A misspelling of former President Donald Trump's name occurred on an optional ballot review screen in Virginia, prompting an unfounded claim on social media of "election fraud." The error was a typo that appeared only on the ballot review screen, not on actual ballots, and would not affect any votes, election officials said.

Bamako is a 2006 film directed by Abderrahmane Sissako, first released at the 2006 Cannes Film Festival on 21 May and in Manhattan by New Yorker Films on 14 February 2007. The film depicts a trial taking place in Bamako, the capital of Mali, amid the daily life that is going on in the city. In the midst of that trial, two sides argue whether the World Bank and International Monetary Fund are guided by special interest of developed nations, or whether it is corruption and the individual nations' mismanagement, that is guilty of the current financial state of many poverty-stricken African countries as well as the rest of the poor undeveloped world. The film even touches on European colonization and discusses how it plays a role in shaping African societies and their resulting poverty and issues.

Malês portrays the difficult living conditions of Black men and women in 19th-century Bahia, fighting against extreme racism, poverty, and religious intolerance. The film features a prominent cast, including Rocco and Camila Pitanga, Wilson Rabelo, Bukassa Kabengele, Samira Carvalho, Rodrigo de Odé, Heraldo de Deus, and Patricia Pillar, along with the director Antonio Pitanga. The script is written by Manuela Dias, and the cinematography is by Pedro Farkas. The film, based on historical facts, depicts the Malês Revolt, the largest organized uprising by enslaved people in Brazilian history. The insurrection mobilized the enslaved and free Black population through the streets of Salvador against slavery in 1835. Led by Muslim Africans, called Malês, the rebellion took place at the end of Ramadan, celebrated in January by Islam. After the revolt’s failure, the demonstrators were severely punished, and repression against Black people in Brazil increased. In the feature, Antonio Pitanga plays Pacífico Licutan, one of the leaders of the uprising, who emphasized the importance of participation from different tribes and religions for the revolt’s success and the end of slavery. The film also presents other participants of the revolt, such as Anuna (Rodrigo dos Santos), Manuel Calafate (Bukassa Kabengele), Vitério Sule (Heraldo de Deus), and Luis Sanim (Thiago Justino), along with fictional characters who portray real-life dramas, such as Dassalu (Rocco Pitanga), Sabina (Camila Pitanga), and Abayome (Samira Carvalho).

What is driving the trend towards greater use of investment screening by nation states and regional economic groupings?

This event is part of the Inclusive Governance Initiative, which is examining how to build more inclusive models and mechanisms of global governance fit for purpose in today’s world.

Pyrroline-5-carboxylate reductase 1 (PYCR1) catalyzes the biosynthetic half-reaction of the proline cycle by reducing Δ1-pyrroline-5-carboxylate (P5C) to proline through the oxidation of NAD(P)H. Many cancers alter their proline metabolism by up-regulating the proline cycle and proline biosynthesis, and knockdowns of PYCR1 lead to decreased cell proliferation. Thus, evidence is growing for PYCR1 as a potential cancer therapy target. Inhibitors of cancer targets are useful as chemical probes for studying cancer mechanisms and starting compounds for drug discovery; however, there is a notable lack of validated inhibitors for PYCR1. To fill this gap, we performed a small-scale focused screen of proline analogs using X-ray crystallography. Five inhibitors of human PYCR1 were discovered: l-tetrahydro-2-furoic acid, cyclopentanecarboxylate, l-thiazolidine-4-carboxylate, l-thiazolidine-2-carboxylate, and N-formyl l-proline (NFLP). The most potent inhibitor was NFLP, which had a competitive (with P5C) inhibition constant of 100 μm. The structure of PYCR1 complexed with NFLP shows that inhibitor binding is accompanied by conformational changes in the active site, including the translation of an α-helix by 1 Å. These changes are unique to NFLP and enable additional hydrogen bonds with the enzyme. NFLP was also shown to phenocopy the PYCR1 knockdown in MCF10A H-RASV12 breast cancer cells by inhibiting de novo proline biosynthesis and impairing spheroidal growth. In summary, we generated the first validated chemical probe of PYCR1 and demonstrated proof-of-concept for screening proline analogs to discover inhibitors of the proline cycle.

In-depth coverage of proteomic analysis could enhance our understanding to the mechanism of the protein functions. Unfortunately, many highly hydrophobic proteins and low-abundance proteins, which play critical roles in signaling networks, are easily lost during sample preparation, mainly attributed to the fact that very few extractants can simultaneously satisfy the requirements on strong solubilizing ability to membrane proteins and good enzyme compatibility. Thus, it is urgent to screen out ideal extractant from the huge compound libraries in a fast and effective way. Herein, by investigating the interior mechanism of extractants on the membrane proteins solubilization and trypsin compatibility, a molecular dynamics simulation system was established as complement to the experimental procedure to narrow down the scope of candidates for proteomics analysis. The simulation data shows that the van der Waals interaction between cation group of ionic liquid and membrane protein is the dominant factor in determining protein solubilization. In combination with the experimental data, 1-dodecyl-3-methylimidazolium chloride (C12Im-Cl) is on the shortlist for the suitable candidates from comprehensive aspects. Inspired by the advantages of C12Im-Cl, an ionic liquid-based filter-aided sample preparation (i-FASP) method was developed. Using this strategy, over 3,300 proteins were confidently identified from 10³ HeLa cells (~100 ng proteins) in a single run, an improvement of 53% over the conventional FASP method. Then the i-FASP method was further successfully applied to the label-free relative quantitation of human liver cancer and para-carcinoma tissues with obviously improved accuracy, reproducibility and coverage than the commonly used urea-based FASP method. The above results demonstrated that the i-FASP method could be performed as a versatile tool for the in-depth coverage proteomic analysis of biological samples.

Purpose:

Colorectal cancer (CRC) screening is recommended starting at age 45, but there has been little research on strategies to promote screening among patients younger than 50. This study assessed the effect of a multicomponent intervention on screening completion in this age group.
