Background: This journal paper seeks to understand what executives value in data visualization, based on the literature published from prior data studies. Methodology: The qualitative methodology was used to understand the sentiments of executives and data analysts using semi-structured interview techniques. Contribution: The preliminary findings can provide practical knowledge for data visualization designers, but can also provide academics with knowledge to reflect on and use, specifically in relation to information systems (IS) that integrate human experience with technology in more valuable and productive ways. Findings: Preliminary results from interviews with executives and data analysts point to the relevance of understanding and effectively presenting the data source and the data journey, using the right data visualization technology to fit the nature of the data, creating an intuitive platform which enables collaboration and newness, the data presenter’s ability to convey the data message and the alignment of the visualization to core the objectives as key criteria to be applied for successful data visualizations Recommendations for Practitioners: Practitioners, specifically data analysts, should consider the results highlighted in the findings and adopt such recommendations when presenting data visualizations. These include data and premise understanding, ensuring alignment to the executive’s objective, possessing the ability to convey messages succinctly and clearly to the audience, having knowledge of the domain to answer questions effectively, and using the right technology to convey the message. Recommendation for Researchers: The importance of human cognitive and sensory processes and its impact in IS development is paramount. More focus can be placed on the psychological factors of technology acceptance. The current TAM model, used to describe use, identifies perceived usefulness and perceived ease-of-use as the primary considerations in technology adoption. However, factors that have been identified that impact on use do not express the importance of cognitive processes in technology adoption. Future Research: Future research requires further focus on intangible and psychological factors that could affect technology adoption and use, as well as understanding data visualization effectiveness in corporate environments, not only predominantly within the Health sector. Lessons from Health sector studies in data visualization should be used as a platform. Full Article

Mosquitoes are ideal model organisms for studying egg maturation in arthropods, as their follicle development is initiated only following the ingestion of a blood meal, followed by a carefully orchestrated series of hormonally regulated events leading to egg maturation. A cohort of approximately 100 follicles per mosquito ovary begin developing synchronously. However, a significant fraction of follicles ultimately undergo apoptosis and oosorption, especially when available resources from the blood meal are limited. Therefore, simple, rapid, and reliable techniques to accurately evaluate follicular atresia are necessary to understand mechanisms underlying follicle development in insects. This protocol describes how to detect apoptotic follicle cells within the Aedes aegypti mosquito ovaries using a commercially available fluorescent-labeled inhibitor of caspases (FLICA). Caspases are key players in animal apoptosis. In this assay, the FLICA reagent enters the intracellular compartment of follicles in dissected mosquito ovaries and covalently binds to active caspases. The bound reagent remains within the cell and its fluorescent signal can be observed by confocal microscopy. Although this method was specifically developed for visualizing apoptotic ovarian follicles during Ae. aegypti mosquito egg development, it should be applicable to other mosquito tissues that undergo caspase-mediated program cell death in a time-dependent manner. visualization Scanning electron microscopy as a method for sample visualization in protein X-ray crystallography By scripts.iucr.org Published On :: 2020-04-10 Developing methods to determine high-resolution structures from micrometre- or even submicrometre-sized protein crystals has become increasingly important in recent years. This applies to both large protein complexes and membrane proteins, where protein production and the subsequent growth of large homogeneous crystals is often challenging, and to samples which yield only micro- or nanocrystals such as amyloid or viral polyhedrin proteins. The versatile macromolecular crystallography microfocus (VMXm) beamline at Diamond Light Source specializes in X-ray diffraction measurements from micro- and nanocrystals. Because of the possibility of measuring data from crystalline samples that approach the resolution limit of visible-light microscopy, the beamline design includes a scanning electron microscope (SEM) to visualize, locate and accurately centre crystals for X-ray diffraction experiments. To ensure that scanning electron microscopy is an appropriate method for sample visualization, tests were carried out to assess the effect of SEM radiation on diffraction quality. Cytoplasmic polyhedrosis virus polyhedrin protein crystals cryocooled on electron-microscopy grids were exposed to SEM radiation before X-ray diffraction data were collected. After processing the data with DIALS, no statistically significant difference in data quality was found between datasets collected from crystals exposed and not exposed to SEM radiation. This study supports the use of an SEM as a tool for the visualization of protein crystals and as an integrated visualization tool on the VMXm beamline. Full Article text

In addition, information about the shape and internal defects of crystals allows the optimization of data-collection strategies. Here, it is demonstrated that the X-ray beam available on the macromolecular crystallography beamline P14 at the high-brilliance synchrotron-radiation source PETRA III at DESY, Hamburg, Germany can be used for high-energy phase-contrast microtomography of protein crystals mounted in an optically opaque lipidic cubic phase matrix. Three-dimensional tomograms have been obtained at X-ray doses that are substantially smaller and on time scales that are substantially shorter than those used for diffraction-scanning approaches that display protein crystals at micrometre resolution. Adding a compound refractive lens as an objective to the imaging setup, two-dimensional imaging at sub-micrometre resolution has been achieved. All experiments were performed on a standard macromolecular crystallography beamline and are compatible with standard diffraction data-collection workflows and apparatus. The large diffraction data volumes require high-throughput software to preprocess the raw images for subsequent analysis. ClickX is a program designated for serial crystallography data preprocessing, capable of rapid data sorting for online feedback and peak-finding refinement by parameter optimization. The graphical user interface (GUI) provides convenient access to various operations such as pattern visualization, statistics plotting and parameter tuning. A batch job module is implemented to facilitate large-data-volume processing. A two-step geometry calibration for single-panel detectors is also integrated into the GUI, where the beam center and detector tilting angles are optimized using an ellipse center shifting method first, then all six parameters, including the photon energy and detector distance, are refined together using a residual minimization method. Implemented in Python, ClickX has good portability and extensibility, so that it can be installed, configured and used on any computing platform that provides a Python interface or common data file format. ClickX has been tested in online analysis at the Pohang Accelerator Laboratory X-ray Free-Electron Laser, Korea, and the Linac Coherent Light Source, USA.
