• As October comes to a close, so too does the Bureau of Land Management’s annual Bat Beauty Contest, which asks where the "most stunning bat photographed on BLM public lands" can be found? It's Oregon, motherfuckers! For the third year running WE HAVE THE MOST BEAUTIFUL BAT. YES! Despite J. K. Rowling's tarnished-as-hell legacy, we can still celebrate this feisty male hoary bat, Hoary Potter. It's also worth noting that this is the second year a bat photographed by wildlife technician Emma Busk took top beauty honors. Busk wrote, "anyone who knows me knows that I’ve been wanting to photograph a hoary bat,"—a sentiment we can all echo.

• There's also terrible news, unfortunately. Yesterday evening, a shooting at Vancouver Mall, in Vancouver, WA, killed one person and injured two in the building's food court.  A witness who spoke to KOIN described "a harrowing scene, as the shopping center was full of kids trick-or-treating." So far, police have said they do not have a suspect in custody and the person may still be armed. They told KOIN they were reviewing surveillance footage, but have not yet released a description of the shooter.

• Oh my god, Clark County are you okay? KPTV reports that Clark County Elections Office is warning voters to beware of unofficial ballot boxes. Unofficial ballot boxes are not illegal in Washington! (This also appears to be true of Oregon!) Clark County Elections encouraged voters official ballot return locations.

• The Oregonian is returning to broadsheet size, they announced yesterday in a post that did not explain what that is for readers who didn't go to graphic design school / work in at newspaper with some old dude named Phil. Somewhat surprisingly, Willamette Week's Nigel Jaquiss stepped in to clarify. Tabloid is hotdog style fold; broadsheet is burger. Both WW and the Mercury are hotdogs. The New York Times is burger fold.

• NPR's Up First weirdly tried to do a report on Vice President Kamala Harris "highlighting Trump's own language and using it against him," as if that's somehow something of which to be critical? Gave me the vapors of the new Hasaan Hates Portland sketch from this week:

• In two week's the city's beloved queer documentary festival QDoc will host the Oregon debut of The World According to Allee Willis, which explores the complicated life of a queer "song doctor" who co-wrote not only Earth, Wind and Fire's "September," but the Friends TV show theme. • Atlanta rap star Young Thug, born Jeffery Williams, finally walked free last night, after pleading guilty to "six counts, including possession of drugs and firearms... leading a criminal street gang and conspiracy to violate the RICO act, the state’s racketeering law," the New York Times reports. As part of the plea, the judge sentenced Williams to a total sentence of 40 years, but commuted the five years of prison time to the two and a half years that Williams has already served. Williams will remain on probation for 15 years, and will "be required to stay away from metro Atlanta for the first 10 years of his probation." He is also required to take random drug tests, make anti-gang presentations for children's organizations four times a year, perform 100 hours of community service a year, and "refrain from promoting gangs or being around known gang members." The ongoing Young Slime Life (YSL) court proceedings—which caused NYT's Joe Coscarelli to ask "is YSL was a record label, street gang, or both?"—have stretched into what is now the longest trial in Georgia history. Despite three other plea deals this week, it appears the trial will continue with the two remaining co-defendants.

Kalashnikov AK-203 assault rifles to be produced in India

Russian-Indian joint enterprise Indo-Russian Rifles Private Limited intends to completely localise the production of AK-203 assault rifles in India, TASS reports with reference to Russia's defence export giant Rosoboronexport. "The production plans of the joint venture include plans for the 100-percent localisation of the production of AK-203 assault rifles in India," the company noted. "In addition, the joint venture may increase production and upgrade to produce prospective models on the platform of the Kalashnikov assault rifle,” Rosoboronexport said. In October, the company promised that the Russian-Indian plant for the production of AK-203 Kalashnikov assault rifles (7.62 by 39 mm caliber) in the Indian city of Corva would be ready to begin the production of weapons before the end of 2022.

