Archaeologists verify Florida's Mound Key as location of elusive Spanish fort

(Florida Museum of Natural History) Florida and Georgia archaeologists have discovered the location of Fort San Antón de Carlos, home of one of the first Jesuit missions in North America. The Spanish fort was built in 1566 in the capital of the Calusa, the most powerful Native American tribe in the region, on present-day Mound Key in the center of Estero Bay on Florida's Gulf Coast.

Can fluorescence-guided surgery help identify all lesions in unknown locations or is the integrated use of a roadmap created by preoperative imaging mandatory? A blinded study in prostate cancer patients.

Rationale: Lymphatic tracers can help visualize the lymphatic drainage patterns and sentinel nodes of individual prostate cancer patients. To determine the role of nuclear medicine, in particular the positional guidance of a SPECT/CT-based 3D imaging roadmap, in this process we studied to which extend fluorescence-guidance underestimated the number of target lesions. Methods: SPECT/CT imaging was performed after intraprostatic tracer administration of either ICG-99mTc-nanocolloid (hybrid tracer group) or 99mTc-nanocolloid to create a roadmap that depicted all sentinel nodes (SNs). Patients who received 99mTc-nanocolloid were injected with "free" ICG immediately prior to surgery ("free" ICG group). Before unblinding, fluorescence-guidance was used for intraoperative SN identification. This was followed by extended pelvic lymph node dissection (ePLND). Following unblinding of the SPECT/CT images, the number of missed SN’s were recorded and their resection was pursued when the anatomy allowed. Results: Preoperative SPECT/CT revealed no differences in the SN identification rate between ICG-99mTc-nanocolloid and 99mTc-nanocolloid. However, fluorescence-guidance only allowed intraoperative removal of all SNs in 40% of patients in the hybrid tracer group and in 20% of patients in the "free" ICG group. Overall, 75.9% of the intraoperatively resected SNs in the hybrid tracer group and 51.8% of the SNs in the "free" ICG group were removed solely under fluorescence-guidance. During ePLND 22 additional SNs were resected (7 in the hybrid tracer group and 15 in the "free" ICG group). After unblinding 18 remaining SNs were identified (6 in the hybrid group and 12 in the "free" ICG group). In the "free" ICG group, ex vivo evaluation of the excised specimens revealed that 14 SNs removed under ePLND or after unblinding contained radioactivity but no fluorescence. Conclusion: The preoperative imaging roadmap provided by SPECT/CT enhanced the detection of prostate SNs in more ectopic locations in 17 of the 25 patients and the hybrid tracer ICG-99mTc-nanocolloid was shown to outperform "free" ICG. Overall, fluorescence-guided pelvic nodal surgery underestimated the number of SNs in 60-80% of patients.

SSO and other putative inhibitors of FA transport across membranes by CD36 disrupt intracellular metabolism, but do not affect FA translocation [Research Articles]

Membrane-bound proteins have been proposed to mediate the transport of long-chain FA (LCFA) transport through the plasma membrane (PM). These proposals are based largely on reports that PM transport of LCFAs can be blocked by a number of enzymes and purported inhibitors of LCFA transport. Here, using the ratiometric pH indicator (2',7'-bis-(2-carboxyethyl)-5-(and-6-)-carboxyfluorescein and acrylodated intestinal FA-binding protein-based dual fluorescence assays, we investigated the effects of nine inhibitors of the putative FA transporter protein CD36 on the binding and transmembrane movement of LCFAs. We particularly focused on sulfosuccinimidyl oleate (SSO), reported to be a competitive inhibitor of CD36-mediated LCFA transport. Using these assays in adipocytes and inhibitor-treated protein-free lipid vesicles, we demonstrate that rapid LCFA transport across model and biological membranes remains unchanged in the presence of these purported inhibitors. We have previously shown in live cells that CD36 does not accelerate the transport of unesterified LCFAs across the PM. Our present experiments indicated disruption of LCFA metabolism inside the cell within minutes upon treatment with many of the "inhibitors" previously assumed to inhibit LCFA transport across the PM. Furthermore, using confocal microscopy and a specific anti-SSO antibody, we found that numerous intracellular and PM-bound proteins are SSO-modified in addition to CD36. Our results support the hypothesis that LCFAs diffuse rapidly across biological membranes and do not require an active protein transporter for their transmembrane movement.

On estimating the location parameter of the selected exponential population under the LINEX loss function

Mohd Arshad, Omer Abdalghani.

Source: Brazilian Journal of Probability and Statistics, Volume 34, Number 1, 167--182.

