Maternal Epigenetic Regulation Contributes to Prevention of Estrogen Receptor-negative Mammary Cancer with Broccoli Sprout Consumption

Cruciferous vegetables have been of special interest due to the rich presence of bioactive compounds such as sulforaphane which show promising potential on cancer prevention and therapy as an epigenetic dietary strategy. Abnormal epigenetic alteration as one of the primary contributors to tumor development is closely related to breast cancer initiation and progression. In the present study, we investigated the effect of dietary broccoli sprouts (BSp), a common cruciferous vegetable, on prevention of estrogen receptor (ER)-negative mammary tumors at three different temporal exposure windows using a spontaneous breast cancer mouse model. Our findings indicate that maternal BSp treatment exhibited profound inhibitory and preventive effects on mammary cancer formation in the nontreated mouse offspring. The BSp diet administered to adult mice also showed suppressive effects on mammary cancer but was not as profound as the maternal BSp preventive effects. Moreover, such protective effects were linked with differentially expressed tumor- and epigenetic-related genes, as well as altered global histone acetylation, DNA methylation, and DNA hydroxymethylation levels. We also found that the expression changes of tumor-related genes were associated with the levels of histone methylation of H3K4 and H3K9 in the gene promoter regions. In addition, BSp-enriched sulforaphane was shown to increase protein expression of tumor suppressor genes such as p16 and p53 and inhibit the protein levels of Bmi1, DNA methyltransferases, and histone deacetylases in ERα-negative breast cancer cell lines. Collectively, these results suggest that maternal exposure to key phytochemicals may contribute to ER-negative mammary tumor prevention in their offspring through epigenetic regulations.

Advanced ADC Histogram, Perfusion, and Permeability Metrics Show an Association with Survival and Pseudoprogression in Newly Diagnosed Diffuse Intrinsic Pontine Glioma: A Report from the Pediatric Brain Tumor Consortium [FUNCTIONAL]

BACKGROUND AND PURPOSE:

Diffuse intrinsic pontine glioma is a lethal childhood brain cancer with dismal prognosis and MR imaging is the primary methodology used for diagnosis and monitoring. Our aim was to determine whether advanced diffusion, perfusion, and permeability MR imaging metrics predict survival and pseudoprogression in children with newly diagnosed diffuse intrinsic pontine glioma.

MATERIALS AND METHODS:

A clinical trial using the poly (adenosine diphosphate ribose) polymerase (PARP) inhibitor veliparib concurrently with radiation therapy, followed by maintenance therapy with veliparib + temozolomide, in children with diffuse intrinsic pontine glioma was conducted by the Pediatric Brain Tumor Consortium. Standard MR imaging, DWI, dynamic contrast-enhanced perfusion, and DSC perfusion were performed at baseline and approximately every 2 months throughout treatment. ADC histogram metrics of T2-weighted FLAIR and enhancing tumor volume, dynamic contrast-enhanced permeability metrics for enhancing tumors, and tumor relative CBV from DSC perfusion MR imaging were calculated. Baseline values, post-radiation therapy changes, and longitudinal trends for all metrics were evaluated for associations with survival and pseudoprogression.

RESULTS:

Fifty children were evaluable for survival analyses. Higher baseline relative CBV was associated with shorter progression-free survival (P = .02, Q = 0.089) and overall survival (P = .006, Q = 0.055). Associations of higher baseline mean transfer constant from the blood plasma into the extravascular extracellular space with shorter progression-free survival (P = .03, Q = 0.105) and overall survival (P = .03, Q = 0.102) trended toward significance. An increase in relative CBV with time was associated with shorter progression-free survival (P < .001, Q < 0.001) and overall survival (P = .004, Q = 0.043). Associations of longitudinal mean extravascular extracellular volume fraction with progression-free survival (P = .03, Q = 0.104) and overall survival (P = .03, Q = 0.105) and maximum transfer constant from the blood plasma into the extravascular extracellular space with progression-free survival (P = .03, Q = 0.102) trended toward significance. Greater increases with time were associated with worse outcomes. True radiologic progression showed greater post-radiation therapy decreases in mode_ADC_FLAIR compared with pseudoprogression (means, –268.15 versus –26.11, P = .01.)

CONCLUSIONS:

ADC histogram, perfusion, and permeability MR imaging metrics in diffuse intrinsic pontine glioma are useful in predicting survival and pseudoprogression.

Comparison of Enhancement of the Vestibular Perilymph between Variable and Constant Flip Angle-Delayed 3D-FLAIR Sequences in Meniere Disease [HEAD & NECK]

BACKGROUND AND PURPOSE:

Endolymphatic hydrops in patients with Menière disease relies on delayed postcontrast 3D-FLAIR sequences. The purpose of this study was to compare the degree of perilymphatic enhancement and the detection rate of endolymphatic hydrops using constant and variable flip angles sequences.

MATERIALS AND METHODS:

This was a retrospective study performed in 16 patients with Menière disease who underwent 3T MR imaging 4 hours after gadolinium injection using two 3D-FLAIR sequences with a constant flip angle at 140° for the first and a heavily-T2 variable flip angle for the second. The signal intensity ratio was measured using the ROI method. We graded endolymphatic hydrops and evaluated the cochlear blood-labyrinth barrier impairment.

