Understanding the mechanical limitations of the performance of soft X-ray monochromators at MAX IV laboratory

MAX IV is a fourth-generation, or diffraction-limited, synchrotron light source with a number of state-of-the-art beamlines. The performance of a beamline is, to a high degree, set by the energy resolution it can achieve, which in turn is governed to a large extent by the monochromator. During the design phase of a monochromator, the mechanical requirements must be fully understood and met with margin. During commissioning, the performance must be verified and optimized. In this paper, six soft X-ray monochromators at MAX IV beamlines (Bloch, Veritas, HIPPIE, SPECIES, FinEstBeAMS and SoftiMAX) are examined with a focus on their resolving power, energy range and the time required to change measurement range, as those parameters are dependent on each other. The monochromators have a modern commercial design, planned and developed in close collaboration with the vendors. This paper aims to present the current status of the commissioning at MAX IV with emphasis on elucidating the mechanical limitations on the performance of the monochromators. It contains analysis of the outcome and our approach to achieve fast and high-resolution monochromators.

Campgrounds reopen in Iowa Friday, see takers despite some health limitations

Some Eastern Iowans are ready to go camping.

With Gov. Kim Reynolds allowing campgrounds across the state to open Friday, some people wasted little time in heading outdoors.

“They’re already starting to fill up,” said Ryan Schlader of Linn County Conservation. “By about 7 this morning, we had a dozen at Squaw Creek Park. People were coming in bright and early to camp. We’re not surprised.”

Schlader said Linn County Conservation tried to have the campgrounds open at the county’s Squaw Creek, Morgan Creek and Pinicon Ridge parks at 5 a.m. Friday. He expected all of them would be busy.

“I think people were ready to go,” he said.

Lake Macbride State Park in Johnson County didn’t see quite as much of a rush for campsites, park manager Ron Puettmann reported Friday morning, saying he’d had six walk-ins for the park’s 42 campsites.

Camping this weekend will be done on a first-come, first-served basis. Sites won’t be available for reservations until next week, though online reservations can be made now, Puettmann said.

“I’m quite sure people were waiting anxiously to get on,” he said.

While Reynolds’ campground announcement came Wednesday, Schlader and Puettmann said they had no issues having the campgrounds ready for Friday.

Schlader said county staff have been in touch with the Iowa Department of Natural Resources and other county conservation boards to discuss protocols for reopening to ensure a safe experience for campers and employers.

“We anticipated at some point the order would be lifted,” Schlader said. “We were anticipating maybe May 15. The campgrounds were in good shape and ready to go.”

For now, camping comes with some limitations:

• Campers can camp only in a self-contained unit with a functioning restroom, such as a recreational vehicle.

• Shower houses with restrooms will remain closed for the time being.

• Campsites are limited to six people unless they are from the same household.

• No visitors are allowed at the campsites.

Puettmann said staffers and a DNR officer will be on hand to make sure guidelines are followed, but he didn’t anticipate enforcement would be an issue.

“For the most part, we’re going to allow people to police themselves,” he said.

It’s hard to gauge demand, Schlader said.

The weather isn’t yet deal for camping, and some people might not be ready to camp, given the continuing coronavirus.

“There is a lot of uncertainty,” he said. “Do people feel like they need to get out and enjoy a camping experience within their own campsite, or do people still feel under the weather and think it’s not a good idea for my family to go right now? ... We just want this to be an option for people.”

Metabolism of Gluconeogenic Substrates by an Intracellular Fungal Pathogen Circumvents Nutritional Limitations within Macrophages

ABSTRACT

Microbial pathogens exploit host nutrients to proliferate and cause disease. Intracellular pathogens, particularly those exclusively living in the phagosome such as Histoplasma capsulatum, must adapt and acquire nutrients within the nutrient-limited phagosomal environment. In this study, we investigated which host nutrients could be utilized by Histoplasma as carbon sources to proliferate within macrophages. Histoplasma yeasts can grow on hexoses and amino acids but not fatty acids as the carbon source in vitro. Transcriptional analysis and metabolism profiling showed that Histoplasma yeasts downregulate glycolysis and fatty acid utilization but upregulate gluconeogenesis within macrophages. Depletion of glycolysis or fatty acid utilization pathways does not prevent Histoplasma growth within macrophages or impair virulence in vivo. However, loss of function in Pck1, the enzyme catalyzing the first committed step of gluconeogenesis, impairs Histoplasma growth within macrophages and severely attenuates virulence in vivo, indicating that Histoplasma yeasts rely on catabolism of gluconeogenic substrates (e.g., amino acids) to proliferate within macrophages.

IMPORTANCE Histoplasma is a primary human fungal pathogen that survives and proliferates within host immune cells, particularly within the macrophage phagosome compartment. The phagosome compartment is a nutrient-limited environment, requiring Histoplasma yeasts to be able to assimilate available carbon sources within the phagosome to meet their nutritional needs. In this study, we showed that Histoplasma yeasts do not utilize fatty acids or hexoses for growth within macrophages. Instead, Histoplasma yeasts consume gluconeogenic substrates to proliferate in macrophages. These findings reveal the phagosome composition from a nutrient standpoint and highlight essential metabolic pathways that are required for a phagosomal pathogen to proliferate in this intracellular environment.

Advancing Biologics Development Programs with Legacy Cell Lines: Advantages and Limitations of Genetic Testing for Addressing Clonality Concerns Prior to Availability of Late Stage Process and Product Consistency Data

The bioprocessing industry uses recombinant mammalian cell lines to generate therapeutic biologic drugs. To ensure consistent product quality of the therapeutic proteins, it is imperative to have a controlled production process. Regulatory agencies and the biotechnology industry consider cell line "clonal origin" an important aspect of maintaining process control. Demonstration of clonal origin of the cell substrate, or production cell line, has received considerable attention in the past few years, and the industry has improved methods and devised standards to increase the probability and/or assurance of clonal derivation. However, older production cell lines developed before the implementation of these methods, herein referred to as "legacy cell lines," may not meet current regulatory expectations for demonstration of clonal derivation. In this article, the members of the IQ Consortium Working Group on Clonality present our position that the demonstration of process consistency and product comparability of critical quality attributes throughout the development life cycle should be sufficient to approve a license application without additional genetic analysis to support clonal origin, even for legacy cell lines that may not meet current day clonal derivation standards. With this commentary, we discuss advantages and limitations of genetic testing methods to support clonal derivation of legacy cell lines and wish to promote a mutual understanding with the regulatory authorities regarding their optional use during early drug development, subsequent to Investigational New Drug (IND) application and before demonstration of product and process consistency at Biologics License Applications (BLA) submission.

Impact bonds, also known as Pay for Success contracts in the United States, have leveraged over $200 million in upfront private capital for social services worldwide over the last six years, and by 2020 the market is expected to triple. Brookings experts have published two reports analyzing the market, the first of which is a comprehensive review of the global impact bond market and the second of which examines applications to Early Childhood Development programs.

On February 29, the Global Economy and Development program at Brookings hosted a discussion on the scope for social and development impact bonds to address social challenges globally. Sessions reflected on the types of challenges for which these new financing modalities are best suited, and the factors critical for their success. Sir Ronald Cohen, chairman of the Global Social Impact Investment Steering Group, provided keynote remarks, followed by presentations from Emily Gustafsson-Wright, fellow at the Center for Universal Education at Brookings and lead author of both reports on impact bonds.  The event included two panel discussions and a networking lunch.