Expression, purification and crystallization of the photosensory module of phytochrome B (phyB) from Sorghum bicolor

Sorghum, a short-day tropical plant, has been adapted for temperate grain production, in particular through the selection of variants at the MATURITY loci (Ma1–Ma6) that reduce photoperiod sensitivity. Ma3 encodes phytochrome B (phyB), a red/far-red photochromic biliprotein photoreceptor. The multi-domain gene product, comprising 1178 amino acids, autocatalytically binds the phytochromobilin chromophore to form the photoactive holophytochrome (Sb.phyB). This study describes the development of an efficient heterologous overproduction system which allows the production of large quantities of various holoprotein constructs, along with purification and crystallization procedures. Crystals of the Pr (red-light-absorbing) forms of NPGP, PGP and PG (residues 1–655, 114–655 and 114–458, respectively), each C-terminally tagged with His6, were successfully produced. While NPGP crystals did not diffract, those of PGP and PG diffracted to 6 and 2.1 Å resolution, respectively. Moving the tag to the N-terminus and replacing phytochromobilin with phycocyanobilin as the ligand produced PG crystals that diffracted to 1.8 Å resolution. These results demonstrate that the diffraction quality of challenging protein crystals can be improved by removing flexible regions, shifting fusion tags and altering small-molecule ligands.

Multi-species cryoEM calibration and workflow verification standard

Cryogenic electron microscopy (cryoEM) is a rapidly growing structural biology modality that has been successful in revealing molecular details of biological systems. However, unlike established biophysical and analytical techniques with calibration standards, cryoEM has lacked comprehensive biological test samples. Here, a cryoEM calibration sample consisting of a mixture of compatible macromolecules is introduced that can not only be used for resolution optimization, but also provides multiple reference points for evaluating instrument performance, data quality and image-processing workflows in a single experiment. This combined test specimen provides researchers with a reference point for validating their cryoEM pipeline, benchmarking their methodologies and testing new algorithms.

Crystallographic fragment-binding studies of the Mycobacterium tuberculosis trifunctional enzyme suggest binding pockets for the tails of the acyl-CoA substrates at its active sites and a potential substrate-channeling path between them

The Mycobacterium tuberculosis trifunctional enzyme (MtTFE) is an α2β2 tetrameric enzyme in which the α-chain harbors the 2E-enoyl-CoA hydratase (ECH) and 3S-hydroxyacyl-CoA dehydrogenase (HAD) active sites, and the β-chain provides the 3-ketoacyl-CoA thiolase (KAT) active site. Linear, medium-chain and long-chain 2E-enoyl-CoA molecules are the preferred substrates of MtTFE. Previous crystallographic binding and modeling studies identified binding sites for the acyl-CoA substrates at the three active sites, as well as the NAD binding pocket at the HAD active site. These studies also identified three additional CoA binding sites on the surface of MtTFE that are different from the active sites. It has been proposed that one of these additional sites could be of functional relevance for the substrate channeling (by surface crawling) of reaction intermediates between the three active sites. Here, 226 fragments were screened in a crystallographic fragment-binding study of MtTFE crystals, resulting in the structures of 16 MtTFE–fragment complexes. Analysis of the 121 fragment-binding events shows that the ECH active site is the `binding hotspot' for the tested fragments, with 41 binding events. The mode of binding of the fragments bound at the active sites provides additional insight into how the long-chain acyl moiety of the substrates can be accommodated at their proposed binding pockets. In addition, the 20 fragment-binding events between the active sites identify potential transient binding sites of reaction intermediates relevant to the possible channeling of substrates between these active sites. These results provide a basis for further studies to understand the functional relevance of the latter binding sites and to identify substrates for which channeling is crucial.

C-SPAM: an open-source time-resolved specimen vitrification device with light-activated molecules

Molecular structures can be determined in vitro and in situ with cryo-electron microscopy (cryo-EM). Specimen preparation is a major obstacle in cryo-EM. Typical sample preparation is orders of magnitude slower than biological processes. Time-resolved cryo-EM (TR-cryo-EM) can capture short-lived states. Here, Cryo-EM sample preparation with light-activated molecules (C-SPAM) is presented, an open-source, photochemistry-coupled device for TR-cryo-EM that enables millisecond resolution and tunable timescales across broad biological applications.