RESULTS:

Both for symptomatic and asymptomatic ears, the median signal intensity ratio was significantly higher with the constant flip angle than with the heavily-T2 variable flip angle (7.16 versus 1.54 and 7.00 versus 1.45, P < .001). Cochlear blood-labyrinth barrier impairment was observed in 4/18 symptomatic ears with the heavily-T2 variable flip angle versus 8/19 with constant flip angle sequences. With heavily-T2 variable flip angle sequences, endolymphatic hydrops was observed in 7–10/19 symptomatic ears versus 12/19 ears with constant flip angle sequences. We found a significant association between the clinical symptomatology and the presence of endolymphatic hydrops with constant flip angle but not with heavily-T2 variable flip angle sequences. Interreader agreement was always perfect with constant flip angle sequences while it was fair-to-moderate with heavily-T2 variable flip angle sequences.

CONCLUSIONS:

3D-FLAIR constant flip angle sequences provide a higher signal intensity ratio and are superior to heavily-T2 variable flip angle sequences in reliably evaluating the cochlear blood-labyrinth barrier impairment and the endolymphatic space.

For weeks after Ahmaud Arbery was killed while running down a road in coastal southern Georgia, there were few public developments in the case of a 25-year-old unarmed black man who was shot while being pursued by two white men with weapons in February.Then a graphic video of the shooting surfaced online, spurring widespread outrage.Within days, the Georgia Bureau of Investigation had taken over the case. The video was criticized by celebrities and politicians alike, including President Donald Trump, who called the footage "very, very disturbing," and former Vice President Joe Biden, the presumptive Democratic presidential nominee, who said Arbery had essentially been "lynched before our very eyes."And in a major turn, the authorities announced Thursday night that they had arrested two suspects in the case and charged them with murder and aggravated assault.The video -- which by Friday officials had described as "a very important piece" of evidence in moving forward with criminal charges -- was first posted by WGIG, a radio station in Brunswick, Georgia, which said it had obtained the footage from an anonymous source.But in a twist emblematic of the small-town politics that have defined the case, that source turned out to be a criminal defense lawyer in town who had informally consulted with the suspects.The lawyer, Alan Tucker, said in an interview Friday that the video had come from the cellphone of a man who had filmed the episode and that he later gave the footage to the radio station. Tucker's role was confirmed by Scott Ryfun, who oversees the station's programming.Asked why he had leaked the video, Tucker said he had wanted to dispel rumors that he said had fueled tension in the community. "It wasn't two men with a Confederate flag in the back of a truck going down the road and shooting a jogger in the back," Tucker said."It got the truth out there as to what you could see," he added. "My purpose was not to exonerate them or convict them."The video, taken from inside a vehicle, shows Arbery running when he comes upon a white truck, with one man standing next to its open driver's-side door and another in the bed of the pickup. Arbery runs around the truck and disappears briefly from view. Then the man standing outside the truck tussles with him, and three gunshots are heard.The authorities identified Travis McMichael, 34, as the person who shot Arbery. His father, Gregory McMichael, 64, a retired investigator at the local district attorney's office, was also charged.Before the charges were filed this week, two prosecutors had recused themselves from handling the case, citing professional ties to Gregory McMichael. Tucker, too, said he had been an acquaintance of McMichael's from their work in legal circles.Reports suggest Tucker had consulted with the McMichael family in some capacity during the investigation, although it is not clear to what extent. Reached by The Washington Post before his arrest Thursday, Gregory McMichael referred questions to Tucker.Tucker declined to comment on his conversations with the McMichaels on Friday, citing attorney-client privilege."I'm not going to tell you what I told them or what they told me," he said, using profanity to say that any conversations -- had they occurred, he said -- were none of the public's business.At times during the interview, a woman could be heard in the background whispering suggested answers to Tucker.By Friday afternoon, Tucker said that it had been decided that he would not be retained as the lawyer for either of the McMichaels, and it was unclear who was representing them.Tucker said he would not be representing anyone else involved in the case, as the authorities announced Friday that they were pursuing a number of leads, including investigating the man who took the video.The man, Roddie Bryan, lives in the neighborhood. "If anybody wanted a copy of the video, he would give it to them."But he said the added attention, including the scrutiny from the authorities, had come as a shock to his client, a mechanic who had since lost his job and received threats. "The atmosphere down here is very volatile," Gough said. "People are in fear. That's all a result of the last few days."The latest developments in the case on Friday fell on Arbery's birthday, when he would have turned 26. YMMV obviously--I ordered two of these a month ago and they have been running fine for me.  All I had to do was reinstall the newest system update and they have both been fine with no issues.  One system's issue noted was "freezes" and the other was "no video". 

 

It says he has only 2 in stock, but I bought two a while back and the listing has remained active for the entire time with stock available, so he probably leaves the number low to keep demand going. This is probably a bulk buyer who doesn't extensively troubleshoot the systems.  I am not affiliated with the seller.  Cosmetically, the systems are nice enough to trade in without taking a refurb fee when stores re-open for business.
