The human brain is often said to be the most complex object in the known universe, and there’s good reason to believe that it is. That lump of jelly inside your head contains at least 80 billion nerve cells, or neurons, and even more of the non-neuronal cells called glia. Between them, they form hundreds of trillions of precise synaptic connections; but they all have moveable parts, and these connections can change. Neurons can extend and retract their delicate fibres; some types of glial cells can crawl through the brain; and neurons and glia routinely work together to create new connections and eliminate old ones.

These processes begin before we are born, and occur until we die, making the brain a highly dynamic organ that undergoes continuous change throughout life. At any given moment, many millions of them are being modified in one way or another, to reshape the brain’s circuitry in response to our daily experiences. Researchers at Yale University have now developed an imaging technique that enables them to visualise the density of synapses in the living human brain, and offers a promising new way of studying how the organ develops and functions, and also how it deteriorates in various neurological and psychiatric conditions.

We spend one third of our lives sleeping, but we still do not know exactly why we sleep. Recent research shows that that the brain does its housekeeping while we sleep, and clears away its waste. According to another hypothesis, sleep plays the vital role of restoring the right balance of brain synapses to enhance learning, and two studies published in today’s issue of Science now provide the most direct evidence yet for this idea.

We do know that sleep is important for consolidating newly formed memories. During waking hours, we learn all kinds of new information, both consciously and unconsciously. To store it, the brain modifies large numbers of synaptic connections, making some of them stronger and larger, and it’s now thought that as we sleep other synapses are weakened or destroyed, so that the important new information is stored for later use, while irrelevant material, which could interfere with learning, is not.

Actor Taapsee Pannu put out yet another throwback picture from her archives on Saturday, and shared her childhood experience, noting how the school does play an important part in shaping one's personality. Mumbai: Three rescued, five feared trapped as wall collapses in Kandivali

The National Disaster Response Force (NDRF) on Sunday informed that few people were feared to be trapped under the debris after a wall of a house collapsed in Kandivali (West) area of Mumbai. According to NDRF, so far three people have been rescued.

"Four to five people possibly trapped after wall of a house collapsed in Kandivali (West) area of Mumbai. 3 people rescued till now," the NDRF said.

A rescue operation is underway at the mishap site.

The fact remains that the villagers had on several occasions complained to the district administration and the State Government with regard to the danger posed by the compound wall. The incident led to scores of people, including relatives of the deceased and the members of the pro-Dalit organisations like Tamil Tigers and other political parties protest against the district administration and pressing for reasonable demand to accommodate them inside the Mettupalayam Government Hospital Campus.

Rescue operation was still underway at the site, he said.

Characterization of the Pseudomonas aeruginosa T6SS PldB immunity proteins PA5086, PA5087 and PA5088 explains a novel stockpiling mechanism

The bacterial type VI secretion system (T6SS) secretes many toxic effectors to gain advantage in interbacterial competition and for eukaryotic host infection. The cognate immunity proteins of these effectors protect bacteria from their own effectors. PldB is a T6SS trans-kingdom effector in Pseudomonas aeruginosa that can infect both prokaryotic and eukaryotic cells. Three proteins, PA5086, PA5087 and PA5088, are employed to suppress the toxicity of PldB-family proteins. The structures of PA5087 and PA5088 have previously been reported, but the identification of further distinctions between these immunity proteins is needed. Here, the crystal structure of PA5086 is reported at 1.90 Å resolution. A structural comparison of the three PldB immunity proteins showed vast divergences in their electrostatic potential surfaces. This interesting phenomenon provides an explanation of the stockpiling mechanism of T6SS immunity proteins.

Factor for inversion stimulation (Fis) is a versatile bacterial nucleoid-associated protein that can directly bind and bend DNA to influence DNA topology. It also plays crucial roles in regulating bacterial virulence factors and in optimizing bacterial adaptation to various environments. Fis from Pseudomonas aeruginosa (PA4853, referred to as PaFis) has recently been found to be required for virulence by regulating the expression of type III secretion system (T3SS) genes. PaFis can specifically bind to the promoter region of exsA, which functions as a T3SS master regulator, to regulate its expression and plays an essential role in transcription elongation from exsB to exsA. Here, the crystal structure of PaFis, which is composed of a four-helix bundle and forms a homodimer, is reported. PaFis shows remarkable structural similarities to the well studied Escherichia coli Fis (EcFis), including an N-terminal flexible loop and a C-terminal helix–turn–helix (HTH) motif. However, the critical residues for Hin-catalyzed DNA inversion in the N-terminal loop of EcFis are not conserved in PaFis and further studies are required to investigate its exact role. A gel-electrophoresis mobility-shift assay showed that PaFis can efficiently bind to the promoter region of exsA. Structure-based mutagenesis revealed that several conserved basic residues in the HTH motif play essential roles in DNA binding. These structural and biochemical studies may help in understanding the role of PaFis in the regulation of T3SS expression and in virulence.

The second benzene mol­ecule has all atoms on a general position: it is disordered over two equally occupied orientations. No directional inter­actions beyond normal van der Waals contacts occur in the crystal.
