tap A structural study of TatD from Staphylococcus aureus elucidates a putative DNA-binding mode of a Mg2+-dependent nuclease By scripts.iucr.org Published On :: 2020-04-17 TatD has been thoroughly investigated as a DNA-repair enzyme and an apoptotic nuclease, and still-unknown TatD-related DNases are considered to play crucial cellular roles. However, studies of TatD from Gram-positive bacteria have been hindered by an absence of atomic detail and the resulting inability to determine function from structure. In this study, an X-ray crystal structure of SAV0491, which is the TatD enzyme from the Gram-positive bacterium Staphylococcus aureus (SaTatD), is reported at a high resolution of 1.85 Å with a detailed atomic description. Although SaTatD has the common TIM-barrel fold shared by most TatD-related homologs, and PDB entry 2gzx shares 100% sequence identity with SAV0491, the crystal structure of SaTatD revealed a unique binding mode of two phosphates interacting with two Ni2+ ions. Through a functional study, it was verified that SaTatD has Mg2+-dependent nuclease activity as a DNase and an RNase. In addition, structural comparison with TatD homologs and the identification of key residues contributing to the binding mode of Ni2+ ions and phosphates allowed mutational studies to be performed that revealed the catalytic mechanism of SaTatD. Among the key residues composing the active site, the acidic residues Glu92 and Glu202 had a critical impact on catalysis by SaTatD. Furthermore, based on the binding mode of the two phosphates and structural insights, a putative DNA-binding mode of SaTatD was proposed using in silico docking. Overall, these findings may serve as a good basis for understanding the relationship between the structure and function of TatD proteins from Gram-positive bacteria and may provide critical insights into the DNA-binding mode of SaTatD. One of the last widely circulated print satirical magazines in America will leave newsstands after this year, according to sources at DC Comics, which publishes MAD magazine. While the Harvard Lampoon remains in business, The Onion hasn't been in print since 2013. The once-influential Spy was a casualty of the 1990s. At MAD's peak in the early 1970s, more than 2 million people subscribed to it, both for its pungent political humor and deeply adolescent jokes. In 2017, that number had reportedly dropped to 140,000. MAD isn't completely shutting down, but it will be radically downsized and changed. Readers will only be able to find the 67-year-old humor magazine at comic book stores and through subscriptions. After issue No. 10 this fall, there will no longer be new content, except for end-of-year specials which will be all new. If no local team is in the running (or exists), I'm always glad to root for a Bay Area team that makes it. My tweet got responses from some Southern Californians who have no interest in supporting a San Francisco team, especially given the Giants' World Series championship. It goes without saying that many Dodger fans are loathe to support the Giants, under any circumstances. Given the historic bad blood between the teams, that's no surprise, but I think it runs even deeper. The divide between Northern and Southern California is about more than sports, or even water rights. It's rooted in distinct cultural differences between the two. However, California has evolved to the point where the bigger cultural divide now might be between coastal and inland regions. Rural Northern Californians typically dislike San Francisco far more than Angelenos do. Similarly, inland Southern California residents often see Los Angeles as the prohibitively expensive home of two-hour traffic jams. Until the Inland Empire or the San Joaquin Valley get major league teams, we won't see that rivalry playing out at a stadium near you.