Abstract:
Suppose that $pi_{1},pi_{2},ldots ,pi_{k}$ be $k(geq2)$ independent exponential populations having unknown location parameters $mu_{1},mu_{2},ldots,mu_{k}$ and known scale parameters $sigma_{1},ldots,sigma_{k}$. Let $mu_{[k]}=max {mu_{1},ldots,mu_{k}}$. For selecting the population associated with $mu_{[k]}$, a class of selection rules (proposed by Arshad and Misra [ Statistical Papers 57 (2016) 605–621]) is considered. We consider the problem of estimating the location parameter $mu_{S}$ of the selected population under the criterion of the LINEX loss function. We consider three natural estimators $delta_{N,1},delta_{N,2}$ and $delta_{N,3}$ of $mu_{S}$, based on the maximum likelihood estimators, uniformly minimum variance unbiased estimator (UMVUE) and minimum risk equivariant estimator (MREE) of $mu_{i}$’s, respectively. The uniformly minimum risk unbiased estimator (UMRUE) and the generalized Bayes estimator of $mu_{S}$ are derived. Under the LINEX loss function, a general result for improving a location-equivariant estimator of $mu_{S}$ is derived. Using this result, estimator better than the natural estimator $delta_{N,1}$ is obtained. We also shown that the estimator $delta_{N,1}$ is dominated by the natural estimator $delta_{N,3}$. Finally, we perform a simulation study to evaluate and compare risk functions among various competing estimators of $mu_{S}$.

The equivalence of dynamic and static asset allocations under the uncertainty caused by Poisson processes

Yong-Chao Zhang, Na Zhang.

Source: Brazilian Journal of Probability and Statistics, Volume 33, Number 1, 184--191.

Abstract:
We investigate the equivalence of dynamic and static asset allocations in the case where the price process of a risky asset is driven by a Poisson process. Under some mild conditions, we obtain a necessary and sufficient condition for the equivalence of dynamic and static asset allocations. In addition, we provide a simple sufficient condition for the equivalence.

Robust location estimators in regression models with covariates and responses missing at random. (arXiv:2005.03511v1 [stat.ME])

This paper deals with robust marginal estimation under a general regression model when missing data occur in the response and also in some of covariates. The target is a marginal location parameter which is given through an $M-$functional. To obtain robust Fisher--consistent estimators, properly defined marginal distribution function estimators are considered. These estimators avoid the bias due to missing values by assuming a missing at random condition. Three methods are considered to estimate the marginal distribution function which allows to obtain the $M-$location of interest: the well-known inverse probability weighting, a convolution--based method that makes use of the regression model and an augmented inverse probability weighting procedure that prevents against misspecification. The robust proposed estimators and the classical ones are compared through a numerical study under different missing models including clean and contaminated samples. We illustrate the estimators behaviour under a nonlinear model. A real data set is also analysed.

Optimal asset allocation with multivariate Bayesian dynamic linear models

Jared D. Fisher, Davide Pettenuzzo, Carlos M. Carvalho.

Source: The Annals of Applied Statistics, Volume 14, Number 1, 299--338.

Abstract:
We introduce a fast, closed-form, simulation-free method to model and forecast multiple asset returns and employ it to investigate the optimal ensemble of features to include when jointly predicting monthly stock and bond excess returns. Our approach builds on the Bayesian dynamic linear models of West and Harrison ( Bayesian Forecasting and Dynamic Models (1997) Springer), and it can objectively determine, through a fully automated procedure, both the optimal set of regressors to include in the predictive system and the degree to which the model coefficients, volatilities and covariances should vary over time. When applied to a portfolio of five stock and bond returns, we find that our method leads to large forecast gains, both in statistical and economic terms. In particular, we find that relative to a standard no-predictability benchmark, the optimal combination of predictors, stochastic volatility and time-varying covariances increases the annualized certainty equivalent returns of a leverage-constrained power utility investor by more than 500 basis points.

Clostridium difficile Infection Among Children Across Diverse US Geographic Locations

Little is known about the epidemiology and pathogenicity of Clostridium difficile infection among children, particularly those aged ≤3 years in whom colonization is common and pathogenicity uncertain.

Young children, 1 to 3 years of age, had the highest Clostridium difficile infection incidence. Considering that clinical presentation, outcomes, and disease severity were similar across age groups, C difficile infection in the youngest age group likely represents true disease and not asymptomatic colonization. Impact Locations and Concussion Outcomes in High School Football Player-to-Player Collisions

Recent concussion research has examined the role of impact location (ie, the area on the head to which impact occurred); however, no studies exist regarding impact location’s association with concussion outcomes (eg, symptomatology, symptom resolution time, return to play).

This study is the first to examine the association of impact location and concussion outcomes in young athletes. Our findings suggest that impact location, as assessed by sideline observers/player report, is likely of little use in predicting clinical outcomes. (Read the full article)

Variation in Rotavirus Vaccine Coverage by Provider Location and Subsequent Disease Burden

Uptake of rotavirus vaccines has increased steadily since introduction. Despite their demonstrated impact, rotavirus vaccine coverage is lower than for other vaccines recommended in infancy and disease continues to occur.

We observed higher rotavirus detection rates among patients from provider locations with lower rotavirus vaccine coverage; providers who do not offer rotavirus vaccine to age-eligible children may create pockets of susceptible children that serve as reservoirs of ongoing disease transmission.
