New research shows that the drug makes cells in the retina more sensitive to light

25 years ago, pharmacologist M. E. West of the University of the West Indies in Kingston, Jamaica, noted that local fisherman who smoke cannabis or drink rum made with the leaves and stems of the plant had “an uncanny ability to see in the dark,” which enabled them to navigate their boats through coral reefs. “It was impossible to believe that anyone could navigate a boat without compass and without light in such treacherous surroundings,” he wrote after accompanying the crew of a fishing boat one dark night, “[but] I was then convinced that the man who had taken the rum extract of cannabis had far better night vision than I had, and that a subjective effect was not responsible.”

Some of these crew members told West that Moroccan fishermen and mountain dwellers experience a similar improvement after smoking hashish, and in 2002, another research team travelled to the Rif mountains in Morocco to investigate further. They gave a synthetic cannabinoid to one volunteer, and hashish to three more, then used a newly developed piece of kit to measure the sensitivity of their night vision before and after. Confirming West’s earlier report, they found that cannabis improved night vision in all three of their test subjects.

Crystal engineering of exemestane to obtain a co-crystal with enhanced urease inhibition activity

Co-crystallization is a phenomenon widely employed to enhance the physio-chemical and biological properties of active pharmaceutical ingredients (APIs). Exemestane, or 6-methyl­ideneandrosta-1,4-diene-3,17-dione, is an anabolic steroid used as an irreversible steroidal aromatase inhibitor, which is in clinical use to treat breast cancer. The present study deals with the synthesis of co-crystals of exemestane with thio­urea by liquid-assisted grinding. The purity and homogeneity of the exemestane–thio­urea (1:1) co-crystal were confirmed by single-crystal X-ray diffraction followed by thermal stability analysis on the basis of differential scanning calorimetry and thermogravimetric analysis. Detailed geometric analysis of the co-crystal demonstrated that a 1:1 co-crystal stoichiometry is sustained by N—H⋯O hydrogen bonding between the amine (NH2) groups of thio­urea and the carbonyl group of exemestane. The synthesized co-crystal exhibited potent urease inhibition activity in vitro (IC50 = 3.86 ± 0.31 µg ml−1) compared with the API (exemestane), which was found to be inactive, and the co-former (thio­urea) (IC50 = 21.0 ± 1.25 µg ml−1), which is also an established tested standard for urease inhibition assays in vitro. The promising results of the present study highlight the significance of co-crystallization as a crystal engineering tool to improve the efficacy of pharmaceutical ingredients. Furthermore, the role of various hydrogen bonds in the crystal stability is successfully analysed quantitatively using Hirshfeld surface analysis.

Microtubules dynamics is regulated by the plus end-tracking proteins (+TIPs) in cells. End binding protein 1 (EB1) acts as a master regulator in +TIPs networks by targeting microtubule growing ends and recruiting other factors. However, the molecular mechanism of how EB1 binds to microtubule ends with a high affinity remains to be an open question. Using single-molecule imaging, we show that the end-binding kinetics of EB1 changes along with the polymerizing and hydrolysis rate of tubulin dimers, confirming the binding of EB1 to GTP/GDP-Pi tubulin at microtubule growing ends. The affinity of wild-type EB1 to these sites is higher than monomeric EB1 mutants, suggesting that two CH domains in the dimer contribute to the end-binding. Introducing phosphomimicking mutations into the linker domain of EB1 weakens the end-binding affinity and confers a more curved conformation to EB1 dimer without compromising dimerization, suggesting that the overall architecture of EB1 is important for the end-binding affinity. Taken together, our results provide insights into understanding how the high-affinity end-binding of EB1 can be achieved and how this activity may be regulated in cells.

Titanium dioxide-water nanofluids enhance the performance of solar collectors

Adding nanoparticles to water in solar collectors, which are used to capture the sun’s energy, can considerably improve their performance, a recent study on nanofluids has found. The energy efficiency of the collector can be increased by up to 76.6% when using water containing 0.1% by volume of titanium dioxide nanoparticles, compared with water alone.
