Sri Lanka's first transgender candidate hopes to break political, social barriers

COLOMBO — Chanu Nimesha is contesting Sri Lanka's parliamentary election on Thursday (Nov 14) as the first openly transgender candidate, hoping to forge a more inclusive and tolerant political culture on the South Asian island. Transgender people are estimated to make up about one per cent of Sri Lanka's 22 million people, according to Equal Ground, a local civil society group. They frequently face social rejection, lack legal protection and have almost no representation in political parties. Nimesha, who is contesting from Kegalle — about 80km east of Colombo — said she was the first transgender person to run for a seat in the 225-strong parliament and that her message of social justice had been well received. "I'm not concerned about winning or losing," she said, sitting in her one-bedroom flat. "But it is important for me to be present in this space, to be seen, to inspire others like me. I want to help everyone, not just my community."

2021 Miss Nevada Will Be The First Openly Transgender Miss USA Contestant

Wearing a rainbow sequin gown she designed herself in honor of Pride Month, Kataluna Enriquez made history last weekend when she was crowned Miss Nevada USA — the win will make her the first openly transgender contestant to compete in the upcoming Miss USA pageant this fall.

"My win is our win," she posted afterward on her Instagram in a message to the LGBTQ community. "We just made history. Happy pride."

Enriquez, who was also Miss Nevada USA's first trans contestant, beat out 21 other women for the top spot. She will represent the Silver State at the 2021 Miss USA pageant that will be held on Nov. 29 in Tulsa, Okla., where she will have a chance to be crowned Miss USA and advance to the Miss Universe pageant.

If crowned Miss USA, Enriquez will become the second trans contestant to compete for Miss Universe, after Angela Ponce, who represented Spain in the 2018 Miss Universe pageant. The pageant began allowing transgender contestants in 2012.

Enriquez began competing in pageants in 2015. Unable to afford custom designer gowns that fit her body at the time, she began designing her own to wear for competitions and eventually started her own line, Kataluna Kouture (@katalunakouture). In March, Enriquez became the first trans woman to win Miss Nevada's preliminary pageant, Miss Silver State USA.

The journey has not been easy, and Enriquez has faced discrimination. While competing in a pageant outside of Nevada, she had not been given a roommate when pageant organizers learned she was trans. A doctor had also been sent to certify that she was a woman before she could continue.

But Enriquez told NPR's Weekend All Things Considered that her determination to make history was what motivated her to keep competing.

"I had a purpose and I had a dream," she said. "I wanted to compete on the Miss USA stage. When I was young, I always wanted to see someone on the Miss USA stage — someone like me. And it just happened to be that I was the person that I needed to make history."

As she prepares for the Miss USA pageant, Enriquez said she plans to advocate for equality and mental health.

"My win is not just a win for the trans community," she said. "It's a win for all women to be represented."

Generating and Validating Transgenic Mosquitoes with Transposon-Mediated Transgenesis

Transposon-mediated transgenesis has revolutionized both basic and applied studies of mosquito vectors of disease. Currently, techniques such as enhancer traps and transposon tagging, which rely on remobilizable insertional mutagenesis, are only possible with transposon-based vector systems. Here, we provide general descriptions of methods and applications of transposon-based mosquito transgenesis. The exact procedures must be adapted to each mosquito species and comparisons of some differences among different mosquito species are outlined. A number of excellent publications showing detailed and specific protocols and methods are featured and referenced.

Mosquito Transposon-Mediated Transgenesis

Transposon-mediated transgenesis of mosquito vectors of disease pathogens followed the early success of transgenesis in the vinegar fly, Drosophila melanogaster. The P transposable element used in Drosophila does not function canonically in mosquitoes, and repeatable, routine transgenesis in mosquitoes was not accomplished until new transposable elements were discovered and validated. A number of distinct transposons were subsequently identified that mediate the introduction of exogenous DNA in a stable and heritable manner in mosquito species, including members of the genera Aedes, Anopheles, and Culex. The most versatile element, piggyBac, is functional in all of these mosquito genera, as well as in many other insects in diverse orders, and has been used extensively outside the class. Transposon-mediated transgenesis of recessive and dominant marker genes and reporter systems has been used to define functional fragments of gene control sequences, introduce exogenous DNA encoding products beneficial to medical interests, and act as "enhancer traps" to identify endogenous genes with specific expression characteristics.

Early Prediction and Impact Assessment of CYP3A4-Related Drug-Drug Interactions for Small-Molecule Anticancer Drugs Using Human-CYP3A4-Transgenic Mouse Models [Articles]

Early detection of drug-drug interactions (DDIs) can facilitate timely drug development decisions, prevent unnecessary restrictions on patient enrollment, resulting in clinical study populations that are not representative of the indicated study population, and allow for appropriate dose adjustments to ensure safety in clinical trials. All of these factors contribute to a streamlined drug approval process and enhanced patient safety. Here we describe a new approach for early prediction of the magnitude of change in exposure for cytochrome P450 (P450) CYP3A4-related DDIs of small-molecule anticancer drugs based on the model-based extrapolation of human-CYP3A4-transgenic mice pharmacokinetics to humans. Victim drugs brigatinib and lorlatinib were evaluated with the new approach in combination with the perpetrator drugs itraconazole and rifampicin. Predictions of the magnitude of change in exposure deviated at most 0.99- to 1.31-fold from clinical trial results for inhibition with itraconazole, whereas exposure predictions for the induction with rifampicin were less accurate, with deviations of 0.22- to 0.48-fold. Results for the early prediction of DDIs and their clinical impact appear promising for CYP3A4 inhibition, but validation with more victim and perpetrator drugs is essential to evaluate the performance of the new method.

SIGNIFICANCE STATEMENT

The described method offers an alternative for the early detection and assessment of potential clinical impact of CYP3A4-related drug-drug interactions. The model was able to adequately describe the inhibition of CYP3A4 metabolism and the subsequent magnitude of change in exposure. However, it was unable to accurately predict the magnitude of change in exposure of victim drugs in combination with an inducer.
