Killer Archaea: Virus-Mediated Antagonism to CRISPR-Immune Populations Results in Emergent Virus-Host Mutualism

ABSTRACT

Theory, simulation, and experimental evolution demonstrate that diversified CRISPR-Cas immunity to lytic viruses can lead to stochastic virus extinction due to a limited number of susceptible hosts available to each potential new protospacer escape mutation. Under such conditions, theory predicts that to evade extinction, viruses evolve toward decreased virulence and promote vertical transmission and persistence in infected hosts. To better understand the evolution of host-virus interactions in microbial populations with active CRISPR-Cas immunity, we studied the interaction between CRISPR-immune Sulfolobus islandicus cells and immune-deficient strains that are infected by the chronic virus SSV9. We demonstrate that Sulfolobus islandicus cells infected with SSV9, and with other related SSVs, kill uninfected, immune strains through an antagonistic mechanism that is a protein and is independent of infectious virus. Cells that are infected with SSV9 are protected from killing and persist in the population. We hypothesize that this infection acts as a form of mutualism between the host and the virus by removing competitors in the population and ensuring continued vertical transmission of the virus within populations with diversified CRISPR-Cas immunity.

IMPORTANCE Multiple studies, especially those focusing on the role of lytic viruses in key model systems, have shown the importance of viruses in shaping microbial populations. However, it has become increasingly clear that viruses with a long host-virus interaction, such as those with a chronic lifestyle, can be important drivers of evolution and have large impacts on host ecology. In this work, we describe one such interaction with the acidic crenarchaeon Sulfolobus islandicus and its chronic virus Sulfolobus spindle-shaped virus 9. Our work expands the view in which this symbiosis between host and virus evolved, describing a killing phenotype which we hypothesize has evolved in part due to the high prevalence and diversity of CRISPR-Cas immunity seen in natural populations. We explore the implications of this phenotype in population dynamics and host ecology, as well as the implications of mutualism between this virus-host pair.

A few days ago, she shared some 'bored' selfies on her Instagram account. Ira has been spending her time at home due to the lockdown imposed to curb the coronavirus pandemic.

For the unversed, Ira has made her directorial debut with a theatre production titled Euripides' Medea. The play is an adaptation of Euripides' Greek tragedy Medea and stars Aamir's son Junaid Khan and actress Hazel Keech among others.

On the professional front, her dad, Aamir Khan will be seen opposite Kareena Kapoor Khan in Laal Singh Chaddha, which is an official adaptation of the Hollywood film, Forrest Gump. The film is written by Atul Kulkarni, helmed by Secret Superstar director Advait Chandan and co-produced in association with Viacom 18 Motion Pictures.

Mrs. Serial Killer Review: Miss Jacqueline Fernandez robs off all the fun from this film

Forget everything! Forget that filmmaker Shirish Kunder's last feature film was Joker, forget that the leading lady of his fresh film, Mrs. Serial Killer, is Jacqueline Fernandez. And just read this premise- A helpless and hapless wife takes the suicidal decision of going on a murderous rage and becoming a serial killer when her husband is arrested on the charges of brutally killing six unmarried and pregnant women. In short, a woman will go to any lengths to save her husband.

Any film with this plot can amalgamate its narrative with both thrill and tragedy. You first feel a sense of sympathy for the man who has been wrongly accused and unfairly imprisoned. You feel empathetic towards the woman who now has to fight what could be the most traumatic battle of her life. And then you feel the tension and thrill of wanting to know how she would execute all her murders! What unfolds is your perfect Bollywood potboiler, driven by drama and fueled by unpredictability. But what we see in Mrs. Serial Killer is a puerile and almost-preposterous telling of an unseen and unknown CID episode.

The performances are all ham-and-cheese, and not even worthy of being your enjoyable and entertaining guilty pleasure stuff. It's juvenile and joyless from start to finish. If Manoj Bajpayee played your perfect hero in The Family Man, Fernandez plays your loyal family woman in this film who readily agrees to become Mrs. Serial Killer to save her husband. Kunder does what no other filmmaker even imagined he would, he gives her more screen time than Bajpayee and let her have her moment of glory. To say she falters would be an understatement, she collapses ever before her mission has begun.

Her performance is both annoying and alarming and makes you smile more than scared and that too in all the incorrect ways and at the incorrect moments. Look how she reacts when a gatekeeper refuses to let her in, look how she reacts when a receptionist refuses to let her meet a gynecologist, look how she reacts when she successfully unlocks her husband's laptop, and look how she reacts when she's about to drug and almost kill her prey. And after a while, you stop looking at all. Kunder has certainly been unwise while casting someone deadpan for a deep role.

