or serological evidence of autoimmune disease. IgA autoantibodies were present in all patients, and IgG4 autoantibodies were present in 19 of 22 patients. Patients with GPIHBP1 autoantibodies had low plasma LPL levels, consistent with impaired delivery of LPL into capillaries. Plasma levels of GPIHBP1, measured with a monoclonal antibody–based ELISA, were very low in 17 patients, reflecting the inability of the ELISA to detect GPIHBP1 in the presence of autoantibodies (immunoassay interference). However, GPIHBP1 levels were very high in five patients, indicating little capacity of their autoantibodies to interfere with the ELISA. Recently, several GPIHBP1 autoantibody syndrome patients were treated successfully with rituximab, resulting in the disappearance of GPIHBP1 autoantibodies and normalization of both plasma triglyceride and LPL levels. The GPIHBP1 autoantibody syndrome should be considered in any patient with newly acquired and unexplained chylomicronemia.

EphB2 Signaling Is Implicated in Astrocyte-Mediated Parvalbumin Inhibitory Synapse Development

Impaired inhibitory synapse development is suggested to drive neuronal hyperactivity in autism spectrum disorders (ASD) and epilepsy. We propose a novel mechanism by which astrocytes control the development of parvalbumin (PV)-specific inhibitory synapses in the hippocampus, implicating ephrin-B/EphB signaling. Here, we utilize genetic approaches to assess functional and structural connectivity between PV and pyramidal cells (PCs) through whole-cell patch–clamp electrophysiology, optogenetics, immunohistochemical analysis, and behaviors in male and female mice. While inhibitory synapse development is adversely affected by PV-specific expression of EphB2, a strong candidate ASD risk gene, astrocytic ephrin-B1 facilitates PV->PC connectivity through a mechanism involving EphB signaling in PV boutons. In contrast, the loss of astrocytic ephrin-B1 reduces PV->PC connectivity and inhibition, resulting in increased seizure susceptibility and an ASD-like phenotype. Our findings underscore the crucial role of astrocytes in regulating inhibitory circuit development and discover a new role of EphB2 receptors in PV-specific inhibitory synapse development.

HBM3E: All About Bandwidth

The rapid rise in size and sophistication of AI/ML training models requires increasingly powerful hardware deployed in the data center and at the network edge. This growth in complexity and data stresses the existing infrastructure, driving the need for new and innovative processor architectures and associated memory subsystems. For example, even GPT-3 at 175 billion parameters is stressing the bandwidth, capacity, training time, and power of the most advanced GPUs on the market.

To this end, Cadence has shown our HBM3E memory subsystem running at 12.4Gbps at nominal voltages, demonstrating the PHY’s robustness and performance margin. The production version of our latest HBM3E PHY supports DRAM speeds of up to 10.4Gbps or 1.33TB/s per DRAM device. This speed represents a >1.6X bandwidth increase over the previous generation, making it ideal for LLM training.

Cadence has been the HBM performance leader since 2021, when we announced our first 8.4Gbps HBM3E PHY supporting >1TB/s of memory bandwidth per HBM DRAM. Customers building advanced AI processors have used this speed while building margin into their systems. Recall that HBM3E is a 3D stacked DRAM with 1024-bit wide data (16 64-bit channels). While this wide data bus enables high data transfer, routing these signals requires interposer technology (2.5D) capable of routing close to 2000 signals (data and control), including silicon, RDL, and silicon bridges.

The interposer design is critical for the system to operate at these data rates. Cadence provides 2.5D reference designs, including the interposer and package, as part of our standard IP package. As demonstrated in our test silicon, these designs give customers confidence they will meet their memory bandwidth requirements. The reference design is also a good starting point, helping to reduce development time and risk. Our expert SI/PI and system engineers work closely with customers to analyze their channels to ensure the best system performance.

Even as HBM3E delivers the highest memory bandwidth today, the industry keeps pushing forward. JEDEC recently announced that HBM4the next version of the HBM DRAM standard, is nearing completion. JEDEC calls HBM4 an “evolutionary step beyond the currently published HBM3 standard.” They also claim HBM4 “enhancements are vital for applications that require efficient handling of large datasets and complex calculations.” HBM4 will support AI training applications, high-performance computing (HPC), and high-end graphics cards.

Cadence will continue to push the HBM performance boundaries to ensure designers of these data-intensive systems can take advantage of the highest memory bandwidth available.

UVM Adapter for Pipelined protocols like AHB, AXI etc

Hello,

I have been running this `uvm_reg_hw_reset_seq` sequence for the AHB protocol. My UVM Adapter looks like:


Issue: When I use basic reg.write, my write access are working well, as that is managed by the driver i.e. once adapter gives the packet to the driver, the driver supplies the address and the control signals to the DUT on the first clock cycle and then the write data on the next clock cycle. But when I am performing the read operation, somehow the UVM adapter is reading the data at the same clock cycle where read address + Controls are supplied and this is triggering read failure messages from the `uvm_reg_hw_reset_seq` sequence. What should I modify in the driver/sequencer/adapter so that the UVM adapter can read the data on the next cycle instead of the same clock cycle.

Just FYI: The waveforms of the read operation are correct, it is just the Adapter and the `uvm_reg_hw_reset_seq`. The AHB Driver + AHB Monitor is fully proven and verified to be working correctly.