There's barely anything that works, not even Manoj Bajpayee. He's hardly there and dedicates all his screen time in hamming. A languid Mohit Raina nearly puts you off to sleep, and the climatic twist suggests nothing but the sheer limitations and lunacy of everyone involved in this Netflix nonsense. Kunder's debut, Jaan-E-Mann, although a commercial failure, was earnest in its craftsmanship and entertaining in its moments, but his repertoire has deteriorated ever since. Mrs. Serial Killer is his weakest film to date, worsened by its frustratingly limited leading lady. It does deliver unintentionally comical moments and there are plenty of them. Watch out for the implausible and innocuous climax where Bajpayee snarls, "Comedy Kahaan Hai?" It's right there, sir!

Govind Pansare killers now have Rs 50 lakh bounty on their heads

The monetary reward for the person providing solid leads about the two wanted suspects in Comrade Govind Pansare's murder case has been increased by a whopping Rs 40 lakh by the state home department.

The Communist Party of India (CPI) leader was killed in February 2015, with one team each in every district of Maharashtra probing the case. Police teams on Saturday began pasting posters declaring the hike in reward money at all major public places.

The initial prize amount declared by the Special Investigation Team (SIT) for the informant was R5 lakh which was increased to R10 lakh in February 2016 and has now been revised to Rs 50 lakh, according to the SIT's orders dated Saturday.

Comrade Pansare and his wife Uma were returning from their morning walk in Sagar Mahal, Kolhapur, when two bike-borne persons fired five rounds at Pansare on February 16, 2015.

Police teams have begun pasting the announcement posters at public places across the state

The communist was airlifted and rushed to Mumbai for treatment where he died four days later, on February 20. While suspects Sameer Gaikwad, Amol Kale and Mumbai-based ENT specialist Dr Virendrasinh Tawade have been arrested by the police, Vinay Baburao Pawar and Sarang Deelip Akolkar alias Kulkarni are absconding.

After the SIT, investigating the Pansare murder case, issued the orders for increasing the informant's prize amount last month, the Maharashtra police prepared posters declaring this in English, Hindi and Marathi. The whereabouts of the informant will be kept secret, the SIT has assured.

A senior officer who is part of the investigating team said, “The reward is Rs 25 lakh each for information about Pawar and Akolkar. We had received some clues in the past too but those did not lead to anything while probe on those lines is still on too.”

Pansare's daughter-in-law Megha, however, wasn't too impressed with the hike in prize money. “It is too late now but there is some hope. The Bombay High Court too was amused and sceptical of a cash prize leading to clues,” she said. “Do you think people will take you to the accused. For all you know, they might be getting paid more to keep quiet,” the court had remarked at the last case hearing.

Wanted in Dabholkar case too

Pawar and Akolkar, who are associated with the Sanatan Sanstha, are also wanted in connection with the murder of rationalist Dr Dabholkar. Akolkar, who hails from Pune, is an electronic engineer and has been on the run since his name cropped up during the Goa blast case being probed by the National Investigation Agency.

The NIA had even secured an Interpol Red Corner Notice against him in 2012. Pawar, a resident of Miraj in Sangli district of western Maharashtra, was last located in Umbaraj in Karad taluka of Satara district. He has been untraceable ever since.

Farah Khan's life comes to full circle with Aamir Khan's niece Zayn making her debut in Mrs Serial Killer

On May 1, Shirish Kunder's directorial Mrs Serial Killer was released on Netflix. The film is co-produced by Farah Khan. While Jacqueline Fernandez, Manoj Bajpayee and Mohit Raina play the lead roles, the film also marks the debut of Zayn Khan, who is the daughter of Mansoor Khan and niece of Aamir Khan.

On Thursday, Farah Khan took to her Instagram handle to share a picture with Zayn Khan. Sharing the picture she also revealed how life came a full circle for her with Zayn's debut in her film. "In 1992 her father, Mansur khan gave me a break as choreographer in #jojeetawohisikandar .. 28 yrs later his daughter @zaynmarie makes her debut n wins hearts in our film , #mrsserialkiller.. life has a way of coming full circle," she wrote.


Mrs Serial Killer is the story of a renowned gynaecologist who is accused of murdering five unmarried pregnant girls. All the evidence is against him due to which his bail is rejected. With no other option in hand, the gynaecologist’s wife decides to commit a copycat crime to prove his innocence. While Manoj Bajpayee plays the gynaecologist, Jacqueline Fernandez plays the dutiful better half. Mohit Raina essays the role of the cop. As for Zayn Marie Khan, she plays Anushka, who is selected by Jacqueline’s character Sona as her target.
